High level expression and immunochemical characterization of botulinum neurotoxin type F light chain.

Botulinum neurotoxins (BoNTs) are the most toxic biological substances known. Their potential use as biological warfare agent results in their classification as category A biowarfare agent by Centers for Disease Control and Prevention (CDC), USA. Presently, there are no approved detection system and pharmacological treatments for BoNT intoxication. Although a toxoid vaccine is available for immuno-prophylaxis, vaccines cannot reverse the effect of pre-translocated toxin. Direct handling of the live BoNTs for developing detection and therapeutics may pose fatal danger. This concern was addresse

Beyond Fungitoxicity: Recent Achievements in Targeted Fungicide Discovery.

Phytopathogenic fungi pose a constant threat to worldwide agricultural production. Given the widespread development of fungicide resistance and increasing environmental and regulatory constraints, precision disease-control strategies are urgently needed to enhance selectivity, durability, and sustainability. This review critically examines recent advances in targeted fungicide discovery against phytopathogenic fungi. We categorize these strategies into three complementary groups: (1) targeting fungal biological processes that are essential or infection-associated; (2) disarming pathogen virule

Friends or foes: Unraveling the tsetse fly-Spiroplasma symbiosis.

Tsetse flies (Glossina spp.) transmit African trypanosomes, the causative agents of human African and African animal trypanosomiases (HAT and AAT, respectively). These neglected tropical diseases impose significant public health and economic burdens across sub-Saharan Africa. Trypanosome transmission by tsetse flies is influenced by multiple factors, including host genetic background, ecological factors, and interactions with heritable microbial endosymbionts. Spiroplasma glossinidia has recently emerged as an important modulator of tsetse reproductive fitness and vector competence, making it

[Secondary osteoporosis or secondary contributors to bone loss in fracture. Therapeutic intervention of rheumatoid arthritis bone loss].

In rheumatoid arthritis (RA) , the osteoclast pathway is activated by abnormal immune conditions accompanied by chronic inflammation resulting in periarticular osteoporosis and local bone destruction around joints. In addition, multiple factors lead to systemic osteoporosis, including reduced physical activity and pharmacotherapies such as steroids. These conditions cause decreased bone mineral density and bone quality, and expose patients to an increased risk of fracture. When treating patients with RA osteoporosis, it is important to improve systemic osteoporosis using anti-osteoporotic agen

Bioelectric Modulation as a Systems-Level Adjunct to Regenerative Therapies in Facial Plastic Surgery and Hair Restoration.

Regenerative strategies in facial plastic surgery increasingly incorporate platelet-rich fibrin, platelet-rich plasma, fat grafting, and emerging biologic agents. Despite widespread adoption, outcomes remain variable, in part due to limited control of upstream processes such as perfusion, extracellular matrix remodeling, and stem cell niche stability. To review the mechanistic basis and emerging clinical evidence supporting bioelectric modulation as a systems-level regulator of regenerative pathways relevant to facial rejuvenation, wound healing, and hair restoration. Bioelectric modulation re

Biologic modifiers in periodontal regeneration.

The specific objective of this article was to update the reader on biologic modifiers being used or suggested for use in therapies directed at regenerating periodontal tissues. As indicated from the studies presented here, many of these biologic modifiers have significant influences on cell behavior and show great promise for use in regenerative therapies. As discussed here, however, additional investigations are required both at the molecular level and at the clinical level to improve the predictability of regenerative therapies. With active investigations directed toward understanding the bi

Biologic Disease-Modifying Antirheumatic Drug Use During Pregnancy and Lactation.

Autoimmune rheumatic diseases (ARDs) are chronic diseases characterized by periods of flare and remission. Autoimmune rheumatic diseases are often diagnosed in reproductive-aged women, and treatment decisions must factor in pregnancy potential and intent. Well-controlled maternal disease in the prepregnancy period and during pregnancy portends improved maternal and fetal outcomes. Although conventional immunosuppressant and agents remain a mainstay of treatment, use of biologic disease-modifying antirheumatic drugs (bDMARDs), either alone or in combination with conventional agents, is increasi

Novel Therapeutic Approaches and Targets for the Treatment of Neutrophilic Dermatoses, Management of Patients with Neutrophilic Dermatoses and Future Directions in the Era of Biologic Treatment.

Neutrophilic dermatoses are a heterogeneous group of inflammatory skin disorders characterized by the presence of a sterile, predominantly neutrophilic infiltrate on histopathology. Universally accepted and validated guidelines for the management of neutrophilic dermatoses do not exist, also given the paucity of randomized controlled study and high-quality data. However, the literature on the effective use of biologic therapies is rapidly expanding. This article reviews the epidemiology, clinical characteristics, histopathologic features, and management of pyoderma gangrenosum as well as Sweet

Rapid biofilm disruption by ionic buffers enables calcium phosphate cement functionalization.

Antimicrobial resistance and biofilm-associated infections continue to undermine standard antibiotic therapies, prompting the need for physicochemical adjuvant strategies. In this study, we evaluated the antibiofilm and antimicrobial activity of 7 biological buffer systems including citrate, acetate, sodium phosphate, potassium phosphate, borate, carbonate, and Tris-buffered saline against methicillin-susceptible and methicillin-resistant Staphylococcus aureus (MSSA and MRSA). Using a range of biofilm, metabolic, and cytotoxicity assays coupled with bioluminescence imaging, we demonstrate that

Empathy in the Soil, Agents on the Desktop: Why Plant Pathology Needs a Scientist-in-the-Loop AI Paradigm.

Plant pathology faces a cognitive bottleneck caused by the growing volume of biological data. Addressing it requires a shift from passive chatbots to autonomous AI research agents that operate in Goal-Action-State cycles. Researchers can set up a local, desktop-based agentic framework that combines semantic discovery, LLM-Wiki knowledge synthesis, and in silico prevalidation, managed under a human-directed three-tier organizational model. Strict "Scientist-in-the-Loop" boundaries, which means that a human researcher sets the goals, reviews AI-generated output before it is treated as evidence,

The emerging pathogen Candida auris: host interactions and disease drivers.

Candida auris has emerged over the past fifteen years as a multidrug-resistant human fungal pathogen responsible for hospital-acquired infections associated with high mortality rates. Clinical isolates frequently exhibit resistance to one or more first-line antifungal drugs, and management is further complicated by persistent colonization, biofilm formation, and historical misidentification in diagnostic laboratories. Rapid species-level identification and accurate antifungal susceptibility testing are critical for effective patient care and infection control. In this article, we summarize the

Colonic Mucormycosis in Fistulizing Crohn's Disease.

Gastrointestinal mucormycosis, a rare fatal fungal infection in an immunocompromised host, affects mainly the stomach. Colonic mucormycosis is infrequent and is associated with high mortality. Perianal involvement is seen in almost one-third of patients with Crohn's disease. Perianal Crohn's disease is a particularly debilitating form of the disease, which requires multidisciplinary care. It may also require profound immunosuppression with biological agents to control disease activity. Opportunistic infections can complicate the disease course in these patients. We present a case of a middle-a

Bioanalytical methods for the detection of antidiabetic drugs: a review.

Type 2 diabetes mellitus, a disease which prevalence has been progressively increasing worldwide, is characterized by chronic hyperglycemia resulting from the combination of inappropriate insulin secretion and/or resistance to insulin action. If left uncontrolled, diabetes is associated with complications such as dysfunction and failure of various organs, and even premature death. Along with lifestyle-modification strategies, several classes of oral antidiabetic agents can be employed for glycemic control. Thus, therapeutic drug monitoring of these drugs is essential to maintain appropriate tr

Optimizing Crops for Biocontrol of Pests and Disease.

Volatile compounds and extrafloral nectar are common defenses of wild plants; however, in crops they bear an as-yet underused potential for biological control of pests and diseases. Odor emission and nectar secretion are multigene traits in wild plants, and thus form difficult targets for breeding. Furthermore, domestication has changed the capacity of crops to express these traits. We propose that breeding crops for an enhanced capacity for tritrophic interactions and volatile-mediated direct resistance to herbivores and pathogens can contribute to environmentally-friendly and sustainable agr

Comparative susceptibility of adult house fly (Diptera: Muscidae) and an important predator, Carcinops pumilio (Coleoptera: Histeridae), to Beauveria bassiana (Hypocreales: Cordycipitaceae).

House flies (Musca domestica L.) (Diptera: Muscidae) are challenging pests to control. Biological control using Carcinops pumilio beetles may help to reduce house fly populations. However, it is unknown if C. pumilio beetles are compatible with Beauveria bassiana, another house fly biological control option. Five strains of commercially available (GHA, HF23, and L90) and newly discovered (NFH10 and PSU1) strains of B. bassiana were used to test the comparative susceptibility of adult house flies and adult C. pumilio using different laboratory exposure methods. Adult house flies were susceptibl
