Moderate mental retardation and nonspecific dysmorphic syndrome associated with ring chromosome 9.

A ring chromosome 9 is reported in a 12-year-old, moderately mentally retarded boy. As in other ring chromosome 9 patients, the clinical stigmata are nonspecific and their expressivity is mild. The finding of two normal cells of a total of 121 cells examined favors the hypothesis of a postzygotic, mitotic origin of the ring chromosome formation.

GM1-gangliosidosis: chromosome 3 assignment of the beta-galactosidase-A gene (beta GALA).

The structural gene (beta GALA) coding for lysosomal beta-galactosidase-A (EC 3.2.1.23) has been assigned to human chromosome 3 using man--m … ate fusion experiments utilizing cells from 9 individuals. Enzyme segregation analysis excluded all chromosomes for beta GALA assignment except chromosome 3. Concordant segregation of chromosomes and enzymes in 16 cell hybrids demonstrated assignment of beta GALA to chromosome 3; all other chromosomes were excluded. The evidence suggests that GM1 gangliosidos …

The effect of oxygen concentration on the X-ray induction of chromosome aberrations in human lymphocytes.

In vitro dose--response curves, for unstable chromosome aberration induction in human lymphocytes under conditions of full oxygenation or of anoxia, have been obtained using 250 kVp X-rays. Dicentric yields have been fitted to the quadratic function Y = alpha D + beta D2. An Oxygen Enhancement Ratio (OER) ranging from 7.2 to 2.7 was calculated from the coefficients of these curves possibly indicating that the data do not fit to the dose-modifying model of the oxygen effect although the differences are not statistically significant. A similar analysis of total aberration data yields an OER of 3

Fluorescent light-induced chromosome damage and its prevention in mouse cells in culture.

Twenty-hour-exposure to fluorescent light produces chromatid breaks in a line of adult mouse lung cells grown in Dulbecco-Vogt medium supplemented with fetal bovine serum. The light-induced damage appears to be enhanced by increasing the concentration of oxygen in the gas phase of the culture. The effective wavelength(s) of light is in the visible range between 400 and 450 nm andis probably the mercury emission peak at 405 or 436 nm. Addition of catalase or glutathione with ascorbic acid to the culture medium reduced the number of chromatid breaks to a level not significantly different from th

Spontaneous Chromosome Breakage at Male Meiosis Associated with Male Recombination in DROSOPHILA MELANOGASTER.

An inbred line (OK1) of Drosophila melanogaster , recently derived from a natural population in Oklahoma, has been found by Woodruff and Thompson to exhibit a low frequency of spontaneous male recombination when outcrossed to marker stocks. There is also a reciprocal-cross effect, such that recombination is found only if OK1 males are used in the initial cross. When OK1 females are used, however, male recombination is again found if their male progeny are used for a subsequent cross.-In the present cytological analysis, chromosome behavior at male meiosis was studied in reciprocal crosses betw

True hermaphroditism: a clinical description and a proposed function for the long arm of the Y chromosome.

True hermaphroditism is a very rare form of ambiguous genitalia characterized by the presence of both ovarian and testicular tissue in the same individual. Sixty percent of these patients have a 46,XX karyotype; however, most have H-Y antigen detectable. Since H-Y antigen is thought to be the gene product of the short arm of the Y, then the short arm must be present somewhere in the karyotype. Products of the short arm loci are thought to initiate testicular differentiation and male determination. The long arm of the Y has not been detected in true hermaphrodites. Testicular histology in true

Chromosome analyses in man in the course of chemoprophylaxis against tuberculosis and of antituberculosis chemotherapy with isoniazid.

Cytogenetic analyses were carried out in lymphocytes of 37 persons before and during a primary chemoprophylaxis or a preventive chemotherapy with isoniazid as well as of 30 patients suffering from active pulmonary tuberculosis before and during a chemotherapy with isoniazid in combination with two other antimycobacterial drugs. The results of the four participating research groups consistently reveal no indication for a chromosome damaging activity of isoniazid, of its metabolites in the human organism and of the administered drug regimens.

Y autosome translocation and complex chromosome rearrangement in cri du chat syndrome.

An unbalanced Y autosome translocation t(5;Y) and an apparently balanced translocation t(2;13) are identified with the Q and R banding in a 7-year-old boy with severe encephalopathy and a multiple malformation syndrome. At birth, the clinical diagnosis of 'cri du chat' syndrome based on the characteristic crying was not confirmed after karyotyping, using conventional staining techniques.

The early childhood development of 17 boys with sex chromosome anomalies: a prospective study.

Seventeen boys with sex chromosome anomalies were identified at birth and followed up developmentally for two to five years. The 17 included 12 boys with 47,XXY, four with 47,XYY, and one with 46,XY/XXY mosaicism. None had major disabilities in physical, intellectual, or emotional growth. Some of the boys with 47,XXY were characterized as tall, inactive, poorly organized in motor functions, placid, and delayed in language; two of the four with 47,XYY were impulsive at 1 year of age. These deviations from normal were relatively mild; the boys were not found to be at high risk for grossly abnorm

Risk for recombinants in pericentric inversions of the (p11 leads to q21) region of chromosome 18.

A child with female hypospadia complicated by bilateral hydronephrosis, hydroureter, and hydrocolpos was heterozygous for a pericentric inversion of chromosome 18, 46,XX,inv(18)(p11q21). The normal mother and her father had the same inversion. The abnormal phenotype of the girl could be due to undetectable recombination or to a position effect. She had a low level of the enzyme peptidase-A whose locus is on 18q, while her mother and grandfather had normal levels. The two other cases of familial inversions for chromosomes 18 in the literature both involve the same (p11 leads to q21) region. The…

Idiopathic acquired refractory sideroblastic anemia: Banded chromosome analysis in six patients.

Chromosome analyses with banding were performed on six patients with idiopathic acquired refractory sideroblastic anemia (IARSA). One patient was found to have an extra chromosome No. 8, but had a normal level of red cell glutathione reductase. Bone marrow chromosomes from the other patients showed a normal karyotype. 28 patients with IARSA, including our 6 patients, have had chromosomal analyses. Two consistent chromosomal abnormalities have been described: a + 8 in three patients and a 20q- in three others. Despite the presence of chromosomal abnormalities in abouut one half of the patients,…

Genetic counselling and prenatal diagnosis for chromosome anomalies. Use of study of spontaneous abortions.

Genetic counselling for chromosome abnormalities must take into account the fact that such anomalies may develop as an error in oogenesis or spermatogenesis or may be transmitted from parents. Since many chromosomally abnormal embryos are spontaneously aborted, parents with repeated unfavorable outcomes of pregnancy or difficulty in conceiving should be studied by karyotype. The relationship between chromosomal abnormalities and various environmental or biologic factors are discussed. The outcome of a previous pregnancy may indicate the necessity of subsequent fetal evaluation even with chromo

Interstitial deletion of the long arm of chromosome no. 7 (7q-) in an infant with multiple anomalies.

An infant is reported with partial deletion of the long arm of chromosome no. 7. She presented with hypertonia, seizures, feeding difficulty, and multiple congenital anomalies. The abnormalities include low-set dysplastic ears, hypoplastic orbital bones, upslanting and small palpebral fissures, prominent cheeks with a relatively large mouth, micrognathia, abnormal creases of the hands and a congenital heart defect. With age her hypotonia and feeding difficulty have improved. Her mother has no detectable chromosome abnormality.

X-chromosome inactivation in human liver: confirmation of X-linkage of ornithine transcarbamylase.

Histochemical assay for ornithine transcarbamylase (OTC) activity in fixed frozen hepatic sections from a woman heterozygous for OTC deficiency revealed two populations of hepatocytes: those with normal activity and those with no activity. This observation, in conjunction with data from previous family studies, confirms the hypothesis that the gene for OTC is X-linked. It also provides the first cytologic demonstration of cellular mosaicism for a liver-specific cell product.

Supernumerary bisatellited chromosome in a family ascertained through a patient with Sturge-Weber syndrome.

The presence of a structurally abnormal extra chromosome in a patient with Sturge-Weber syndrome and several members of her family is described. With routine techniques the abnormal chromosome is slightly submetacentric, of the size of a G group chromosome and shows satellites on both arms. C-banding suggested the presence of 2 centromeric regions rather than one, and to explain this finding, in addition to the segregation of the abnormal chromosome through 3 generations and why only one centromere is visible with the usual cytogenetic technique, an hypothesis is advanced suggesting that it re
