Gut mucosal immune responses - Implications for celiac disease and food allergy.

The intestinal mucosa employs a diverse set of defense mechanisms-from mucosal barrier regulation to immunoregulatory pathways-to maintain homeostasis despite constant exposure to microbial and dietary antigens. Failure to establish immune tolerance to food antigens can lead to pathological conditions such as celiac disease (CeD) and food allergies. This review provides an overview of the sequential processes that support tolerance to food antigens and examines how gene-environment interactions contribute to the pathogenesis of CeD and food allergies. Although the exact nature of the environme

Candida albicans: Insights into the Biology and Experimental Innovations of a Commonly Isolated Human Fungal Pathogen.

Candida albicans is an opportunistic pathogen associated with healthy humans. It is the major causative agent for superficial and invasive candidiasis in immunocompromised individuals globally. Lack of awareness toward fungal infections, poor disease management, and increasing drug resistance have contributed to the burden of C. albicans-associated diseases. C. albicans displays polymorphism, assisting in host interaction, virulence, and sustenance of the pathogen. Well-known polymorphic forms of C. albicans include yeast, hyphal, pseudohyphal, white, opaque, gray, GUT, and chlamydospores. Whi

Genetics and humoral responses of the immune system to Aspergillus fumigatus.

Due to the increasing incidence of invasive aspergillosis (IA) caused by the opportunistic fungal pathogen Aspergillus fumigatus, coupled with rising antifungal resistance and high mortality rates globally, novel antifungal development is a critical priority for reducing disease burden. The opportunistic fungus Aspergillus fumigatus, prevalent in the environment alongside other fungi, infects immunocompromised individuals with weakened immune systems. The pathogen can cause severe diseases with fatal outcomes. Risk factors contributing to the severity of A. fumigatus-associated diseases have b

Trypanosoma cruzi/Triatomine Interactions-A Review.

This review summarizes the interactions between Trypanosoma cruzi, the etiologic agent of Chagas disease, and its vectors, the triatomines, and highlights open questions. Four important facts should be emphasized at the outset: (1) The development of T. cruzi strains and their interactions with the mammalian host and the insect vector vary greatly. (2) Only about 10 of over 150 triatomine species have been studied for their interactions with the protozoan parasite. (3) The use of laboratory strains of triatomines makes generalizations difficult, as maintenance conditions influence the interact

Paternal contributions to mammalian zygote - Beyond sperm-oocyte fusion.

Contrary to a common misconception that the fertilizing spermatozoon acts solely as a vehicle for paternal genome delivery to the zygote, this chapter aims to illustrate how the male gamete makes other essential contributions , including the sperm borne-oocyte activation factors, centrosome components, and components of the sperm proteome and transcriptome that help to lay the foundation for pregnancy establishment and maintenance to term, and the newborn and adult health. Our inquiry starts immediately after sperm plasma membrane fusion with its oocyte counterpart, the oolemma. Parallel to an

Dysferlin Protein-Protein Interaction Pathways in the Organ of Corti and Spiral Ganglion Intersect with Alzheimer's Protein Pathways.

Dysferlin direct protein-protein interactions (PPI) previously have been elucidated with surface plasmon resonance (SPR) and predicted to underlie membrane repair in mechanotransducing myofibrils. In mechanotransducing inner ear hair cells, dysferlin is detected with Z-stack confocal immunofluorescence in the stereocilia and their inserts in the tectorial membrane (TM) co-localizing with FKBP8, consistent with the SPR determination of tight, positively Ca2+-dependent interaction. FKBP8, a direct binding partner of mechanotransducing TMC1, when overexpressed, evokes an elevation in anti-apoptot

Paramyxovirus matrix protein redirects METTL3 for dual regulation of viral replication and immune evasion.

N6-methyladenosine (m6A) epitranscriptomic modifications play crucial roles in regulating both host and viral gene expression. Here, we revealed a novel mechanism by which paramyxoviruses exploit host m6A machinery to simultaneously enhance viral replication and suppress host immunity. Our results demonstrated that the viral matrix protein (M) of bovine parainfluenza virus type 3 (BPIV3) binds to the methyltransferase domain of METTL3 in the nucleus and facilitates its translocation to the cytoplasm through an exportin-1-dependent pathway. This mechanism is conserved across multiple paramyxovi

Metabolic regulation of interferon-mediated innate antiviral immunity.

Respiratory viral infections pose a major global public health challenge, with pathogens such as influenza viruses, novel coronaviruses, and respiratory syncytial virus exerting serious threats to human health. These infections often progress to severe disease in children, the elderly, and individuals with compromised immunity. Due to their anatomical exposure and relatively weak mucosal defenses, the upper respiratory tract-including the nasal cavity and throat-serves as a primary portal of entry for these pathogens. Such infections can lead to otolaryngological conditions such as anosmia and

Aspergillus and host-pathogen interaction: focus on treatment-relevant aspects.

BACKGROUND: Invasive aspergillosis (IA) remains a major cause of morbidity and mortality in immunocompromised individuals, with an expanding spectrum of at-risk populations, particularly in intensive care settings. Despite advances in antifungal pharmacotherapy, treatment outcomes remain suboptimal, and the rise of antifungal resistance highlights the need for adjunctive therapy that leverages host immune mechanisms. The pathogenesis of IA is primarily driven by impaired or dysregulated immune responses to Aspergillus species, mainly Aspergillus fumigatus. Host defence involves a complex inter

The innate immune response to A. fumigatus in alveolar macrophages: Roles of LC3-associated phagocytosis, apoptosis and inflammation.

Aspergillus fumigatus (A. fumigatus) has a life-threatening effect in immunocompromised populations, underscore the vital role played by the interplay between the host immune response and this fungal pathogen. Alveolar macrophage (AM) situated within the alveolar spaces serve as the primal immune cells to engage with inhaled Aspergillus conidia. These AMs deploy various strategies to resist A. fumigatus assaults. Their capacity to eliminate A. fumigatus is contingent upon LC3-associated phagocytosis (LAP), an immunological safeguard against microbial intrusions. Furthermore, the propensity for

Dysregulated macrophage immunity in Helicobacter pylori infection: unveiling mechanistic insights and therapeutic implications.

Helicobacter pylori (H. pylori) is a microaerophilic, gram-negative spirochete that primarily colonizes the human gastric mucosa. It is strongly linked to gastritis, ulcers, and the development of malignant tumors. Macrophages, as one of the key components of the innate immune system, play a crucial role in maintaining immune homeostasis through a range of functions, including pathogen phagocytosis, antigen recognition and presentation, inflammation regulation and tumor immune surveillance. Emerging evidence suggests that H. pylori employs diverse molecular mechanisms to evade immune clearance

Fructose catabolism and its metabolic effects: Exploring host-microbiota interactions and the impact of ethnicity.

Important health disparities are observed in the prevalence of obesity and associated non-communicable diseases (NCDs), including type 2 diabetes (T2D) and metabolic dysfunction-associated steatotic liver disease (MASLD) among ethnic groups. Yet, the underlying factors accounting for these disparities remain poorly understood. Fructose has been widely proposed as a potential mediator of these NCDs, given that hepatic fructose catabolism can result in deleterious metabolic effects, including insulin resistance and hepatic steatosis. Moreover, the fermentation of fructose by the gut microbiota c

Early host-parasite interaction models reveal a key role for fibrinolysis in Fasciola hepatica intestinal migration.

BACKGROUND: Fasciola hepatica is the most common etiologic agent of fasciolosis, a parasitic disease that affects millions of ruminants worldwide and a zoonotic human infection of public health concern. Upon ingestion of infective metacercariae, F. hepatica newly excysted juveniles (FhNEJ) emerge in the duodenum and cross the intestinal wall to initiate a migration route that culminates with their establishment within the hepatic bile ducts. The ability of FhNEJ to exploit the broad-spectrum activities of host plasmin, the central protease of the fibrinolytic system, has been proposed as a str

Blood group antigens and malaria susceptibility.

Blood-group antigens are increasingly recognized as key modulators of malaria susceptibility. This review synthesizes current evidence on how erythrocyte surface molecules shape parasite invasion, cytoadherence, immune evasion, and disease severity. We focus on a curated set of systems, ABO, Duffy, MNS, Gerbich, Knops, Diego, and others, and outline their molecular mechanisms affecting malaria. Population heterogeneity, parasite-host adaptations/counter-adaptations, and clinical implications for risk stratification, transfusion safety, and vaccine design are also discussed.

Redefining the immune landscape of hepatitis A virus infection.

Despite the development of effective vaccines against hepatitis A virus (HAV) infection, outbreaks of acute hepatitis A still occur globally, such that HAV remains a major cause of acute viral hepatitis. Most patients with acute hepatitis A recover spontaneously; however, some adult cases result in acute liver failure due to immune-mediated liver damage. Previous studies suggested that HAV evades the innate immune response through strong counteractive mechanisms, and that HAV-specific CD8+ T cells contribute to liver damage in patients with acute hepatitis A. However, recent research findings

Mechanistic Insights into Hepatic Pathogenesis of Dengue Virus Serotype-2: Host-Virus Interactions, Immune Dysregulation, and Therapeutic Perspectives.

Dengue virus serotype 2 (DENV-2) is a predominant cause of severe dengue and a key determinant of dengue-associated liver injury. This review integrates recent findings on the molecular and cellular mechanisms of DENV-2 hepatotropism, focusing on viral replication, cellular stress responses, and immune-mediated damage. The interplay between hepatocytes, Kupffer cells, and innate and adaptive immune responses, culminating in cytokine storm and immune-mediated hepatocyte apoptosis, is dissected. Integrating in vitro and in vivo findings, this review highlights how viral replication and immune dy

Recent Advances in Biology, Host and Microbe Interactions of the Human Sexually Transmitted Parasite Trichomonas vaginalis.

Trichomoniasis is the most common non-viral sexually transmitted infection in humans, with over 200 million people affected each year. This disease is associated with pre-term birth, low birth weight, and premature membrane rupture. Its causal pathogen, Trichomonas vaginalis (TV), is a prevalent sexually transmitted protozoan parasite that infects the urogenital tract through cytoadherence. TV infection alters TV gene expression and induces host immune responses, while TV-secreted exosomes carry RNA and protein cargoes that mediate extracellular signaling. This review summarizes recent discove

VPS18 contributes to phagosome membrane integrity in Mycobacterium tuberculosis-infected macrophages.

Mycobacterium tuberculosis (Mtb) has evolved to be exquisitely adapted to survive within host macrophages. The capacity to damage the phagosomal membrane has emerged as central to Mtb virulence. While Mtb factors driving membrane damage have been described, host factors that maintain phagosomal integrity or repair Mtb-induced damage to contain the pathogen remain largely unknown. We used a genome-wide CRISPR screen to identify host factors required to repair Mtb-damaged phagosomal membranes. Macrophage Differentiation and Metabolic Adaptation in Mycobacterial Infections.

The adaptation of macrophages-the most common tissue-resident immune cells-to metabolic and microbial cues with high local variability is essential for the maintenance of organ integrity. In homeostasis, macrophages show largely predictable tissue-specific differentiation, as recently revealed by multidimensional methods. However, chronic infections with human-adapted pathogens substantially contribute to the differentiation complexity of tissue macrophages, which has been only partially resolved. Specifically, the response to mycobacterial species-which range from Mycobacterium tuberculosis (
