A case of pathology-proven neuromyelitis optica spectrum disorder with Sjögren syndrome manifesting aphasia and apraxia due to a localized cerebral white matter lesion.

A woman with Sjögren syndrome manifesting as aphasia with a left deep cerebral white matter lesion tested positive for anti-aquaporin 4 (AQP4) antibody. Open biopsy of the lesion revealed active demyelination with edematous changes and the preservation of most axons, indicating a non-necrotic demyelinating lesion. Immunostaining for AQP4 was diffusely lost, whereas the loss of glial fibrillary acidic protein immunostaining was limited but with highly degenerated astrocytic foot processes in perivascular areas. These results suggested neuromyelitis optica spectrum disorder (NMOSD) pathology rat

Neuromyelitis optica: potential roles for intravenous immunoglobulin.

Neuromyelitis optica (NMO) is an idiopathic central nervous system inflammatory demyelinating disease that causes optic neuritis, transverse myelitis, and other CNS syndromes. It is distinct from multiple sclerosis and is associated with autoantibodies that target aquaporin-4 (AQP4), an astrocyte water channel. Evidence indicating antibody-mediated immune injury in NMO includes its association with other autoimmune diseases, lesional pathology that reveals prominent complement activation and immunoglobulin deposition, pathogenic potential of AQP4 autoantibodies based on in vitro studies, and r

Central nervous system tissue heterotopia of the nose: case report and review of the literature.

The authors present a case of heterotopic central nervous system tissue observed in an 81-year-old male in the form of an ethmoidal polyp. A review of the literature indicates that this is a rare condition characterised by a connective tissue lesion with astrocytic and oligodendrocytic glial cells, which may be located outside the nasal pyramid in some cases and inside the nasal cavity in others. The most important diagnostic aspect involves differentiating these from meningoencephalocele, which maintains an anatomical connection with central nervous system tissue. Contrast-enhanced imaging is

Multi-focal cerebral oligoastrocytoma in a puppy.

A nine-month-old puppy was presented for investigation of seizures. Neurological deficits were found localising to the prosencephalon, mesencephalon and myelencephalon. Magnetic resonance imaging identified multiple, large lesions involving both cerebral hemispheres. Management with antiepileptic and immunomodulatory drugs was instituted; however, the seizures became progressively refractory and the puppy was euthanased. Histopathology following post-mortem examination found a mixed glial cell tumour with discrete areas where neoplastic cells appeared oligodendroglial or astrocytic. Primary in

Therapeutic immunization protects dopaminergic neurons in a mouse model of Parkinson's disease.

Degeneration of the nigrostriatal dopaminergic pathway, the hallmark of Parkinson's disease, can be recapitulated in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-intoxicated mice. Herein, we demonstrate that adoptive transfer of copolymer-1 immune cells to MPTP recipient mice leads to T cell accumulation within the substantia nigra pars compacta, suppression of microglial activation, and increased local expression of astrocyte-associated glial cell line-derived neurotrophic factor. This immunization strategy resulted in significant protection of nigrostriatal neurons against MPTP-induce

Early post-traumatic cerebral blood flow mapping: correlation with structural damage after focal injury.

Focal post traumatic mass lesions such as contusions and intracerebral haematomas are common, and often difficult for neurosurgeons to manage, because little is known of their pathophysiology. We have mapped cerebral blood flow, and studied small vessel ultrastructure at different time points within the first three weeks of head injury, in patients with these lesions. A zone of ischaemic brain is always present around these lesions, and persists for weeks or months. This accords with astrocyte swelling and microvascular compression seen on electron microscopy. Focal zones of hyperaemia were al

Increased proteins in post-mortem brain in a case of Pick's disease and in Huntington's disease.

Soluble proteins from temporal cortex and caudate nucleus from a case of Pick's disease, 5 cases of Huntington's disease and 5 controls were analysed by SDS-polyacrylamide gel electrophoresis. Acidic proteins of molecular weight 39 000-42 000, which showed glial fibrillary acidic protein immunoreactivity, were increased in temporal cortex of the Pick's case. Proteins of these molecular weights were also increased in caudate nucleus of the Huntington's cases. Our results show that the astrocytic gliosis observed in temporal cortex in Pick's disease and in caudate nucleus in Huntington's disease

Biological behavior of the primitive neuroectodermal tumors: significant supratentorial childhood gliomas.

Supratentorial gliomas in the pediatric age group at the University of Kentucky Medical Center accounted for 20% of all childhood brain tumors. Seventeen of the 20 children with these tumors, regardless of histologic type, mode of therapy, or other parameters died within five years after tissue diagnosis, and most were dead within two years. Unlike the astrocytic gliomas, the poorly differentiated primitive neuroectodermal tumors tended to spread diffusely throughout the central nervous system and accounted for 35% of the supratentorial gliomas in children at our institution. Results of this s

In the brain, loss of primary cilia or GBA1 deficiency impairs Hedgehog signaling, reducing production of neuroprotective factors needed to support vulnerable dopamine neurons. Genome-scale metabolic modeling uncovers cell-type specific signatures associated with APOE variants.

Metabolic dysregulation is a key feature of Alzheimer's disease (AD) pathogenesis, with the APOE ε4 variant (APOE4) representing the strongest genetic risk factor. In this study, we utilized a metabolite-centric approach to investigate how APOE4 reshapes cellular metabolism across brain cell types. Transcriptomic data from isogenic iPSC-derived neurons, astrocytes, and microglia were integrated into a human genome-scale metabolic model to identify genotype-specific alterations. These findings were validated using metabolomics data from the same cell types. In addition to cholesterol and fatty …

Interleukin-33 promotes dopaminergic neuron survival and inhibits glial activation in Parkinson's disease models.

Interleukin-33 (IL33) plays a critical role in modulating immune and inflammatory responses across various diseases, yet its influence on Parkinson's disease (PD) remains elusive. In this study, we provide substantial evidence that global depletion of Il33 exacerbates PD pathology and motor deficits in vivo. Moreover, Il33 deficiency diminishes the communication between astrocytes and microglia, leading to a more profound injury to primary neurons in vitro. Notably, the application of recombinant IL33 (rIL33) following MPTP treatment or α-synucleinA53T overexpression partially inhibits the red…

Microglia coordinate activity-dependent protein synthesis in neurons through metabolic coupling.

De novo protein synthesis is required for long-lasting synaptic plasticity and memory, but it comes with a great metabolic cost. In the mammalian brain, it remains unclear which cell types and biological mechanisms are critical for sensing and responding to increased metabolic demand. Here we demonstrate that microglia, resident macrophages of the brain, coordinate metabolic coupling between endothelial cells, astrocytes, and neurons to fuel protein synthesis in active neurons. Increasing metabolic demand via a motor task stimulates microglia to secrete the hypoxia-responsive protein CYR61, in…

Effect of Deoxyglucose on Cultured Glial Cells of the Rat Cerebral Cortex under Conditions of LPS Stimulation.

It was shown that blocking of glycolysis by 2-deoxy-D-glucose (DG) inhibits LPS-induced NO production in glial cell cultures and reduces the mitochondrial membrane potential. Lactate partially prevented the decrease in the mitochondrial membrane potential of gliocytes, but did not affect the inhibitory effect of DG. Immunocytochemical typing showed the presence of a large number of astrocytes and microglia in the culture. In the control, microglial cells had a large number of processes, which is typical of nonactivated cells, whereas in LPS-treated cultures, microglia had a flattened amoeboid …

18 F-FDG PET/CT Findings in an Atypical Case of Glial Fibrillary Acidic Protein Meningomyelitis.

Gilal fibrillary acidic protein (GFAP) meningomyelitis is a rare autoimmune inflammatory disorder of the central nervous system, typically presenting with subacute encephalopathy, ataxia, optic neuritis, and cerebrospinal fluid (CSF) pleocytosis. It is characterized by the presence of GFAP-specific autoantibodies, which target astrocytes and cause widespread neuroinflammation. 18 F-FDG PET/CT plays a critical role in the early diagnosis of autoimmune encephalitides, particularly in cases where conventional MRI findings may be subtle or nonspecific. Here, we present a case of 21-year-old woman …

The Effect of Intranasal Administration of Gangliosides on the Viability of CA1 Hippocampal Neurons in Rat Two-Vessel Occlusion Model of Forebrain Ischemia/Reperfusion Injury.

Intranasal administration of total bovine brain gangliosides (6 mg/kg) to rats protected the CA1 hippocampal neurons from the death caused by two-vessel occlusion model (with hypotension) of forebrain ischemia/reperfusion injury. The immunohistochemical reaction of specific antibodies to marker proteins of activated microglia (Iba1) and astrocytes (GFAP) in hippocampal slices revealed the neuroprotective effect of exogenous gangliosides which can be mostly explained by their ability to suppress neuroinflammation and gliosis. The expression of neurotrophic factor BDNF in the CA1 region of hippo…

Retinal ganglion cell circuits and glial interactions in humans and mice.

Retinal ganglion cells (RGCs) are the brain's gateway for vision, and their degeneration underlies several blinding diseases. RGCs interact with other neuronal cell types, microglia, and astrocytes in the retina and in the brain. Much knowledge has been gained about RGCs and glia from mice and other model organisms, often with the assumption that certain aspects of their biology may be conserved in humans. However, RGCs vary considerably between species, which could affect how they interact with their neuronal and glial partners. This review details which RGC and glial features are conserved b…
