To investigate leukemogenesis in atomic bomb (A-bomb) survivors, chromosome aberrations in bone marrow cells, and T- and B-lymphocytes from … ysis revealed a higher incidence of translocations in 50 bands, including those containing cellular oncogenes such as 8q22, 8q24, and 9q34. Of these 50 bands, 20 were matched with bands specific for leukemia and cancer and 14 with constitutive fragile sites. In leukemic marrow, all 10 patients who had been exposed to radiation of more than 200 rad and then …

The incidence of clinical prostate cancer varies across countries and ethnic groups. Genetic and epigenetic factors have been suggested as possible explanations to these variations, although no mesological factors with clearly significant effects have been identified. About 20% of patients with prostate cancer have a family history for this disease. Several studies have reported links between prostate cancer and breast cancer, suggesting that the same loci may predispose to both diseases. Identification of one or more inherited genes associated with an increased risk of prostate cancer in some

In this issue of Blood, de Yébenes et al identify miR-217 as a promoter of the germinal center reaction, partly by suppressing the DNA damage response. Overactivity of miR-217 is hence potentially dangerous, and, indeed, its uncontrolled expression causes lymphomas.

Soft tissue tumors with chondroid matrix represent a heterogeneous group with persistent diagnostic challenges. Advances in molecular diagnostics have identified recurrent gene fusions in several chondroid neoplasms, predominantly involving FN1. Here, we report two cases of chondroid tumors harboring a novel FOS::PABPN1 fusion. The patients were young and presented with small, deep, peri-osseous nodules located in the extremities. Histologically, the tumors were well-circumscribed and lobulated. Both cases presented fibro-cartilaginous and spindle cell areas. No cytonuclear atypia, mitoses, or

IN BRIEF: During pregnancy and delivery, the myometrium was affected by hypoxia stress, which acts as a regulator of cell proliferation. The proliferation of uterine smooth muscle cells in pregnant mice was inhibited under hypoxia, which was related to the up-regulated autophagy through the mTOR pathway.
ABSTRACT: Hypoxia is closely associated with physiological and pathological conditions in the human body, and the myometrium is affected by hypoxic stress during pregnancy and delivery. Autophagy is a catabolic pathway involved in the regulation of apoptosis, proliferation, and migration of a

AIMS: Ectomesenchymal chondromyxoid tumour (ECT) is a rare benign intraoral tumour which almost exclusively presents as a small mass of the anterior dorsal tongue. Recently, the RREB1-MRTFB (previously known as MKL2) fusion gene has been identified in 90% of ECTs, all located in the tongue, emphasising its genetic distinctiveness. Here, we report two mesenchymal tumours involving the superior mediastinum of adult women with RREB1-MRTFB fusions.
METHODS AND RESULTS: Both tumours presented as well-circumscribed paravertebral masses that were clinically suspected to be schwannoma. After fragment

BACKGROUND: The bcl-2 proto-oncogene codes for a protein which appears to block apoptosis. In our study, we examined bcl-2 protein expression in cervical squamous metaplasia, cervical intraepithelial neoplasia (CIN) and microinvasive squamous carcinoma with the aim of identifying a relationship between bcl-2 protein expression and neoplastic development and progression.
MATERIALS AND METHODS: Cervical bioptic samples were obtained from 86 white women, selected consecutively from our Colposcopic Service from January 1993 to June 1994, because of abnormal pap- smear suspicious for cervical dysp

Cutaneous melanoma is becoming increasingly common. Genetic and environmental factors are thought to play a role in its pathogenesis. We have previously shown that normal human melanocytes strongly express the oncoprotein, Bcl-2. To determine the role of Bcl-2 in melanocytic tumors, we studied human benign nevi and melanomas for expression of Bcl-2 protein using immunohistochemistry. Our results show that benign melanocytes from 3 of 4 normal skin biopsies and 5 of 7 common acquired nevi strongly express Bcl-2. Conversely, only 3 of 23 primary cutaneous melanomas and 3 of 9 metastatic melanoma

The rat sarcoma virus oncogene (RAS) is one of the most frequently mutated drivers in human cancers. Developing targeted therapies against RAS has been challenging due to its structure. The recent breakthroughs with covalent Kirsten RAS (KRAS) inhibitors represent a key milestone in targeting mutant KRAS proteins. However, drug resistance remains a significant obstacle. The proteolysis-targeting chimeras (PROTACs), which degrade mutant KRAS proteins, offer a promising strategy to overcome resistance and expand therapeutic options. This review covers the structural basis of KRAS and signaling n

Bladder cancer (BCa) is among the most prevalent neoplasms of the urinary and reproductive systems, ranking ninth in overall cancer incidence. Existing treatment modalities have not yet achieved a reduction in the elevated incidence and fatality rates of BCa. Consequently, enhancing our comprehension of the molecular pathways involved in the onset and progression of BCa to formulate novel and successful therapeutic strategies is a prominent area of research. Post-translational modifications (PTMs), essential regulators of protein structure, function, and diverse biological processes, play pivo

Osteosarcoma remains the most common primary malignant bone tumor, and survival has improved little over recent decades because of metastasis and therapeutic resistance. Flavones exhibit diverse anti-osteosarcoma activities by suppressing proliferation, inducing apoptosis, ferroptosis and autophagy, inhibiting metastasis, and modulating oncogenic signaling pathways, including PI3K-Akt, Wnt-β-catenin, STAT3, MAPK, and NF-κB. This review summarizes the cell-line-specific molecular mechanisms of representative flavones, critically evaluates current experimental limitations, and discusses strategi

In 2018, there were globally about 350,000 deaths from cervical cancer, despite the fact that the disease can be prevented by screening and in spite of the fact that highly effective human papillomavirus (HPV) tests became available some 25 years ago. WHO recommended HPV as the primary choice for cervical screening in 2014 (WHO 2014). In this chapter, I will outline the major evidence base behind HPV-based screening, discuss some of the major bottlenecks that has hampered the use of HPV-based screening for prevention of cervical cancer, and point out recent developments that may overcome these

Cutaneous human papillomaviruses (HPVs), particularly those of the β-genus, have traditionally been viewed as harmless constituents of the skin microbiome. However, a debate has emerged over the past few years regarding their possible contribution to cancer development. In immunocompetent hosts, commensal β-HPVs may contribute to immune education and cutaneous homeostasis. In contrast, under immunosuppression, their increased viral load (combined with UV-induced DNA damage) can promote early oncogenic events and facilitate cancer initiation, through a "hit-and-run" mechanism. This context-depe

The success of immunotherapies such as immune checkpoint blockade and engineered T-cells has demonstrated the immune system's anticancer potential. Although prophylactic vaccines against oncogenic viruses such as human papillomavirus prevent cancer by blocking infection, therapeutic vaccines targeting tumour-associated or tumour-specific antigens (TAAs/TSAs) have not reliably produced meaningful tumour regressions. Advances in DNA sequencing and computational pipelines for predicting individual, MHC-restricted neoantigens now enable personalized cancer vaccines (PCVs). By targeting neoantigens

Mirdametinib is an MEK1/2 inhibitor having therapeutic potential for the treatment of neurofibromatosis type 1 (NF1)-associated plexiform neurofibromas (PNs). NF1 is a rare and progressive genetic disorder. As a selective MAPK pathway modulator, it targets the dysregulated RAS (Rat Sarcoma viral oncogene homolog)/RAF (Rapidly Accelerated Fibrosarcoma)/MEK (Mitogen-activated protein kinase)/ (ERK) Extracellular signal-regulated kinase) signaling cascade, which plays a crucial role in NF1 tumor growth. PN, often progressive and disfiguring, significantly impairs quality of life (QOL), and it can

p16/Ki-67 dual stain is a biomarker-based test that can identify oncogenic transformation in cervical cells with higher sensitivity than cervical cytology, using the same samples taken for human papillomavirus (HPV) testing and liquid-based cytology. Dual stain is approved by the US Food and Drug Administration (FDA) for triage of women with positive results by primary HPV testing or by HPV/cytology co-testing and has recently been incorporated into management guidelines. In this review, we summarize the data showing the utility of dual stain in detecting precancers, reducing the number of unn

Translocations involving immunoglobin (IG) are common in B-cell neoplasms. These IG translocations lead to the disposition of the enhancer or promoter of an IG locus, typically IGH, to a proto-oncogene, resulting in the elevated expression of the cancer gene. IG fusions play an important role in the diagnosis, prognostication, and therapy selection of B-cell lymphomas. A t(14;22)(q32;q11) translocation involving IGH and IGL is rare in lymphomas. We report herein clinicopathological characteristics, response to treatment, and outcomes of a first splenic marginal zone lymphoma case with a t(14;2

Epidermodysplasia verruciformis (EV) is a rare genodermatosis characterized by chronic infection with human papillomavirus (HPV), leading to disseminated flat-topped papules and pityriasis versicolor-like lesions. Patients with EV are predisposed to developing non-melanoma skin cancers, particularly squamous cell carcinoma (SCC) in sun-exposed areas. Currently, there is no definitive cure for EV, and management remains largely supportive and symptomatic. We report a case of EV in a 38-year-old male, presenting with classical clinical features and histopathological findings, complicated by the

Diethylnitrosamine (DEN) is a chemical hepatocarcinogenic agent that triggers a large array of oncogenic mutations after a single injection. Initiated hepatocytes subsequently undergo clonal expansion within a proliferative environment, rendering the DEN model a comprehensive carcinogen. In rodent studies, DEN finds extensive utility in experimental liver cancer research, mimicking several aspects of human hepatocellular carcinoma (HCC), including angiogenesis, metabolic reprogramming, immune exhaustion, and the ability to metastasize. Beyond the wealth of scientific insights gleaned from this
