Fucoidan attenuates hyperuricemia and renal inflammation in association with gut microbiota remodeling and enrichment of Lactobacillus plantarum.

Fucoidan has shown potential for the management of hyperuricemia (HUA), although its underlying mechanisms remain poorly understood. In this study, fucoidan from Undaria pinnatifida was fractionated, and the 0.8 M NaCl-eluted fraction (UPF-8) was evaluated in a mouse model of HUA. UPF-8 reduced serum uric acid, blood urea nitrogen, and creatinine levels, decreased adenosine deaminase and xanthine oxidase activities, and alleviated renal injury. These effects were accompanied by changes in serum metabolites related to purine and amino acid metabolism and the TCA cycle, improved intestinal barri…

Hyperuricemia in Chronic Kidney Disease: Clinical Evidence and the Missing Role of Urinary Uric Acid Handling.

Hyperuricemia is common in chronic kidney disease (CKD); however, its role in CKD progression remains unclear. A reduced kidney function elevates serum uric acid levels through impaired urinary excretion, while uric acid may injure the kidney. Although early studies suggested renoprotection from urate-lowering therapy (ULT), large randomized trials have not consistently shown that lowering serum uric acid levels slows the progression of CKD. One reason may be that the measurement of serum uric acid alone cannot distinguish between the underlying mechanisms, such as overproduction, reduced glom…

Impact of hyperuricemia on cardiac events in stable coronary artery disease patients with or without renal dysfunction.

Patients with hyperuricemia may have an elevated risk of cardiovascular events. Owing to the relationship between uric acid (UA) levels and renal function, the impact of hyperuricemia may differ depending on the presence or absence of renal dysfunction. This study aimed to evaluate the impact of hyperuricemia on cardiac events in coronary artery disease (CAD) patients with or without renal dysfunction. This single-center observational study enrolled patients with stable CAD who underwent percutaneous coronary intervention. Hyperuricemia (high UA) was defined as a UA level ≥ 7 mg/dL according t…

Discovery of C2: A Novel Lead Compound for the Treatment of Gout and Hyperuricemia via Multi-Pathway Inhibition.

Gout and hyperuricemia, prevalent metabolic disorders, are driven by elevated serum uric acid (UA) levels and subsequent monosodium urate crystal deposition, which provoke inflammatory responses and joint damage. Current therapeutic options remain unsatisfactory, underscoring the urgent need for novel agents with improved efficacy and safety profiles. In this study, we aimed to discover a new lead compound capable of concurrently addressing both hyperuricemia and associated inflammatory pain. Two novel compounds were designed and synthesized, among which C2 was identified as a promising candid…
