Machine learning-assisted dual-mode immunochromatographic detection of pentachlorophenol via oriented antibody-functionalized AFIrTPs nanoprobes.

Pentachlorophenol (PCP), a human carcinogen, is widely detected in human biosamples, despite being banned in aquaculture, continues to show high detection frequencies in aquatic products according to regulatory reports. Therefore, there is an urgent need for reliable on-site testing methods. Immunochromatographic assay (ICA) are promising, but conventional ICA suffers from antibody deactivation and unstable single-signal outputs in complex matrices. Herein, we reported a self-assembly method to fabricate tannic acid (TA)-mediated sea urchin-like Au/Fe/Ir trimetallic nanoparticles (AFIrTPs) tha

Aflatoxin Exposure and Human Health with a Focus on Northern Latin America.

Aflatoxins, mycotoxins produced by Aspergillus flavus and Aspergillus parasiticus, were discovered sixty-five years ago and remain a significant public health threat, particularly amid increasing instances of extreme weather events. Of the four principal forms of aflatoxins found in foods (B1, B2, G1, and G2), aflatoxin B1 is the most potent carcinogen. Aflatoxins commonly contaminate a variety of foodstuffs, with maize being among the most susceptible. Chronic exposure to aflatoxins has been linked to liver cancer, childhood stunting, gallbladder cancer, and other adverse health effects. Due

Extraction, encapsulation, and bioaccessibility of polyphenols from fruit and vegetable by-products and their health effects: a decade overview.

The fruits and vegetable processing industry produces a huge amount of by-products that are rich in bioactive compounds. Therefore, the recovery of these biologically active compounds from industry by-products has received significant attention. Polyphenols are recognized for their therapeutic potential, including anti-carcinogenic, anti-inflammatory, and antioxidant properties. However, they are highly sensitive and easily degraded, and also have lower bioaccessibility due to their instability in gastrointestinal conditions. To increase stability and improve the bioaccessibility of polyphenol

A Smartphone-Assisted Approach to Formaldehyde Detection Using Diethanolamine-Grafted Carbon Nanoparticles.

Formaldehyde (FA) is a well-known indoor pollutant and human carcinogen, making the development of field-deployable sensors for its monitoring crucial for public safety. Herein, we report a facile strategy to synthesize carbon nanoparticles functionalized with diethanolamine (CNPs-DEA) as FA sensors. The strategic surface functionalization dictates a specific spatial arrangement that is key to efficient and cooperative interaction with the analyte. In water solution, the probe exhibits a LOD of 46 ppb with a binding constant (log β) value of 4.31 ± 0.01. By immobilizing the CNPs on polyamide m

Circulating metabolomic and chemical exposomics signatures as potential biomarkers for long-term radon exposure.

Radon is a radioactive gas and a lung carcinogen, yet lacks established blood biomarkers for long-term exposure. We aimed to identify circulating metabolomic and exposomics signatures correlated with reconstructed radon-222 exposure. Metabolites and chemical exposomes were profiled using liquid and gas chromatography-high-resolution mass spectrometry from plasma (n = 80). Batch effects were corrected using ComBat, and network and pathway analyses were performed using xMWAS and MetaboAnalyst, respectively. A total of 124 metabolites were correlated with radon, including 41 positive correlations

Robust discovery of mutational signatures using power posteriors.

Mutational processes, such as the molecular effects of carcinogenic agents or defective DNA repair mechanisms, produce different mutation types with characteristic frequency profiles, known as mutational signatures. Non-negative matrix factorization (NMF) has been successfully used to discover many mutational signatures, yielding novel insights into cancer etiology and informing targeted therapies. However, the NMF model is only a rough approximation to reality, and even small departures from this assumed model can have large negative effects on the accuracy and reliability of the results. We

Targeting SYK to alleviate MDSC-driven immunosuppression and augment anti-PD1 efficacy.

Myeloid-derived suppressor cells (MDSCs) play a crucial role in tumor-associated immune suppression and in impeding immune checkpoint blockade (ICB) therapies; however, effective intervention strategies targeting MDSCs remain limited. In this study, we observed a positive correlation between spleen tyrosine kinase (SYK) activity in immune cells and tumor progression in human cancers. Utilizing a mouse model with a gain-of-function SYKS544Y mutation, we demonstrated that SYK activation promotes tumorigenesis in carcinogen-induced and xenograft models across multiple cancer types, largely throug

Metabolism-Driven Modulation by the Human Microbiota: Implications for Cancer Therapy and Emerging Strategies.

The human microbiome plays a pivotal role in cancer development, progression, and therapeutic response. Epidemiologic studies have established links between microbiome composition and various malignancies, with specific microbial taxa exerting direct carcinogenic effects or influencing tumorigenesis through metabolite production and immune modulation. While the gut microbiome remains the most extensively studied, emerging evidence highlights the significance of microbiomes in other body sites, including the cervix, lung, and skin, which also modulate cancer risk and progression. These site-spe

A pathogenic CD8⁺ TRM-chemokine axis orchestrates liver fibrosis and provides circulating biomarkers during chronic Clonorchis sinensis infection.

Chronic liver fluke infections represent a major global health burden, driving liver fibrosis and elevating cancer risk, yet the immunological mechanisms linking persistent infection to tissue damage remain poorly defined. Using Clonorchis sinensis, a carcinogenic liver fluke infecting millions worldwide, as a representative model, we map immune responses during chronic infection in mice. We show that intrahepatic CD8⁺ tissue-resident memory T cells (TRM) progressively accumulate and drive pathology by producing chemokines that promote immune recruitment and fibroblast activation. Depletion of

Developmental neurotoxic effects of organophosphate flame retardants (OPFRs) across different life stages and the central role of pioneer transcription factors Neurog2/Ascl1 during the embryonic period.

In contemporary consumer products markets, organophosphorus flame retardants (OPFRs) are widely used, associated with non-carcinogenic health risks and exhibit multisystem toxicity. Based on the endocrine-disrupting properties, which are combined with the endocrine system's susceptibility and interaction with the nervous system during early developmental stages, developmental neurotoxicity (DNT) has been increasingly reported in epidemiological studies. Unfortunately, their toxic mechanisms remain unclear. Given the highly stage specific nature of neurogenesis-related gene expression across li

Colon cancer in Appalachian Kentucky: Unique genetic, microbiome and obesity findings in a cohort comparison.

We investigated colon cancer genomics and microenvironmental features in the Appalachian Kentucky population, a group with the highest incidence of colon cancer in the United States. We assessed two inter-related risk factors for colon cancer (obesity and abnormal gut bacterial microbiome) and their genetic associations within this population. To evaluate potential unique characteristics of the high-incidence cohort, we compared 99 propensity-matched colon cancer tumors from Appalachian Kentucky patients to 95 non-Appalachian patient tumors to evaluate driver mutations, differentially expresse

Targeting of the m6A eraser ALKBH5 suppresses stemness and chemoresistance of colorectal cancer.

The role of RNA N6-methyladenoine (m6A) eraser AlkB homologue 5 (ALKBH5) in colorectal cancer (CRC) stem cells (CSCs) is unclear. Here, we find that ALKBH5 expression positively correlates with CSC markers in CRC patients. ALKBH5 induces self-renewal and stemness markers in colorectal CSCs and patient-derived organoids (PDOs). Colon-stem cell specific Alkbh5 knockin accelerates carcinogen-induced CRC, while tumorigenesis is attenuated in colon-stem cell specific Alkbh5 knockout mice. Integrated RNA-seq, MeRIP-seq and Ribo-seq reveal FAM84A as an ALKBH5 target. ALKBH5 demethylates m6A-modified

Merkel Cell Polyomavirus Co-Infection in HIV/AIDS Individuals: Clinical Diagnosis, Consequences and Treatments.

Merkel cell polyomavirus (MCV) was named for its role as the causative agent of Merkel cell carcinoma (MCC), which is MCV positive in approximately 80% of cases. MCV is classified as a Group 2A carcinogen, which promotes carcinogenesis by integrating T-antigen into the cell genome. The prevalence of anti-MCV antibodies in the general population can be as high as 90%. MCV typically promotes cancer by integrating T-antigen genes into the host cell genome, and 80% of MCC cases are attributed to MCV activation. In immunocompetent individuals, MCV usually remains latent after infection. However, th

Recycling Fiber-Reinforced Polyamide Waste from the Automotive Industry: Life Cycle Assessment (LCA) of an Advanced Pyrolysis Process to Reclaim Glass Fibers and Valuable Chemicals.

The generation of pyrolysis liquids and gases with poor quality is a limiting factor for the development of the recycling process of fiber-reinforced plastic waste. In this article, the life cycle assessment (LCA) of an advanced two-step pyrolysis process to recycle glass fiber-reinforced polyamide waste is presented. First, the solid waste is pyrolyzed by heating up at 3 °C/min to 500 °C in a tank reactor. The generated volatiles are subsequently thermally cracked at 900 °C in a tubular packed bed reactor. The process is able to reclaim the glass fibers similarly to the conventional one react
