Cytokine Regulation of the Bone Pre- and Metastatic Niches: Implications for Breast Cancer Dormancy.

Breast cancer relapse in bone is a significant clinical problem that is experienced in ~70-80% of patients with late-stage breast cancer. This condition commonly occurs 5-10+ years following surgical removal of the primary tumour. The long latency seen prior to relapse in bone is a result of tumour cell dormancy. Once disseminated to the bone, tumour cell interaction with the bone metastatic niche (endosteal niche and endovascular cells) maintains cells in a dormant state until changes to the local environment activate the niche to support outgrowth. Amassing evidence suggests that cytokines a

Neuroinflammation in Central Nervous System Tumors.

Neuroinflammation within the tumor microenvironment (TME) of central nervous system (CNS) neoplasms, particularly glioblastoma (GBM), is no longer viewed merely as a reactive phenomenon but rather as a major driver of gliomagenesis and malignant transformation. This process involves a shift from acute immune activation to a chronic, sterile state that reshapes the CNS borders and immune niches to favor tumor evasion. This narrative review provides a comprehensive mechanistically focused analysis of the mechanisms governing the inflammatory stroma in primary and metastatic brain neoplasms. It c

[Regulatory T Cells and Cancer-Focus on Renal Cell Carcinoma].

Recent advances in cancer immunology have revealed that tumor progression is profoundly influenced by interactions between tumor cells and the host immune system. Although the immune system can recognize and eliminate malignant cells through immune surveillance, tumors can evade immune-mediated destruction by acquiring various immune escape mechanisms. Regulatory T cells (Tregs) represent one of the key immunosuppressive components that facilitate tumor immune evasion. Tregs, typically characterized as CD4+CD25+FoxP3+ T cells, play an essential role in maintaining immune tolerance and preventi

Photodynamic therapy remodels cutaneous tumor immunity: innate priming, suppressive feedback, and translational opportunities.

Photodynamic therapy (PDT) is an established local treatment for actinic keratoses, Bowen disease, and selected low-risk basal cell carcinomas, but its potential to reshape antitumor immunity may extend its value beyond direct phototoxicity. We conducted a narrative synthesis of clinical, translational, and mechanistic evidence, supplemented by targeted searches through August 2026, and appraised major findings using a three-level evidence-maturity framework. Current evidence supports a temporally organized model in which PDT-induced reactive oxygen species promote tumor and microvascular inju

Microfluidic Single-Cell Bioanalysis for Decoding Tumor Heterogeneity.

Tumor heterogeneity drives cancer progression, dissemination, therapeutic adaptation, and relapse, yet many clinically relevant cell states are rare, transient, context-dependent, and obscured by population-averaged analysis. This review examines how microfluidic platforms preserve biologically meaningful linkages among cell identity, molecular state, secreted output, functional phenotype, perturbation history, and microenvironmental context, which are frequently disrupted by conventional workflows. We first define analytical requirements imposed by tumor heterogeneity, then examine microwell-

In situ antigen-capture strategies for enhancing dendritic cell-mediated anti-tumor immunity.

Dendritic cell (DC)-mediated tumor immunotherapy has demonstrated considerable potential, effectively bridging tumor antigens with specific anti-tumor immune responses. However, the heterogeneous and immunosuppressive tumor microenvironment (TME) frequently impairs DC function by inhibiting antigen uptake, restricting differentiation into mature DCs (mDCs), and limiting migration to tumor-draining lymph nodes (TDLNs), ultimately resulting in immune tolerance that diminishes specific anti-tumor immune responses. To overcome these limitations and effectively restore the DC-mediated link between

Osteosarcoma Exosome Priming of Primary Human Lung Fibroblasts Induces an Immune Modulatory and Protumorigenic Phenotype.

ABSTRACT: Osteosarcoma (OS) is the most common malignant bone cancer. Thirty to forty percent of all patients with OS develop tumor recurrence, almost exclusively in the form of lung metastasis, which is associated with a dismal 20% 5-year survival rate. Cancer-associated fibroblasts are a critical cell type within the lung tumor microenvironment that promote immune suppression, drug resistance, and tumor cell survival. Prior work shows tumor cells can co-opt fibroblasts to a protumorigenic phenotype via exosome-mediated intercellular communication. Currently, the mechanisms by which OS exosom

Dormancy in Metastatic Colorectal Cancer: Tissue Engineering Opportunities for In Vitro Modeling.

Colorectal cancer (CRC) recurs at a striking rate, specifically in patients with liver metastasis. Dormant CRC cells disseminated following initial primary tumor resection or treatment often resurface years later to form aggressive, therapy-resistant tumors that result in high patient mortality. Routine imaging-based screenings often fail to detect dormant cancer cell clusters, and there are no overt symptomatic presentations, making dormant CRC a major clinical challenge to diagnose and treat. Tissue engineering approaches are ideally suited to model dormant cancer cells and enable the discov

Engineering microcavity arrays as a niche for high-throughput and high-content analysis of tumoroids: A detailed methodology.

Tumoroids, which replicate patient tumor genetics and drug responses, have revolutionized oncology by enabling personalized treatment development and reducing reliance on animal testing. Despite remarkable advances in reproducing ever more sophisticated biology in advanced organoid platforms, there remain foundational challenges in standardizing and scaling-up tumour organoid development. Moreover, recreating key aspects of the tumour niche, namely its biomechanical properties is still overlooked in drug screening. To address these shortcomings, we introduce a technology for high-throughput de

Harnessing the effector-to-regulatory T cell ratio to advance prognosis and precision therapy in pancreatic cancer.

Pancreatic cancer remains one of the most lethal malignancies, characterized by rapid progression, late-stage diagnosis, and resistance to current therapies. A major contributor to its poor prognosis is the highly immunosuppressive tumor microenvironment (TME), where regulatory T cells (Tregs) suppressing effector T cells (Teffs). The ratio of Teffs to Tregs (Teff/Treg ratio) has emerged as a pivotal biomarker that reflects immune balance within the TME, with significant prognostic and therapeutic relevance. A low Teff/Treg ratio correlates with poor clinical outcomes, immunotherapy resistance

Glucose restriction: double edged sword in cancer treatment.

Glucose restriction generally limits tumor growth. Recently, Wu et al. reported that glucose restriction inhibits primary tumors but promotes lung metastasis by forming a macrophage-dominated, natural killer (NK) cell-deficient pre-metastatic niche (PMN). This finding provides a new perspective on understanding the dual role of glucose metabolism regulation in cancer treatment.

Gut microbiota reshaping the pancreatic cancer immune microenvironment: new avenues for immunotherapy.

Pancreatic cancer remains one of the deadliest malignancies, primarily due to its highly immunosuppressive tumor microenvironment (TME) and poor response to conventional therapies. Increasing evidence highlights the gut microbiota as a pivotal regulator of antitumor immunity, modulating T cell activation, macrophage polarization, and dendritic cell function. Microbial communities and their metabolites can either inhibit or enhance immune surveillance, thereby influencing the efficacy of immunotherapies such as immune checkpoint inhibitors (ICIs) and CAR-T cell therapy. Approaches including die

Intratumoral microbiome and pancreatic cancer: an enabling hallmark and path to novel treatments?

In 2022, Hanahan integrated polymorphic microbiomes to the hallmarks of cancer, resulting in 14 overarching features that are considered fundamental to initiation and progression of cancers. It is well acknowledged that genomic instability/genetic alterations together with tumor-associated inflammation are so called "enabling hallmarks" as they drive the acquisition of the other traits. The microbiome is a key component of the inflammatory tumor stroma. Pancreatic ductal adenocarcinoma (PDAC) in particular is characterized by a pronounced stromal compartment whose role in the acquisition of ma

Microglial reprogramming: a potential new frontier in enhancing immunotherapy for melanoma brain metastasis.

The brain is a common site of metastatic dissemination in advanced melanoma. Due to limited access to samples from human brain metastases, our understanding of the tumor microenvironment in the brain lags behind that of other sites, and murine studies are therefore highly informative. Rodriguez-Baena et al. conducted elegant studies of myeloid cells within melanoma brain metastases, demonstrating their plasticity and changes before and after colonization by melanoma cells. The immune-inhibitory changes in microglial cells may be reversed or mitigated by inhibition of RelA/NF-κB.

Crosstalk between lipid droplets and autophagy in cancer: A nexus for therapeutic targeting.

Metabolic reprogramming is a cornerstone of cancer cell adaptation to the demanding tumor microenvironment, requiring fine-tuned control over energy, lipid metabolism, and stress responses. Central to this adaptation is the profound and bidirectional interplay between two key cellular processes: lipid storage in lipid droplets (LDs) and cellular recycling via autophagy. LDs are dynamic organelles that have emerged as critical metabolic and signaling hubs, extending far beyond their role as simple lipid depots. Autophagy, a fundamental degradation system, supplies essential metabolites during s

Role of metabolic reprogramming in colorectal cancer and pharmacologic intervention.

Metabolic reprogramming, a core hallmark of tumorigenesis, is crucial for CRC development and metastasis; this study reviews the process of metabolic reprogramming in CRC and the latest advances in pharmaceutical therapy, examines various intervention options from three perspectives-microenvironmental change, suppression of key enzyme function, and metabolic substrate deprivation-and uses translational medicine to discuss the clinical applications of these strategies, while noting that changes in metabolic and immunological parameters in the tumor microenvironment enhance tumor cell proliferat

TGF‑β at the Crossroads: Orchestrating the Bone Metastatic Microenvironment and Shaping Therapeutic Frontiers.

Bone remains one of the most hospitable-and devastating-destinations for metastatic cancer cells. At the center of this unwelcome alliance is transforming growth factor‑β (TGF‑β), a cytokine stored in the mineralized matrix and unleashed during osteoclastic bone resorption. Once activated, TGF‑β fuels a self‑reinforcing "vicious cycle": it co‑opts tumor cells to undergo epithelial‑to‑mesenchymal transition, recruits and primes osteoclasts, suppresses osteoblast function, and shapes an immunosuppressive niche that shields malignant clones. The result is a micro‑environment exquisitely tuned for
