Statistical methods for geographical surveillance in veterinary epidemiology.

Spatial clustering and cluster detection are statistical analysis developed to address relevant scientific hypothesis. The difficulty stays in the large number of alternative hypothesis due to the different mechanisms that could generate the anomalous cases aggregation. We review methods for marked point data (case/control) aimed to describe spatial intensity of disease risk, to test for randomness and to locate significant excesses. Bayesian Gaussian Spatial Exponential models are used to illustrate probabilistic aspects and the link with simpler non parametric tools are shown. We develop an

Uncertainty distribution associated with estimating a proportion in microbial risk assessment.

The uncertainty associated with estimates should be taken into account in quantitative risk assessment. Each input's uncertainty can be characterized through a probabilistic distribution for use under Monte Carlo simulations. In this study, the sampling uncertainty associated with estimating a low proportion on the basis of a small sample size was considered. A common application in microbial risk assessment is the estimation of a prevalence, proportion of contaminated food products, on the basis of few tested units. Three Bayesian approaches (based on beta(0, 0), beta(1/2, 1/2), and beta(l, 1

[The Bayesian statistical theory in the diagnosis of malignant and non-malignant diseases of the lung, pleura and mediastinum].

The Bayesian algorithm was used to assess the probable diagnosis in 1262 patients with a recently diagnosed finding on X-rays of the chest and the results were compared with the final diagnosis. The patients were with regard to the X-ray picture divided into 9 groups: hilar, solitary, multiple, segmental, non-segmental, cavity, diffuse, pleural and mediastinal lesions. Using the Bayesian algorithm, commonly accessible factors were processed: age, sex, case-history, cigarette smoking, red cell sedimentation rate, number of leucocytes and diameter of solitary parenchymatous lesions and the impac

A Bayesian phase I/II trial design to optimizing dose-schedule regimen with competing risk outcomes.

Identifying the optimal dose-schedule regimen in early-phase oncology trials is complicated by competing risks, such as disease progression (DP) and dose-limiting toxicity (DLT). Many existing dose-finding methods fail to adequately address these events or accommodate varying administration schedules. We propose CR-EffTox, a Bayesian adaptive phase I/II trial design that jointly models time-to-event DLT and DP using cause-specific hazard functions. Besides, the model proposed allows for dynamic information borrowing to account for associations among dose-schedule regimes. To guide regimen sele

Comparative efficacy of different therapeutic approaches in treatment naïve FLT3-mutated AML eligible for intensive chemotherapy: a Bayesian network meta-analysis of randomized trials.

FLT3-mutated acute myeloid leukemia (FLT3mut AML) is associated with poor outcomes. Although FLT3 inhibitors (FLT3is) combined with chemotherapy improve responses, long-term survival remains limited, and the optimal first-line strategy is unclear. We conducted a Bayesian network meta-analysis of eight randomized trials, including 1,793 patients, to compare treatment used in patietne eligible for intensive chemoteherapy for overall survival (OS). Treatments studied were 3 + 7 with midostaurin, quizartinib, sorafenib, gemtuzumab ozogamicin (GO), glasdegib, CPX-351, and decitabine. FLT3i-based re

An evaluation of molecular and statistical methods for the attribution of body fluid sources to donors.

Attributing mixed biological materials in crime stains to individual donors remains a challenge in forensic science. Characterisation of the messenger RNA (mRNA) transcripts in a biological material can be used for body fluid identification (BFID) purposes. Comparing coding region single nucleotide polymorphisms (cSNPs) in mRNA transcripts obtained from crime stains to reference cSNP profiles from known individuals presents a way to assign biological materials to donors. In this study, saliva, blood, skin, and vaginal samples were collected from eleven participants. Based on previous studies,

Metabolomic profiling and machine learning application for discriminating fatal hypothermia in a rat model.

Cause of death discrimination of fatal hypothermia has been a challenge in forensic practice. In the present study, we profiled the non-targeted metabolomic of iliopsoas muscle samples from rats of the fatal hypothermia group (n = 48), hypoxia group (n = 20), control group(n = 22) by ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS). A total of 63 differential metabolites were identified between the hypothermia group and the hypoxia group, while 60 differential metabolites were identified between the hypothermia group and the control group. Six different machine lea

A Systems Pharmacology Model of Aging Identifies Optimal Combination Therapies With Secondary Benefits on Weight Loss and Metabolic Health.

Aging is a systems-level process linking metabolic dysfunction, inflammation, impaired repair, frailty, and multimorbidity, whereas existing pharmacological strategies usually optimize disease-specific endpoints such as weight loss or HbA1c rather than aging-related trajectories. We developed an SBML-compliant quantitative systems pharmacology (QSP) model in which aging is represented as a dynamic, pharmacologically modifiable endpoint. The model integrates four coupled layers: metabolic/pharmacodynamic responses to GLP-1 receptor agonism, SGLT2 inhibition, metformin and rapamycin; adverse-eve

A Bayesian Meta-Analysis of Renin-Angiotensin System Inhibitors Following TAVR.

BACKGROUND: Aortic stenosis leads to left ventricular remodeling, hypertrophy, and fibrosis, increasing the risk of heart failure. Renin-angiotensin system (RAS) inhibitors may mitigate these adverse effects and improve clinical outcomes. Despite advancements in transcatheter aortic valve replacement (TAVR), substantial mortality, heart failure readmissions, and other complications persist. AIMS: This study aimed to evaluate the impact of RAS inhibitor therapy following TAVR on clinical outcomes. METHODS: We conducted a systematic review and Bayesian meta-analysis following the Cochrane Hand

SHAP based feature selection for machine learning prediction of mycoplasma pneumoniae pneumonia with atelectasis in children.

BACKGROUND: Mycoplasma pneumoniae pneumonia (MPP) with atelectasis in children is the most reported complication of MPP in children. It is associated with serious respiratory issues, pulmonary tissue damage and fibrosis, and systemic complications. Present diagnostic methods, which are mainly based on radiographic imaging, have drawbacks related to delayed case detection and radiation exposure. There is a need for a simple, non-invasive approach in the diagnosis. We aimed to develop machine learning (ML) models to identify MPP with atelectasis in children and support early risk stratification.

Latent classes of substance use patterns and cannabis use disorder Risk in U.S. Adults during recreational Legalization expansion (2015-2023).

BACKGROUND: Traditional surveillance may not capture heterogeneous patterns of cannabis use disorder (CUD) risk during policy transitions. We used latent class analysis (LCA) to identify substance use patterns associated with varying CUD risk during U.S. recreational cannabis legalization expansion (2015-2023). METHODS: We analyzed 326,193 U.S. adults aged 18 + from the National Survey on Drug Use and Health (2015-2023, excluding 2021). LCA identified classes based on eight indicators: past-year alcohol and cannabis use disorders, lifetime use of alcohol, cannabis, cocaine, and heroin, and ar

Single-cell expression quantitative trait loci-based Mendelian randomization identifies MAN1A2 in naive CD4+ T cells as a candidate therapeutic target for osteoarthritis.

BACKGROUND: Osteoarthritis (OA) is a leading cause of chronic pain and disability worldwide, with growing evidence implicating immune dysregulation in its pathogenesis. However, the specific peripheral immune cell populations contributing causally to OA risk remain poorly characterized. METHODS: We performed a multi-stage two-sample Mendelian randomization (MR) study integrating single-cell cis-expression quantitative trait loci (cis-eQTLs) for 14 peripheral immune cell types from the OneK1K cohort with two independent OA GWAS datasets - the Million Veteran Program for discovery and the UK Bi

Genomic and evolutionary insights into a novel Neisseria meningitidis genogroup W clonal complex 11 variant with dual resistance to penicillin and azithromycin in Spain.

Neisseria meningitidis clonal complex 11 (cc11) is a hyperinvasive lineage associated with global dissemination and outbreaks, including the 'South American/UK' sublineage. In Spain, cc11 serogroup W cases have increased in recent years; however, the evolutionary dynamics of circulating variants remain insufficiently characterized. This study aimed to assess the clonal diversity, temporal evolution and antimicrobial resistance profiles of cc11 isolates recovered in Spain between 2011 and 2025. A total of 1,226 isolates (406 invasive and 820 non-invasive) were collected across 8 Spanish regions

A real-world pharmacovigilance analysis of hepatitis B vaccine using the U.S. Vaccine Adverse Event Reporting System (VAERS) database.

Hepatitis B vaccines (HBVs) are widely used duo to their high clinical use and mild effects. However, as post-marketing data accumulate, several serious adverse events (SAEs) following HBV have been reported. Currently, quantitative studies based on real-world data are lacking, and information on their adverse events is limited. Adverse reaction signals of the HBV were mined and analyzed using the U.S. Vaccine Adverse Event Reporting System (VAERS) to provide a reference for the safe clinical use of this vaccine. Multiple statistical methods, including the reporting odds ratio (ROR) method, th

A heterogeneity analysis of health-related quality of life in early adults born very preterm or very low birthweight across the sociodemographic spectrum.

Preterm birth and very low birthweight (VP/VLBW) are associated with poorer health-related quality of life (HRQoL) outcomes extending into adulthood, yet it remains unclear how these effects differ across sociodemographic subgroups. This study aimed to identify heterogeneity in the association of VP/VLBW on HRQoL in early adulthood, specifically examining maternal age, education, and ethnicity. Individual-level data from three longitudinal cohorts within the Research on European Children and Adults Born Preterm Consortium were analysed, including adults born VP (< 32 weeks' gestation) or VLBW

A Spatiotemporal Analysis of Hemodialysis Incidence Rate and Impact of the COVID-19 Pandemic on the End-Stage Kidney Disease Trend.

INTRODUCTION: Limited research has been conducted on the spatial trend of end-stage kidney disease (ESKD), particularly hemodialysis (HD), in developing countries. This is problematic considering the growing demand for dialysis services in these regions. To bridge this research gap, this study evaluates the spatial trend of chronic dialysis incidence rates over a significant period (2007 to early 2021) in a large population, while also investigating the influence of the COVID-19 pandemic on this trend. METHODS: We used data from the Shiraz University of Medical Sciences Specific Diseases Affa

Masking in Active Comparator Designs in Pharmacovigilance: A Retrospective Bias Analysis on the Spontaneous Reporting of Thiazolidinediones and Cardiovascular Events.

INTRODUCTION: Masking is a reporting bias where drug safety signals are muffled by elevated reporting of other medications in spontaneous reporting databases. While the impact of masking is often limited, its effect when using restricted designs, such as active comparators, can be consequential. METHODS: We used data from the US Food and Drugs Administration Adverse Event Reporting System (1999Q3-2013Q3) to study masking in a real-world example. Rosiglitazone, a thiazolidinedione with elevated reporting after safety concerns over cardiovascular risks, was the masking candidate. We hypothesize
