OBJECTIVE: Citrobacter spp. are commonly found in the human intestine and can cause antibiotic-resistant infections. Inhibitors of LpxC, an enzyme involved in the synthesis of lipid A, have demonstrated potent activity against multiple Gram-negative species, but their activity in Citrobacter spp. has not been well-characterised. The objective of the present study was to evaluate the antimicrobial activity of LpxC inhibitors against carbapenemase-producing Citrobacter spp. clinical isolates.
METHODS: The MICs of LpxC-2, LpxC-4, and CHIR-090 LpxC inhibitors were determined for 61 clinical isola

Antimicrobial drug design is an active area of research. Drug resistance in microorganisms to conventional antibiotics imposes a huge burden on the healthcare and economy worldwide. Understanding the structural parameters controlling the antimicrobial activity enables researchers to develop effective antimicrobial agents. Post-modification of artificial peptides as a method to introduce amphiphilicity, a structural parameter to exhibit antimicrobial activity, is described in the current work. Using two hybrid sequences composed of 5-amino salicylic acid and natural amino acid leucine (Leu) or

Candida albicans is the most common human fungal pathogen and one of the major causes of drug-resistant, life-threatening fungal infections. The Neosartorya (Aspergillus) fischeri antifungal protein 2 (NFAP2), exhibits potent antifungal activity in both planktonic and biofilm-forming Candida cells in vitro and in vivo. However, the exact antifungal mechanism of NFAP2 remains unelucidated, limiting its therapeutic application. Based on our observations, we posit that NFAP2 is internalized in Candida cells and slows the growth over time, in addition to its previously characterized rapid plasma m

UNLABELLED: Heteroresistance describes the phenomenon where seemingly isogenic bacterial populations contain subpopulations with elevated resistance compared to the susceptible majority that are often missed in routine susceptibility testing. The enhanced resistance of those subpopulations can either be maintained (stable) or quickly lost (unstable/transient) after treatment, where transient cases return susceptibility results identical to those of original cultures. Recent work has implicated increased gene dosage of resistance determinants as a major cause of unstable heteroresistance in cli

Pseudomonas aeruginosa infections and biofilms are difficult to treat due to their remarkable ability of antibiotic resistance. Consequently, the incidence of drug-resistant P. aeruginosa infection cases is steadily increasing in hospitals worldwide. Herein, the frequency of drug-resistant P. aeruginosa wound infection cases among hospitalized patients is examined. In addition, suitable natural components with potent antibacterial activity against the drug-resistant P. aeruginosa strains are screened. Among 27 specimens, three P. aeruginosa strains (including carbapenem and multidrug-resistant

V. parahaemolyticus poses a remarkable public health threat, accounting for approximately 25% of global seafood-related infections in human consumers and resulting in severe infections and substantial economic losses in the aquaculture. To explore the prevalence, antibiogram, virulence and resistance genes, the multidrug resistance profiles, and the pathogenicity of V. parahaemolyticus recovered from shrimp, 200 Litopenaeus vannamei (clinically healthy: n = 100 and diseased: n = 100) were gathered from commercial shrimp farms in Ismailia, Egypt. Accordingly, clinical and postmortem findings an

The global aquaculture industry, particularly the farming of Pacific white shrimp (Litopenaeus vannamei), faces significant economic challenges due to infectious diseases, with Vibrio spp. being a primary causative agent of vibriosis. The widespread and often indiscriminate use of antibiotics in aquaculture has led to an alarming increase in antimicrobial resistance (AMR) among Vibrio strains, rendering conventional treatments increasingly ineffective. This study aimed to characterize the phenotypic antibiotic resistance profiles and the presence of associated resistance genes in Vibrio spp. i

Lactoferrin is a bioactive dairy protein with multiple beneficial effects. The use of lactoferrin-based combined strategies is not only effective in combating pathogenic bacteria but also remarkably reduces the emergence of bacterial resistance, which is of great practical significance. In this study, gallic acid and epigallocatechin gallate, tea polyphenols with excellent antimicrobial effects, were selected to bind with lactoferrin (LF) in a non-covalent interaction mode. The complex was characterized using a series of spectral experiments and molecular docking, and its antibacterial activit

Bacitracin is employed for topical treatment of staphylococcal infection, though information is lacking regarding the nature of resistance to this antibiotic in the staphylococci. Here we examined bacitracin resistance in a large collection (n = 1,470) of multidrug-resistant isolates of Staphylococcus aureus. Susceptibility testing of the entire collection revealed a broad range of bacitracin minimum inhibitory concentrations (MICs) (from 32 to >4,096 µg/mL) and allowed us to define a tentative epidemiological cut-off value (TECOFF) (apparent upper end of the wild-type distribution in terms of

This study investigated the antifungal mechanisms of geraniol against Candida albicans. Geraniol significantly inhibited growth and biofilm formation, with a minimum inhibitory concentration (MIC) of 0.5 µL/mL. At concentrations of 0.3, 0.45, and 0.6 µL/mL, geraniol substantially reduced biofilm biomass (P < 0.05). Microscopic analyses revealed apoptosis induction, membrane shrinkage, and structural damage. Mechanistically, geraniol disrupted cell membrane permeability, inhibited ergosterol synthesis, and downregulated the expression of key virulence genes (SAP5, SAP6, SAP7). These multifacete

Widespread use of fluconazole has led to the emergence of fluconazole-resistant (FR) Candida spp. causing challenges in clinical treatment. Iron, an essential nutrient, affects the levels of ergosterol (a fluconazole target) in fungal membranes. Our lab-generated FR strain (fluconazole minimum inhibitory concentration [MIC] >125 µg/mL) showed a twofold lower MIC (4.66 µg/mL) for the iron chelator deferasirox (DFX), compared to its patent strain CAI4 (DFX MIC 9.34 µg/mL), suggesting a greater sensitivity to iron chelation. A sublethal dose of DFX (2.33 µg/mL) was able to effectively synergize w

Surface antimicrobial agents provide a first line of defense against pathogens, especially for immunocompromised individuals. Insect-induced plant galls, tumor-like structures formed on plant surfaces by insect larvae, have long been used as sources of antimicrobial compounds. Building on existing knowledge, this study evaluated the surface antimicrobial activity of a standardized, ethanolic extract of Aleppo oak galls (AGE) on agar, abiotic, and biotic surfaces. Using a novel surface antimicrobial assay, we demonstrated that the anti-Escherichia coli, -Staphylococcus aureus, -Candida albicans

Vancomycin continues to be a widely used antibiotic of last resort in treating drug-resistant pathogens despite the emergence of vancomycin-resistant strains such as vancomycin-resistant Enterococci (VRE). This communication reports that conjugation of vancomycin to a second antibiotic that targets a different region of lipid II enhances and rescues its antibiotic activity. Conjugation of vancomycin to a minimal teixobactin pharmacophore in which residues 1-6 are replaced with an aromatic amide results in substantial enhancement in activity over the individual components or mixtures thereof.
