Cystatin C, a protein inhibitor of lysosomal cysteine proteinases, was demonstrated by immunohistochemical techniques to be present in the birefringent amyloid deposits of the small arteries in the cerebrum, cerebellum, and leptomeninges of 10 Icelandic individuals with hereditary cerebral hemorrhage with amyloidosis. Specimens from other organs were investigated in one of the patients, and amyloid angiopathy characterized by an immunoreactivity of cystatin C was found in a submandibular lymph node. No immunoreactivity of amyloid fibril protein AA, kappa or lambda immunoglobulin light chain, o…

The clinical picture of 15 patients (10 male, five female) with amyloid arthropathy secondary to chronic renal failure treated with haemodialysis has been studied. The average period of haemodialysis was 10.8 years. Joint symptoms appeared between three and 13 years after starting haemodialysis. No patient had renal amyloidosis. Early symptoms were varied and often overlapped: knee swelling (seven patients), painful and stiff shoulders (seven), and carpal tunnel syndrome (six) were the most prominent. Follow up showed extension to other joints. Joint effusions were generally of the non-inflamm…

We studied the clinical and radiological features of pulmonary metastases present in 4 out of 22 patients with medullary carcinoma of thyroid. Two patients presented with common metastases: macronodules in one, and micronodules in the other one. The other two patients presented initially with reticulonodular perihilar lesions on chest X-ray, leading to the diagnosis of sarcoidosis in both of them. The observation of such pulmonary metastases is original: initially latent, they progress very slowly, and they correspond to a lymphangitic spread of the tumour with amyloid deposition in peribronch…

New historic, morphologic, localisatory, chemical, etiological and pathogenetical observations urge the revision of problems of the so-called amyloidosis and suggest a new logical terminology. Cerebral amyloidosis, which corresponds to the clinical picture of senile dementia and Alzheimer's disease, is the local manifestation of a generalized systemic degeneration typified by amyloidotic deposits in aged persons. Chronic lingering tuberculosis, traumatic cerebral injury, genic aberrations, ionising radiations as well as a chronic "slow" virus infection (in cases of Creutzfeldt-Jakob's disease)…

Oxidative stress is observed in Alzheimer's disease (AD) brain, including protein oxidation and lipid peroxidation. One of the major pathological hallmarks of AD is the brain deposition of amyloid beta-peptide (Abeta). This 42-mer peptide is derived from the beta-amyloid precursor protein (APP) and is associated with oxidative stress in vitro and in vivo. Mutations in the PS-1 and APP genes, which increase production of the highly amyloidogenic amyloid beta-peptide (Abeta42), are the major causes of early onset familial AD. Several lines of evidence suggest that enhanced oxidative stress, infl…

Pancreatic lipase (PL) and cholesterol esterase (CE) are dominant lipid-hydrolyzing enzymes involved in the digestion and absorption of lipids, making them attractive targets for obesity prevention. β-lactoglobulin (β-LG) can transform into amyloid fibrils with potential inhibitory activity against PL and CE because of its amphiphilic and self-assembling properties. In this study, Dielectric Barrier Discharge atmospheric cold plasma treatment was employed to regulate the fibrillation process of β-LG (P-β-LGF) at three stages of fibril growth, namely, the initial stage (t0), the end of the lag …

INTRODUCTION: Plasma phosphorylated tau (p-tau)217 is increasingly used to triage amyloid testing, but the biological and clinical meaning of discordant plasma p-tau217 and amyloid positron emission tomography (PET) results in cognitively normal adults remains uncertain.
METHODS: We analyzed cognitively normal participants from the Chinese Preclinical Alzheimer's Disease Study (CPAS; n = 594) and the Alzheimer's Disease Neuroimaging Initiative (ADNI; n = 475). Amyloid PET positivity was defined as Centiloid ≥ 20. Cohort-specific p-tau217 positivity was defined using PET-anchored cutoffs with …

BACKGROUND: Amyloid-β (Aβ) accumulation begins before cognitive impairment, highlighting the need for scalable blood-based biomarkers to identify individuals at risk during the preclinical stage of Alzheimer's disease.
OBJECTIVE: To evaluate whether plasma %p-tau217 detects Aβ-PET positivity and whether longitudinal plasma %p-tau217 change is associated with early Aβ accumulation in cognitively unimpaired participants from the Alzheimer's Disease Neuroimaging Initiative (ADNI).
METHODS: This retrospective longitudinal study included 98 cognitively unimpaired ADNI participants with plasma %p-…

Lecanemab and donanemab are the first disease-modifying therapies for early Alzheimer's disease. These humanized IgG1 antibodies promote the clearance of aggregated amyloid-beta peptides and, in the pivotal CLARITY AD and TRAILBLAZER-ALZ 2 trials, significantly delayed cognitive and functional decline, with an approximately 30% relative risk reduction for the clinical stage progression; however, the clinical meaningfulness remains contested: critics point to the small between-group difference in relation to the total score. This is weighed against the first disease-modifying effect and the fir…

OBJECTIVE: Abnormalities in posteriorly dominant resting-state eyes-closed electroencephalographic (rsEEG) alpha rhythms are associated with cerebrospinal fluid (CSF) tau neuropathology and Mini-Mental Status Examination (MMSE) score, reflecting global cognition, in patients with mild cognitive impairment due to Alzheimer's disease (MCI-AD). The present study examined the relationships between white matter cerebrovascular lesions (WMLs) and CSF amyloid-tau markers, posterior rsEEG alpha rhythms, and cognitive status in patients with MCI-AD versus those with mild cognitive impairment not due to…

Alzheimer's disease (AD) is a progressive neurodegenerative condition with a rising global prevalence. Although clinical immunoassays offer high sensitivity, their dependence on costly and centralized infrastructure hinders widespread screening in resource-limited regions. Herein, we developed an electrical sensing platform utilizing molecularly imprinted polymers (MIPs) as synthetic receptors to detect a critical biomarker, amyloid beta 40 (Aβ40). The analytical performance was evaluated using impedance measurements through a high-precision benchtop LCR meter and a cost-effective miniaturized…

The pathogenesis of Alzheimer's disease (AD) involves amyloid beta (Aβ)-induced oxidative stress, apoptotic cell death, and neuroinflammation, contributing to neuronal dysfunction. In our study, a differentiation protocol using retinoic acid was applied to SH-SY5Y cells to generate a neuron-like phenotype, and the neuroprotective efficacy of Escin was investigated by inducing Aβ1-42-mediated cytotoxicity. The experimental protocol involved an initial treatment with 2 µM Escin prior to Aβ1-42 application. Cell viability, intracellular reactive oxygen species (ROS), apoptosis, and inflammatory m…

Alzheimer's disease (AD) is a neurodegenerative disorder primarily characterized by cognitive decline. Its pathogenesis is complex, involving multiple pathological processes, including amyloid-β (Aβ) deposition, neuroinflammation, and synaptic dysfunction. In recent years, the role of mesenchymal stem cells (MSCs) in AD therapy has garnered significant attention. MSCs, through their multi-directional differentiation potential and paracrine effects, exhibit remarkable neuroprotective and anti-inflammatory properties, influencing AD progression. This review summarizes the potential mechanisms an…

INTRODUCTION: Neuronal networks enable brain's information processing through a well-coordinated activity. Disruptions in this activity can impair key brain functions such as synaptic plasticity and long-term memory. Such dysfunctions are relevant to the cognitive deterioration in Alzheimer's disease (AD). Neuronal circuit alterations in AD are associated with amyloid-β (Aβ) extracellular accumulation across multiple brain regions involved in cognitive regulation. Although several studies have analyzed network topology and examined anatomical, functional, and effective connectivity to understa…

BackgroundApathy is a common neuropsychiatric symptom in Alzheimer's disease (AD) and has been linked to greater levels of AD biomarkers (amyloid-β, tau). However, it is unclear whether early patterns of amyloid deposition may impact development of apathy symptoms in the future.ObjectiveWe sought to examine whether amyloid-β levels, both globally and in brain regions associated with motivation, could predict future apathy symptoms.MethodsParticipants (n = 199, mean age = 79.9) were part of the Harvard Aging Brain Study, a longitudinal observational cohort of individuals without cognitive or ps…

A key characteristic of Alzheimer's disease (AD) is the build-up of amyloid-β (Aβ) fibrillar aggregates, commonly known as plaques, within the brain. Current diagnostic tools, such as MRI, CT, and PET, primarily detect neurodegeneration or plaque burden, limiting their utility to advanced stages of the disorder. Thus, there is a strong need for sensitive approaches that can enable detection at the earliest. Aβ aggregation begins with the formation of small, lipophilic oligomers characterized by antiparallel β-sheet structures. Emerging evidence suggests that these oligomers associated with exo…

BackgroundHippocampal synaptic dysfunction driven by toxic amyloid-β oligomers (AβO) is an early event in the progression of neurodegeneration and cognitive decline in Alzheimer's disease (AD). Non-invasive photobiomodulation therapy (PBM) is a promising intervention that has been shown to reduce amyloid and tau pathology, improve synaptic function, and preserve hippocampal neurogenesis in animal models of AD. Nano-pulsed laser therapy (NPLT) is a type of PBM therapy using pulsed 808 nm near-infrared laser light and optoacoustically generated ultrasound waves to stimulate deeper brain structur…

Background and Objectives: Dual-phase amyloid PET imaging has been proposed to provide complementary information regarding amyloid burden and cerebral perfusion. This exploratory pilot study evaluated whether semiquantitative parameters derived from dual-phase PET/CT could differentiate individuals operationally classified as Alzheimer's disease with mild functional impairment (AD-MFI) from those with mild cognitive impairment (MCI). Materials and Methods: Twenty-four participants (AD-MFI, n = 19; MCI, n = 5) underwent dual-phase amyloid PET/CT and structural MRI. Early phase SUV (eSUV), delay…
