A colloidal gold immunochromatographic test strip based on McAbs anti-S protein to detect porcine epidemic diarrhea virus.

Porcine epidemic diarrhea (PED) is a highly contagious intestinal disease caused by the porcine epidemic diarrhea virus (PEDV), resulting in considerable economic losses for the global swine industry. Rapid diagnosis of PEDV is essential to develop effective prevention and control measures. The PEDV S1 protein serves as an efficient target for PEDV diagnosis. In this study, we successfully produced three monoclonal antibodies (McAbs) specific against the PEDV S1 protein, namely, H6, G3, and F2. ELISA showed that McAb F2 was IgG2a, McAbs H6 and G3 were IgG1, and the antigen site of McAb F2 was

Ultrasensitive Detection of Porcine Epidemic Diarrhea Virus Infections Using Multivalent DNA Nanostructure-Enabled Lateral Flow Assay.

Early, rapid on-site detection of viral pathogens in livestock is critical to prevent disease spread, minimize economic losses, and safeguard food security. However, most current diagnostics are limited to centralized laboratories or require specialized equipment such as electrochemical readers, with a few hours to multi-day turnaround times that hinder field deployment. Here, we introduce a DNA nanostructure-enabled lateral flow assay (LFA) for rapid, ultrasensitive detection of porcine epidemic diarrhea virus (PEDV) directly on farms. This assay employs net-shaped DNA nanostructures presenti

Validation of a Bayesian Ebola Diagnostic Model Using Data from the Tenth Epidemic of 2018 to 2020 in the Democratic Republic of Congo.

Background/Objectives: Diagnosing Ebola virus disease (EVD) based solely on clinical signs remains challenging in settings where its manifestations overlap with other endemic infections. This study aimed to validate a Bayesian clinical diagnostic model to support decision-making for suspected EVD cases in resource-limited settings without access to laboratory confirmation. Methods: We conducted a retrospective analysis using data from the 10th Ebola outbreak in the Democratic Republic of the Congo (2018-2020). Clinical information from 450 suspected and laboratory-confirmed EVD cases was evalu

Early-life nutritional environment is associated with late-life cognition in the Health and Retirement Study, a pellagra epidemic natural experiment.

Early-life exposures are important to several late-life health outcomes. We sought to study the effect of an in utero nutritional environment and its interaction with Alzheimer's disease (AD) genetic risk on late-life cognitive function. We used a natural experiment created by the pellagra epidemic, a nutritional disease caused by a vitamin B3 deficiency, to evaluate the association between in utero pellagra epidemic exposure and late-life cognitive function in the Health and Retirement Study (N = 18,285). We also evaluated whether the in utero exposure could modify the AD polygenic score's (P

Comparison of excess deaths and laboratory-confirmed COVID-19 deaths during a large Omicron epidemic in 2022 in Hong Kong.

BACKGROUND: Using an elimination strategy, Hong Kong was able to minimise COVID-19 mortality in 2020 and 2021, but a large epidemic caused by the Omicron variant occurred in 2022. We aimed to estimate the overall, age-, sex-, epidemic period- and cause-specific excess mortality in 2022 in Hong Kong and compare excess mortality to laboratory-confirmed COVID-19 mortality. METHODS: We used negative binomial regression analysis to model time series of weekly all-cause and cause-specific deaths from 2010 to 2021 to predict the weekly number of deaths in 2022 against counterfactual baselines projec

Equity in epidemic response: an action-oriented framework for guiding public health in equitable responses to major infectious disease emergencies.

BACKGROUND: A rapid and equitable response is paramount to mitigating the spread and impact of an infectious disease public health emergency. Unfortunately, public health responses often integrate equity as a secondary component rather than a foundational one-a decision that can result in disproportionate effects of the epidemic on vulnerable populations and that may further fuel or worsen the ongoing health emergency. This paper introduces a framework grounded in health equity principles to guide the design and implementation of response efforts during infectious disease emergencies. METHODS

COVID-19 Double Annual Epidemic Peaks in Summer and in Winter from 2022, Irrespective of the Rate of Mask Wearing and Vaccination.

Although vaccination for COVID-19 and mask wearing were two of the main preventive measures against infection, their impact is unclear. In the present study, by using national surveillance data in Japan, we compared the incidence rate and weekly case increase ratios of COVID-19 with the domestic stocks of masks and vaccination coverage. The trajectory of epidemic growth increased rapidly in the summer of 2021, concomitant with the launch of the mass national vaccination program. The most rapid spread of the epidemic was found in 2022, approximately 6 months after the national mass vaccination

Bacteriophage M13KE as a nanoparticle platform to display and deliver a pathogenic epitope: Development of an effective porcine epidemic diarrhoea virus vaccine.

Porcine epidemic diarrhoea virus (PEDV) is a porcine enteric coronavirus, outbreaks and epidemics of which have caused huge economic losses to the livestock industry. The disadvantage of existing PEDV vaccines is that the unstable efficacy and high cost limit their widespread use. Therefore, there is an urgent need to develop a recombinant transgenic vaccine candidate for PEDV. In this study, three linear epitopes on the PEDV spike (S) were screened using peptide scanning. The screened epitopes were linked to targeting peptides for lung and intestinal epithelial cells, respectively, and displa

Novel KPC-2 variants and epidemic ST463 clones underlie ceftazidime/avibactam resistance in carbapenem-resistant Pseudomonas aeruginosa.

Carbapenem-resistant Pseudomonas aeruginosa (CRPA) presents a serious clinical challenge due to limited treatment options. Ceftazidime-avibactam (CZA) is a critical last-resort agent against multidrug-resistant Gram-negative pathogens; however, resistance is emerging, particularly mediated by blaKPC variants. In this study, we investigated the prevalence and mechanisms of CZA resistance among CRPA isolates collected from two tertiary hospitals in Hangzhou between 2021 and 2022. A total of 273 non-duplicate CRPA isolates were screened, among which 13.9% (38/273) exhibited CZA resistance (MIC ≥

Molecular basis for the increased fusion activity of the Ebola virus glycoprotein epidemic variant A82V: Insights from simulations and experiments.

During the 2013-2016 Ebola virus (EBOV) epidemic in Western Africa, an A82V mutation emerged in the envelope glycoprotein (GP) that persisted in most circulating isolates. Previous studies demonstrated that A82V increased GP-mediated membrane fusion and altered its dependence on host factors. The mechanistic basis for these observations, in particular the impact of A82V on the conformational changes in GP that are needed for membrane fusion, has not been evaluated in molecular detail. Here, using molecular dynamics simulations, fluorescence correlation spectroscopy, and single-molecule Förster
