Transient Hypertension after Spinal Cord Injury Leads to Cerebrovascular Endothelial Dysfunction and Fibrosis.

We aimed to create a clinically relevant pre-clinical model of transient hypertension, and then evaluate the pathophysiological cerebrovascular processes resulting from this novel stimulus, which has recently been epidemiologically linked to cerebrovascular disease. We first developed a clinically relevant model of transient hypertension, secondary to induced autonomic dysreflexia after spinal cord injury and demonstrated that in both patients and rats, this stimulus leads to drastic acute cerebral hyperperfusion. For this, iatrogenic urodynamic filling/penile vibrostimulation was completed wh

Modelling hepatitis C virus incidence, prevalence and long-term sequelae in Australia, 2001.

BACKGROUND: To plan an appropriate public health response to the hepatitis C virus (HCV) epidemic requires that estimates of HCV incidence and prevalence, and projections of the long-term sequelae of infection, are as accurate as possible. In this paper, mathematical models are used to synthesize data on the epidemiology and natural history of HCV in Australia to estimate HCV incidence and prevalence in Australia to end 2001, and project future trends in the long-term sequelae of HCV infection. METHODS: Mathematical models of the HCV epidemic in Australia were developed based on estimates of

Effect of age and loading rate on human cervical spine injury threshold.

STUDY DESIGN: Statistical analysis of human cadaver cervical spine compression experiments. OBJECTIVES: To quantify the cervical spine compressive injury threshold as a function of the person's age, gender, and external loading rate. SUMMARY OF BACKGROUND DATA: Results of epidemiologic studies have indicated that most survivors of cervical spinal cord injury have spinal column fractures and dislocations that result from a compression or compression-flexion force vector. Cervical spinal column injury thresholds are dependent on many factors. Delineation of the injury thresholds according to a

Prevalence of dementia in selected Middle East and North Africa (MENA) countries: A systematic review and meta-analysis.

Data on dementia epidemiology in the Middle East and North Africa (MENA) region is limited. This systematic review and meta-analysis examined dementia prevalence across MENA. Databases were searched up to October 2024. Analyses were stratified by country and sex. Pooled prevalence was estimated using a random-effects model with a 95% confidence interval (CI). Fifty-two studies on the selected countries met inclusion criteria, covering 87,219 individuals with dementia from a total population of 1,045,908. The pooled prevalence was 12.16% (95% CI: 9.61-14.96) for the region and the Israel had th

Neurobiology of Posttraumatic Stress Disorder: Circuits, Genomics, and Treatment.

Posttraumatic stress disorder (PTSD) is unique in its requirement of an external stressor for the development of disease, leading to dysregulated mood, intrusive memories, and avoidance symptoms. Over the past decade, insights from human neuroimaging, mechanistic neural circuitry studies in animal models, and genomic work have revolutionized our understanding of this psychiatric illness, with emerging data from large-scale PTSD genome-wide association studies providing novel insights into the mechanism of PTSD. Despite these advances, therapeutic interventions remain limited, and psychotherapy

[Syphilis and human treponemes: a long evolutionary history revealed by paleogenomics].

Recent discoveries in paleogenomics have revolutionized our understanding of syphilis and other human treponematoses. Far from being a pathogen that suddenly appeared in Europe in the late Middle Ages, we now know that Treponema pallidum has been circulated among human populations for millennia. Ancient genomes recovered from pre-Columbian contexts in the Americas show that major treponemal lineages had already diversified well before the modern era, often in the absence of recognizable skeletal lesions. Genomic analyses further indicate that treponemal diversity is not the result of extensive

Multiple System Atrophy: Pathology, Pathogenesis, and Path Forward.

Multiple system atrophy (MSA) is a fatal neurodegenerative disease characterized by autonomic failure and motor impairment. The hallmark pathologic finding in MSA is widespread oligodendroglial cytoplasmic inclusions rich in aggregated α-synuclein (αSyn). MSA is widely held to be an oligodendroglial synucleinopathy, and we outline lines of evidence to support this assertion, including the presence of early myelin loss. However, we also consider emerging data that support the possibility of neuronal or immune dysfunction as a primary driver of MSA. These hypotheses are placed in the context of

Current and Emerging Issues in Familial Hypobetalipoproteinemia-related Steatotic Liver Diseases.

Familial hypobetalipoproteinemia (FHBL), caused by apolipoprotein B (APOB) variants, disrupts APOB-containing lipoprotein synthesis, leading to reduced serum total cholesterol, low-density lipoprotein cholesterol, and APOB. Heterozygous carriers are often asymptomatic, while homozygotes exhibit severe manifestations like malabsorption, vitamin deficiencies, and hepatic steatosis. In recent years, FHBL has attracted increasing attention due to its association with liver disease and its role as a unique monogenic model of steatotic liver disease independent of cardiometabolic risk factors. Mecha

Evaluation of substances of abuse consumption and enterovirus prevalence in Barcelona's (Spain) urban wastewater and their correlation with socioeconomic variables.

Wastewater-based epidemiology (WBE) is a powerful tool for tracking chemical and biological markers and for the analysis of population-level health trends. This study adopted a robust multi-stakeholder approach (bringing together key representatives from local administration, health centres, social organizations, water utilities, and academia) to conduct wastewater monitoring using the quadruple helix model, with the aim to enhance stakeholder engagement and improve population health assessments. The substances of abuse of greatest interest to stakeholders, such as cocaine, caffeine, and nicot

Implications of successive blood feeding on Wolbachia-mediated dengue virus inhibition in Aedes aegypti mosquitoes.

Wolbachia is a promising strategy to inhibit dengue virus (DENV) transmission by Ae. aegypti mosquitoes. Laboratory studies assessing DENV inhibition by Wolbachia typically have not considered natural frequent mosquito blood feeding behavior. Here, we determine the impact of successive feeding on DENV-2 transmission by Ae. aegypti in the presence or absence of Wolbachia (wAlbB and wMelM strains). We show that successive feeding shortens the extrinsic incubation period (EIP) in wildtype (WT; without Wolbachia) and wAlbB mosquitoes through enhanced dissemination. Feeding empirical data into mode

IL-17 and Th17 Cells in Chlamydiae Infection.

Chlamydia is a genus of intracellular parasitic microorganisms with a unique developmental cycle that is widely distributed and can cause various human and animal diseases. T helper 17 (Th17) cells are a distinct subpopulation of Th cells with a unique transcriptional profile (STAT3, RORγt and RORα), a pattern of cytokine production (IL-17 family, IL-21, IL-22) and specific cytokines required for differentiation (IL-6, TGF-β, IL-21 and IL-23). Results from mouse models of infection and human epidemiologic studies indicate that Th17 cells may be vital in modulating innate and adaptive immune re

Incidence of infectious diseases in children with Mycobacterium tuberculosis in Japan, 2007-2022.

The objective of this study was to understand the descriptive epidemiology of childhood tuberculosis (TB) in Japan under the 2013 Bacillus Calmette-Guérin (BCG) immunization program modification. The median percentage of annual vaccination coverage for infants aged <13 months was 97.0 % during follow-up (2007-2022). The age at which most infants received their vaccinations was 3 months before 2013 and 5 months after 2013. During follow-up, the number of childhood TB notifications and annual incidence declined by approximately 60 % and 40 %, respectively. A multivariate-adjusted model analysis

Johannes Theodor Wilhelm Petruschky (1863-1945) and the population-level tuberculin experiment for tuberculosis eradication in Hel.

This article examines the experiment conducted by Johannes Theodor Wilhelm Petruschky (1863-1945) on the eradication of tuberculosis using tuberculin. His research was inspired by the work of Robert Koch, who discovered the etiological agent of tuberculosis and conducted experiments using tuberculin as a therapeutic agent. Despite initial hopes, tuberculin proved ineffective in treating tuberculosis. Following Koch's ideas, Petruschky conducted studies in Gdańsk and later in the isolated community of Hel, where he attempted to eliminate tuberculosis through systematic diagnosis and prevention.

Causal effects of endometriosis on cancer risk: A Mendelian randomization study.

Endometriosis has been reported in epidemiological studies to be associated with certain types of cancer. However, the presence of reverse causality and residual confounding due to common risk factors introduces uncertainty regarding the extent to which endometriosis itself contributes to the development of cancer. We performed the Mendelian randomization (MR) to investigate the causal associations between endometriosis and 34 different types of cancers. The results of the inverse-variance-weighted (IVW) model suggested that genetic predisposition to endometriosis was causally associated with

Candida glabrata: A Tale of Stealth and Endurance.

Candida (Nakaseomyces) glabrata, an opportunistic human fungal pathogen, causes mucosal and deep-seated infections in immunocompromised individuals. Recently designated as a high-priority fungal pathogen by the World Health Organization (WHO), C. glabrata exhibits low inherent susceptibility to azole antifungals. In addition, about 10% clinical isolates of C. glabrata display co-resistance to both azole and echinocandin drugs. Molecular mechanisms of antifungal resistance and virulence in C. glabrata are currently being delineated in-depth. This Review provides an overview of the epidemiology,

Epidemiological profiles and pathogenicity of Vancomycin-resistant Enterococcus faecium clinical isolates in Taiwan.

The emerging Vancomycin-resistant Enterococcus faecium (VRE-fm) is an opportunistic pathogen causing nosocomial infections. The identification of VRE-fm is important for successful prevention and control in healthcare settings. VRE-fm clinical isolates obtained from regional hospitals in northern Taiwan were characterized for antimicrobial susceptibility, virulence genes and biofilm production. Most isolates exhibited multi-drug resistance and carried the virulence genes, esp and hyl. While all isolates produce biofilms, those isolates that carried esp exhibited greater biofilm production.
