Impact of Glycosylation of Apolipoprotein D on Its Interaction with Gold Nanoparticles: Insights from Molecular Dynamics Simulations.

Efficient delivery of nanoparticles (NPs) as carriers for biochemical substances is crucial in various biomedical applications. In this study, we systematically investigate the interactions between glycosylated and nonglycosylated forms of Apolipoprotein D (ApoD) with gold nanoparticles (AuNPs) functionalized with different polymer coatings, including polyethylene glycol (PEG) and zwitterionic polymers. Using all-atom molecular dynamics simulations, we demonstrate that glycosylation significantly enhances the adsorption behavior of ApoD on AuNP surfaces, with the extent of this enhancement bei

Understanding the relationship between eosinophils and chronic obstructive pulmonary disease. A new dawn?

INTRODUCTION: Chronic obstructive pulmonary disease (COPD) is a leading cause of global morbidity and mortality. It is a complex and heterogeneous condition defined by the presence of an incompletely reversible airflow obstruction. AREA COVERED: Airway inflammation and remodeling are central to the pathogenesis of this airflow limitation, and eosinophilic or type 2 inflammation has emerged as a measurable and treatable therapeutic target in those patients in whom it is identified. This review addresses the role of type 2 inflammation in COPD pathogenesis as well as current and future therapeu

Fostering diversity in global genomics: a South Asian perspective.

The 'Global South' with excess of two third of the world population is grossly underrepresented in genetic studies. The lack of genetic information disables these populations to adequately utilize the enormous knowledge available connecting genes with diseases and the practicality of genomics informed patient stratification and precision medicine. Here, we discuss how the genomes from the Global South, including the GenomeIndia Project, a pan India initiative to study India's genetic diversity, would enhance our understanding in disease genomics. GenomeIndia Project ensures a diverse represent

Catecholaminergic polymorphic ventricular tachycardia: A narrative review of recent advances in genetics, mechanisms, diagnosis, and treatment.

Catecholaminergic polymorphic ventricular tachycardia (CPVT) is a rare but potentially lethal inherited arrhythmia syndrome. It is characterized by exercise- or emotion-induced bidirectional or polymorphic ventricular tachycardia, which can lead to recurrent syncope and even sudden death. CPVT is mainly caused by mutations in genes related to calcium homeostasis regulation, with ryanodine receptor 2 and calsequestrin 2 gene mutations being the most common. In recent years, with the development of molecular biology techniques and in-depth clinical research, the understanding of CPVT has been co

Single-cell genotyping and transcriptomic profiling of mosaic focal cortical dysplasia.

Focal cortical dysplasia type II (FCDII) is a cortical malformation causing refractory epilepsy. FCDII arises from developmental somatic activating mutations in mTOR pathway genes, leading to focal cortical dyslamination and abnormal cytomegalic cells. Which cell types carry pathogenic mutations and how they affect cell-type-specific transcriptional programs remain unknown. In the present study, we combined several single-nucleus genotyping and transcriptomics approaches with spatial resolution in surgical cortical specimens from patients with genetically mosaic FCDII. Mutations were detected

The role of imaging parameters in the diagnosis of developmental dysplasia of the hip based on artificial intelligence: A perspective.

Developmental dysplasia of the Hip (DDH) is a common pediatric orthopedic disease, characterized primarily by abnormal development of the hip joint structure. The clinical objective is the early detection, diagnosis, and treatment. Current diagnostic strategies rely on a combination of physical examination and imaging assessment, with particular emphasis on the comprehensive assessment of multiple imaging parameters. In recent years, the integration of Artificial Intelligence (AI) with medical imaging has enhanced the accuracy and objectivity of DDH diagnosis and management. Nevertheless, curr

Functional assessment of all ATM SNVs using prime editing and deep learning.

Ataxia telangiectasia mutated (ATM), a large gene with 63 exons, plays a critical role in the DNA damage response, and its loss of function increases cancer risk and affects the prognosis of cancer patients. However, interpreting the functional impact of ATM variants remains challenging because most are variants of uncertain significance (VUSs). Here, we assessed the functions of all 27,513 possible single-nucleotide variants (SNVs) in ATM. By using prime editing, we experimentally evaluated the effects of 23,092 SNVs on cell fitness in the presence of olaparib, thereby identifying critical re

Epigenetic regulation in spinal muscular atrophy: emerging areas and future directions.

Spinal Muscular Atrophy (SMA) is a neuromuscular disorder precipitated by mutations or deletions in the Survival Motor Neuron 1 (SMN1) gene. Although the SMN2 gene partially compensates for SMN1 functional deficiency, its expression is regulated by complex epigenetic and environmental factors. This review comprehensively elucidates the regulatory mechanisms through which epigenetic modifications-encompassing DNA methylation, histone modifications, and non-coding RNAs-modulate SMN2 gene expression and impact SMA pathogenesis and progression. We also briefly discuss how these epigenetic mechanis

PULSAR: a Foundation Model for Multi-scale and Multi-cellular Biology.

Biology emerges from interactions across physical scales, where molecular interactions drive cellular states, which in turn orchestrate multicellular tissue functions that collectively define health and disease. However, current computational models are often constrained to single scales in isolation, failing to integrate the biology that emerges from lower to higher levels [1]. Here we present PULSAR (Patient Understanding Leveraging Single-cell universAl Representation), a multi-scale and multicellular foundation model architecture that explicitly enables information flow from genes to cells

Biomechanical assessment of Hoffa fat pad characteristics with ultrasound: a narrative review focusing on diagnostic imaging and image-guided interventions.

The infrapatellar fat pad (IFP), a key intra-articular knee structure, plays a crucial role in biomechanical cushioning and metabolic regulation, with fibrosis and inflammation contributing to osteoarthritis-related pain and dysfunction. This review outlines the anatomy and clinical value of IFP ultrasonography in static and dynamic assessment, as well as guided interventions. Shear wave elastography (SWE), Doppler imaging, and dynamic ultrasound effectively quantify tissue stiffness, vascular signals, and flexion-extension morphology. Due to the limited penetration capability of ultrasound im

Induced Pluripotent Stem Cells (iPSC) and Their Use in Disease Modeling.

This paper reviews methodologies for culturing induced pluripotent stem cells (iPSCs) and highlights their applications in disease modeling and regenerative medicine. iPSCs can be reprogrammed from multiple somatic cell sources, including keratinocytes, fibroblasts, peripheral blood mononuclear cells, and urinary epithelial cells. Their ability to differentiate into patient-specific cell types provides unique opportunities to model neurodegenerative, cardiovascular, metabolic, and autoimmune disorders in vitro. Pluripotency is typically induced by the overexpression of four canonical transcrip

Decoding eQTLs: unraveling their role in spondyloarthropathies pathogenesis and potential therapeutics.

Expression quantitative trait loci (eQTLs) represent genetic variants that influence gene expression, providing insights into the mechanisms linking genetic predisposition to complex diseases, including spondyloarthropathies (SpAs). SpAs, encompassing ankylosing spondylitis, psoriatic arthritis, reactive arthritis, and enteropathic arthritis, are related heterogeneous conditions with strong genetic components, particularly in HLA-B27 and non-HLA loci, but phenotypically distinct disorders. Recent advances in genome-wide association studies (GWAS) and transcriptomic studies have shown the poten

Toward universal screening for disease-causing alleles: Mendelian susceptibility to mycobacterial disease as a model.

Advances in genomic technologies, including whole exome and genome sequencing, have transformed diagnosis of monogenic disorders such as inborn errors of immunity (IEIs). In high-consanguinity populations like the United Arab Emirates (UAE), where autosomal recessive disorders are prevalent, early genomic screening shifts care from reactive diagnosis to personalized care. UAE national programs remain limited to premarital or neonatal panels, missing disorders with variable onset or incomplete penetrance. We advocate universal genomic screening to integrate disease-causing alleles into clinical

Magnetic-Driven Hydrogel Robots Loaded with Mitomycin for Active Therapy of Bladder Cancer.

Intravesical drug instillation is a common treatment for bladder cancer, but it suffers from poor targeting and suboptimal patient experience due to the influence of gravity. Here, we innovatively designed a bladder cancer treatment method using sodium alginate hydrogel magnetic robots as mitomycin drug carriers. The magnetic-driven hydrogel robots with mitomycin (MHRMs) are fabricated by uniformly mixing alginate solution, neodymium-iron-boron nanoparticles, and mitomycin, followed by the formation of MHRMs capable of carrying mitomycin in a calcium chloride solution. After being injected int

DGAT: A Dual-Graph Attention Network for Inferring Spatial Protein Landscapes from Transcriptomics.

Spatial transcriptomics (ST) technologies provide genome-wide mRNA profiles in tissue context but lack direct protein-level measurements, which are critical for interpreting cellular function and microenvironmental organization. We present DGAT (Dual-Graph Attention Network), a deep learning framework that imputes spatial protein expression from transcriptomics-only ST data by learning RNA-protein relationships from spatial CITE-seq datasets. DGAT constructs heterogeneous graphs integrating transcriptomic, proteomic, and spatial information, encoded using graph attention networks. Task-specifi

α-Synuclein seed amplification assay positivity beyond synucleinopathies.

Neurodegenerative diseases are increasingly recognized as complex disorders involving multiple protein pathologies, with α-synuclein frequently observed beyond classical synucleinopathies such as Parkinson's disease and multiple system atrophy. Recent advances in seed amplification assays (SAAs) have enabled the highly sensitive and specific detection of misfolded α-synuclein in vivo, particularly in cerebrospinal fluid (CSF). This review focuses on CSF-based α-synuclein SAAs and their application in detecting co-pathology across non-synucleinopathies, including Alzheimer's disease, progressiv

Metabolomics in Endocrinology: The Way Forward.

Metabolomics is a type of laboratory science used to understand the cellular and metabolic defects in any disease process. It comprehensively identifies endogenous and exogenous low-molecular-weight (<1 kDa) molecules or metabolites in a high-throughput manner. Mass spectrometry-based methods are used for metabolomics which can be targeted and non-targeted. Metabolomics workflow consists of sample acquisition, its preparation and extraction, separation, ionisation, data analysis, and metabolite detection and identification. Some of the commonly elevated metabolomes are branched-chain amino aci
