Post-covid-19 immune signatures: Antibody and Th1 cytokine profiles among healthcare workers in Côte d'Ivoire.

The post-SARS-CoV-2 immune response involves both neutralizing antibodies (NAbs) and T-cell cooperation. Healthcare workers, highly exposed in daily practice, represent a relevant population for study. This study aimed to describe the post-COVID-19 immune profiles of Ivorian healthcare workers by analyzing the association between antibodies (IgM, IgG, NAbs) and Th1 cytokines (IL-2, IFN-γ, TNF-α). This cross-sectional study, conducted during January 2022 to June 2023 in three university hospitals in Abidjan, included 36 participants with RT-PCR-confirmed infection. IgM and IgG were measured usi

Interleukin-6 Rescues Lymphocyte from Apoptosis and Exhaustion Induced by Chronic Hepatitis C Virus Infection.

Chronic hepatitis C virus (HCV) infection appears to trigger the onset of immune exhaustion and apoptosis to potentially assist viral persistence inside the host, eventually leading to exacerbated conditions of inflammation and hepatocarcinogenesis. Growing evidence suggests that spontaneous apoptosis of peripheral blood mononuclear cells (PBMCs) could be one of the potential immune impairment mechanisms in chronic viral infection. Interleukin-6 (IL-6) is a pleiotropic cytokine that plays an essential role in regulating immune and inflammatory responses. Owing to its known role in priming T ce

Upregulation of miRNA-143, -145, -192, and -194 in esophageal epithelial cells upon acidic bile salt stimulation.

Barrett's esophagus (BE) is a metaplastic condition of the distal esophagus that occurs because of chronic gastroesophageal reflux. Previous studies have identified BE-specific microRNAs (miRNAs) in comparison with normal squamous epithelium (SQ). We hypothesized that BE-specific miRNAs could be induced in esophageal SQ cells by exposure to acid and/or bile salts. We aimed to determine whether BE-specific miRNAs are upregulated in an esophageal SQ cell line (Het-1A) in an environment with acid and/or bile salts and whether this is nuclear factor-κB (NF-κB) dependent. Acid and/or bile salt incu

Enhanced apoptosis of T cells in common variable immunodeficiency (CVID): role of defective CD28 co-stimulation.

CVID is a primary immune disorder in which hypogammaglobulinaemia may be associated with a number of T cell defects including lymphopenia, anergy, impaired lymphocyte proliferation and deficient cytokine secretion. In this study we show that T cells of CVID subjects, in comparison with control T cells, undergo spontaneous apoptosis in culture and markedly accelerated apoptosis after gamma-irradiation. Although costimulation of the CD28 receptor following engagement of the TCR/CD3 receptor normally provides a second signal necessary for IL-2 secretion, CD28 costimulation in CVID does not signif

Genetic analysis of immune dysfunction in non-obese diabetic (NOD) mice: mapping of a susceptibility locus close to the Bcl-2 gene correlates with increased resistance of NOD T cells to apoptosis induction.

The non-obese diabetic (NOD) mouse strain provides a remarkable model for investigating the mechanisms of autoimmunity. Independent genetic analyses of this model have previously shown that chromosome 1-linked loci were involved in the control of periinsulitis and sialitis on the one hand and of insulitis and diabetes on the other hand. In the present work, analysis of a [NOD x (NOD x C57BL/6)F1] backcross progeny allowed us to clearly dissociate two genetic regions: one was associated with periinsulitis and mapped to the middle region of chromosome 1, in the vicinity of the Bcl-2 gene; the ot

Effect of alpha-IFN on cytokine-induced antigen expression and secretion of TNF, LT and IgM in HCL.

In order to investigate the possible mechanisms for the effect of alpha-interferon (alpha-IFN) in hairy cell leukaemia (HCL), blood cells from 4 cases were treated in vitro with alpha-IFN, tumour necrosis factor (TNF) and interleukin 2 (IL-2). Changes in the antigen expression, immunoglobulin (Ig) secretion and the production of TNF and lymphotoxin (LT) were investigated. TNF induced expression of CD4 and CD71, increased the intensity of HLA-DR, CD25, CD11c and CD13 expression and decreased both the intensity and frequency of sIg and cIg positivity. alpha-IFN decreased CD25 expression, the tar

REV-ERBα deficiency exacerbates IL-33-mediated airway inflammation and airway hyperresponsiveness in mice.

Asthma involves type 2 airway inflammation driven by epithelial-derived interleukin-33 (IL-33), a key mediator of allergic responses. REV-ERBα, a circadian transcriptional repressor, modulates pulmonary inflammation, but its role in IL-33-induced airway pathology remains unclear. Here we investigated IL-33-mediated pulmonary inflammation in wild-type and global Rev-erbα knockout mice exposed intranasally to recombinant IL-33. Loss of REV-ERBα markedly increased recruitment of eosinophils, neutrophils, innate lymphoid cells, and Th2 lymphocytes, exacerbated airway hyperresponsiveness, and eleva

A rare case of keloid combined with bullous pemphigoid.

Keloids and bullous pemphigoid (BP) are two clinically and pathophysiologically distinct dermatologic disorders - the former is characterized by abnormal scar formation due to excessive collagen deposition, whereas the latter is an autoimmune blistering disease. We report a case of a 67-year-old man with a 19-year history of abdominal keloids and the subsequent appearance of vesicular skin lesions. Histopathologic examination revealed both keloidal fibrosis and subepidermal blister formation. Serum testing demonstrated elevated anti-BP180 antibody levels. Initial treatment with topical and sys

Immunological role of IL-25 on gut-dwelling helminths.

Interleukin-25 (IL-25) is a crucial cytokine in coordinating type 2 immunity against gut-dwelling helminths. IL-25 is produced by a variety of cells, including epithelial and immune cells, and primarily stimulates Th2-type immune responses characterized by the release of IL-13, IL-5, and IL-4, which play a vital role in eosinophilia induction, goblet cell hyperplasia, and IgE synthesis. This review critically examines IL-25's role in maintaining intestinal barrier integrity and modulating regulatory immune responses. This review addresses how IL-25 integrates signals from the epithelial interf

Terrusnolides A-D, new butenolides with anti-inflammatory activities from an endophytic Aspergillus from Tripterygium wilfordii.

Terrusnolides A-D (1-4), four butenolides were isolated from an endophytic Aspergillus from Tripterygium wilfordii. The structures of 1-4 were established by comprehensive spectroscopic analyses and electronic circular dichroism (ECD) calculation. It is interesting that 1 was a butenolide derived by a triple decarboxylation, while 2-4 were the metabolites with 4-benzyl-3-phenyl-5H-furan-2-one motif possessing an isopentene group fused to the benzene ring. In vitro anti-inflammatory effects of these isolates were evaluated in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. 1-4 exhibit

Adipotoxicity and the insulin resistance syndrome.

During the last decades, nutritional fat intake has continuously inflated in parallel with an enormous rise in the prevalence of obesity and type-2 diabetes in adults but increasingly also in adolescents and even children. Augmented fat intake is associated with an increased mass of adipose tissue which releases free fatty acids (FFA) but also hormones and cytokines such as leptin, adiponectin, resistin, tumor necrosis factor-a and interleukin-6. In particular, FFA decrease insulin-mediated glucose transport/ phosphorylation in skeletal muscle and impair suppression of glucose production by th

A case of Weber-Christian disease associated with myelodysplastic syndrome.

We report the case of a 73-year-old man with myelodysplastic syndrome (MDS) who developed Weber-Christian disease (WCD). Bone marrow aspirates showed refractory anemia with abnormal karyotypes such as trisomy 8, trisomy 8 and 14, and trisomy 8, 9, and 14. The patient had intermittent fever associated with multiple tender erythematous nodules on the skin. A biopsy sample taken from a nodule revealed focal subcutaneous infiltration of neutrophils and necrotizing fat tissue. We diagnosed the patient as having lobular panniculitis associated with myelodysplastic syndrome. The serum levels of solub

Complex association analysis of graves disease using a set of polymorphic markers.

Graves disease is complex autoimmune thyrotoxicosis. A number of genes may contribute to the development of the disorder. Some of them may be genes that encode cytotoxic T-lymphocyte-associated serine esterase-4 (CTLA4), subunit 2 of large multifunctional protease (LMP2), thyroid-stimulating hormone receptor (TSHR), and interleukin 1 receptor antagonist (IL1RN). We studied polymorphism of Ala17Thr CTLA4, H60R LMP2, Pro52Thr TSHR, and IL1RN-VNTR in healthy controls (n = 93) and patients with Graves disease (n = 78) using PCR. To study CTLA4, H60R, and TSHR polymorphism, PCR products were digest

Long-term effect of prenatal exposure to maternal microfilaraemia on immune responsiveness to filarial parasite antigens.

To identify long-term effects of prenatal exposure to maternal filarial-parasite infection, we assessed lymphocyte responses in 21 Polynesian children born 17-19 years previously to mothers diagnosed as being microfilaraemic or infection-free. All children lived on an island endemic for bancroftian filariasis but were free from infection at the time of study. While children (n = 10) of infection-free mothers responded vigorously to microfilarial antigen with lymphocyte proliferation, production of interleukin 2 (IL-2), IL-5, IL-10, granulocyte macrophage colony-stimulating factor (GM-CSF), and

Construction of an Oral Vaccine Based on Multiple Antigens of African Swine Fever Virus Displayed on the Surface of Bacillus Subtilis Spores and Study of its Immune Effect in Rats.

In this study, we constructed an oral vaccine by expressing fusion antigens of African swine fever virus (ASFV) on the surface of Bacillus subtilis spores and explored its immunogenicity. Fusion proteins were designed, and recombinant strains that could stably express target proteins on the spore surface were successfully constructed. After oral immunization of rats, the spleen index and the levels of ASFV-specific IgG, interleukin-2, and interferon-γ were markedly increased, indicating effective activation of humoral and cellular immunity. This study established a reliable spore surface displ

Chronic Hepatitis and Hemophagocytic Lymphohistiocytosis Associated With Systemic Chronic Active Epstein-Barr Virus Disease.

Systemic chronic active Epstein-Barr virus (EBV) disease is a rare, delayed, and life-threatening complication of EBV infection. It is characterized by persistent viremia and a mononucleosis-like syndrome with multisystemic involvement because of lymphocytic tissue infiltration. We present the case of an 82-year-old woman without known immunodeficiency who presented with abnormal liver enzymes and interstitial pneumonia. Investigations revealed hyperferritinemia, elevated soluble interleukin-2 receptor levels, hypofibrinogenemia, and hepatosplenomegaly, consistent with hemophagocytic lymphohis

Sputum interleukin-6 level as a marker of severity during acute chest syndrome in children with sickle cell disease.

Acute chest syndrome (ACS) is a leading cause of morbimortality in sickle cell disease (SCD). In this prospective observational study, we investigated sputum interleukin-6 (IL-6) level as an ACS severity marker during 30 ACS episodes in 26 SCD children. Sputum IL-6 levels measured within the first 72 h of hospitalisation for ACS were significantly higher in patients with oxygen requirement ≥2 L/min, ventilation (invasive and/or non-invasive) length ≥5 days, bilateral and/or extensive opacities on chest X-ray or erythrocytapheresis requirement. Sputum IL-6 could serve as an ACS severity marker

Progressive Thymic Hyperplasia With Graves' Disease: A Case Report.

We treated a 36-year-old man with thymic hyperplasia complicated with Graves' disease. Thymic hyperplasia was observed on thoracic computed tomography (CT) three months after the onset of thyrotoxicosis symptoms. One month after thiamazole initiation, he displayed drug-induced liver injury and underwent a total thyroidectomy. Seven months after surgery, we observed a dramatic reduction of thymic size associated with normalizing the soluble interleukin-2 receptor (sIL-2R) levels. The rapid development of hyperplasia after the onset of thyrotoxicosis and the restoration in thymus volume after to
