Development of experimental meningeal gliomatosis models in rats.

Experimental models of meningeal gliomatosis (MG) have been produced by intracisternal inoculation of rat C6 and 9L glioma cells into Wistar and Fischer 344 rats, respectively. Tumor growth was steady and rapid in both MG models when more than 10(6) tumor cells were implanted. The median survival time of the rats inoculated with tumor cells was inversely related to the number of the cells inoculated. The clinicopathological features observed in both MG models were similar to those seen in diffuse leptomeningeal involvement of gliomas in humans. The models may be useful for investigating the pa

Efficacy of miconazole in experimental keratomycosis.

For the first time comparative efficacy of 1% miconazole in ointment base and drops has been evaluated against Candida, Aspergillus and Fusarium organisms in experimental keratomycosis. The findings of the present study reveal that miconazole in ointment drug delivery system has the advantage of being more effective in healing the fungal ulcers and associated lesions. It requires much less frequent application than drops, a parameter of great importance in improving patient compliance. Miconazole ointment was well tolerated by the rabbit eye and produced no ocular or systemic toxicity. The pre

Experimental acute alcoholic myopathy--a histochemical study.

The histochemical features of muscle injury and repair were examined in an experimental form of acute alcoholic myopathy in the rat. Typical features of rhabdomyolysis were found, which were indicated by scattered muscle fiber necrosis followed by sequential phases of fiber degeneration, phagocytosis, and regeneration. Nests of small, regenerated, or split fibers remained 2 months after the episode of acute myopathy. Vulnerability of type 1 fibers to alcohol-induced muscle injury was evident by selective involvement of muscles with type 1 fiber predominance and by the fact that regenerating fi

Minireview. The hypothalamus and blood glucose regulation.

Complex neural circuits exist in the hypothalamus for the control of metabolic hormones, enzyme activities, and substrate flux that appear to be involved with the normal adaptation to feeding and subsequent blood sugar regulation. Abnormalities within this system have been found in experimental obesity and in human diabetes, and such maladaptive changes are thought to contribute to the pathophysiology of both disorders. The present report is an attempt to present a coherent picture of the manifold factors which may be involved in the homeostatic regulation of blood glucose levels by the hypoth

Central neurogenic hyperventilation in an awake patient with brainstem astrocytoma.

A patient had the rare combination of central neurogenic hyperventilation (PaCO2 of 9 torr) and a normal level of consciousness for eight days. Morphine attenuated but never corrected the hyperventilation. Experimental effects of hypocapnia, which decreases both cerebral blood flow and metabolism in humans, are at odds with the normal mentation initially seen in this patient despite her marked and persistent hypocapnia. Death occurred after progressive brainstem dysfunction. Pathological study showed a well-differentiated astrocytoma involving primarily the medulla and pons, with scattered tum

Hepatotoxicity of hycanthone in patients with metastatic breast cancer.

Hycanthone is an antischistosomal drug with promising antitumor activity against experimental animal tumors. In phase I clinical trials, hepatitis weas the dose-limiting toxic effect and a dose of 60-70 mg/m2/day for 5 days was reported as the maximum-tolerated dose. In a phase II study of hycanthone in patients with breast cancer we have recently observed severe hepatotoxicity, even at lower doses, which resulted in two drug-related deaths. From this study and the previously available data, we conclude that this drug is too toxic for human trials at the currently recommended doses. Its radios

The effects of vasoconstriction on experimental coronary artery stenosis.

In summary, we have examined the response to arterial vasoconstriction in an in vitro coronary artery preparation. Without a preexisting stenosis, arterial vasoconstriction had minimal hemodynamic effects. Similarly, with a stenosis created by a circumferential snare, arterial vasoconstriction had minimal hemodynamic effects. In striking contrast, with a stenosis created by intraluminal obstruction, arterial vasoconstriction dramatically increased the hemodyamic severity of the stenosis. The use of an intraluminal obstruction provides a useful animal model for examining hemodynamics in coronar

Induction of chronic progressive glomerulonephritis with immunofluorescent "mesangial pattern" in rats.

Morphologic changes of extracapillary and chronic progressive glomerulonephritis with contracted kidneys, resembling those of adult human chronic glomerulonephritis, were induced in rats by a single injection of the purified glycopeptide and Freund's incomplete adjuvant into the hind footpads. The purified glycopeptide was prepared by TCA treatment or DEAE-cellulose chromatography from the nephritogenic glycopeptide which is known to have the ability to induce typical morphologic changes of membranous glomerulonephritis. As demonstrated in this report, the purification procedures result in com

Involvement of c-Myc/WWP1/TRIM65 Axis in Renal Fibrosis.

Maladaptive tubular repair is a major contributor to fibrosis and chronic kidney disease (CKD), yet the molecular regulators of this process remain poorly understood. We report that the E3 ubiquitin ligases WWP1 and TRIM65 are novel regulators of tubular fibrosis. Both ligases were markedly induced in human and experimental CKD. WWP1 induction correlates with declining renal function in humans, highlighting the potential clinical relevance of WWP1. Profibrotic factor PAI-1 promotes a robust induction of WWP1 and TRIM65 in both primary human renal epithelial cells as well as cell line (HK-2). T

A DPHV-liver module recapitulates AML infiltration and chemotherapy-induced hepatotoxicity with translational utility.

The liver is central to drug metabolism and a critical site of disease progression, making vascularized in vitro liver models essential for disease modeling, drug discovery, and mechanistic research. However, recapitulating dynamic in vivo drug responses while maintaining clinical relevance remains a major challenge for bioartificial liver systems. In acute myeloid leukemia (AML), patients frequently develop leukemic liver infiltration and chemotherapy-induced hepatotoxicity, both of which compromise therapeutic efficacy and prognosis. Human-relevant experimental models that dynamically simula

Can Micro- and Nanoplastics Modify Food-Allergy-Relevant Pathways?-A Comprehensive Narrative Review.

Micro- and nanoplastics (MNPs) are widely detected in food, drinking water, food-contact materials, and human biological samples. This narrative review evaluates whether experimental evidence supports a role for MNPs as modifiers of food-allergen digestion, intestinal-barrier function, microbiota composition, and immune tolerance. In vitro studies indicate that protein-corona formation can alter allergen conformation, epitope accessibility, and proteolysis, although effects vary by polymer, particle size, dose, and digestive model. Rodent studies provide evidence that MNP exposure can disrupt

Mouse Models of Atherosclerosis: What They Teach Us, What They Miss, and When to Use Them.

Mouse models have been indispensable for defining the mechanisms of atherosclerosis, yet no single model reproduces the full spectrum of human disease. This review provides a practical framework for selecting mouse models according to the biological question being addressed. We compare classical lipoprotein-driven models, including Apoe-/- and Ldlr-/- mice, with models designed to study human-like lipoprotein metabolism, immune and immunometabolic mechanisms, adult-onset hypercholesterolemia, plaque complexity, coronary disease, and myocardial infarction-like outcomes. Particular emphasis is p

Simultaneous quantification of D-penicillamine, D-penicillamine disulfide, and L-cysteine-D-penicillamine disulfide in human plasma: optimization of sample preparation and mass spectrometry procedures to support bioequivalence studies.

AIM: D-penicillamine (PSH) is an active pharmaceutical ingredient used for the treatment of various diseases, such as Wilson's disease, rheumatoid arthritis, cystinuria, and heavy metal poisoning. However, evaluating PSH in plasma poses substantial analytical challenges due to drug instability, thiol-disulfide exchange reactions with endogenous thiols, and the potential formation of multiple chemical forms of PSH. This study describes the development and validation of an analytical method to ensure the stability of PSH during analysis and enable accurate bioavailability measurements and bioequ

From pathology to network failure: a Question-Model-Outcome framework for interpreting animal models of AD-related and dementia-relevant mechanisms.

Animal studies remain indispensable for research on Alzheimer's disease (AD), vascular cognitive impairment, and other dementia-relevant mechanisms. Their value, however, is not that they reproduce human late-life dementia in miniature. A mouse line, vascular manipulation, or mixed-pathology preparation can make one aspect of the problem experimentally visible while not capturing the long preclinical course, clinical heterogeneity, and everyday functional consequences that define the human syndrome. This review therefore treats animal models as instruments for bounded inference. Drawing on tra

Parsing Neurometabolic Signatures of Multiple Sclerosis with MRSI and cPCA.

Magnetic Resonance Spectroscopy Imaging (MRSI) offers spatially-resolved, neurometabolic information, acquired non-invasively at whole-brain scales from human subjects. Analysis of MRSI however, is extremely challenging. The metabolic information is highly convolved, and sparsely distributed across millions of spatial-spectral datapoints, allowing for little direct human interpretation. Conversely, the overall low signal-to-noise with high-intensity artifacts can confound unsupervised machine learning approaches. These technical barriers have left much of the potential of MRSI unrealized. We a

Stochastic analysis of human ovarian aging and menopause timing.

Menopause marks a critically important biological event that ends a woman's fertility. It is a result of ovarian aging and depletion of ovarian reserve. Although many aspects of these processes are now well understood, the overall dynamic picture remains unclear. Here, we present a novel theoretical framework to analyze human ovarian aging dynamics and menopause timing. Our method is based on stochastic analysis of underlying processes stimulated by observing follicles sequentially transitioning between different stages during ovulation. This allows us to obtain a fully quantitative descriptio

A non-apoptotic caspase-8-meteorin pathway in hepatocytes promotes MASH fibrosis.

Metabolic-dysfunction-associated steatohepatitis (MASH) is the leading cause of chronic liver disease, but an incomplete understanding of MASH-induced liver fibrosis has limited therapeutic options. Here we show that hepatocyte caspase-8 drives MASH fibrosis through an apoptosis-independent mechanism. Hepatic caspase-8 expression correlates with liver fibrosis in both human and experimental MASH, and hepatocyte-specific caspase-8 deletion in male mice with MASH suppressed liver fibrosis and hepatic stellate cell (HSC) activation without affecting hepatocyte apoptosis. Mechanistic studies showe

Nonpharmacological and pharmacological influences on opioid effects: A review of clinical research findings.

The current opioid crisis underscores the need for continued research on variables that affect opioid self-administration and strategies to alter this behavior. Decades of human behavioral pharmacology studies suggest that a range of nonpharmacological and pharmacological factors influence the effects of opioids. This review focuses on consolidating findings on the influence of experimental conditions on opioid effects, with a particular emphasis on subjective effects and reinforcement. We describe different schedules of reinforcement that have been used in studies of opioid self-administratio

Treadmill Running Model for Rodents as a Translational Approach Based on Human Physiological Responses.

The concept of "Exercise is Medicine" highlights the preventive effects of physical activity on lifestyle-related diseases, dementia, and mental disorders. However, human studies face limitations in isolating exercise-specific effects due to uncontrolled variables such as diet and living conditions, as well as the constraint of non-invasive methodologies. Animal models offer a valuable alternative, allowing for strict control of experimental conditions and detailed assessment of physiological and neural responses to exercise. While voluntary wheel running has been commonly used, it lacks stand

Cerebro-spinal somatotopic organization uncovered through functional connectivity mapping.

Somatotopy, the topographical arrangement of sensorimotor pathways corresponding to distinct body parts, is a fundamental feature of the human central nervous system (CNS). Traditionally, investigations into brain and spinal cord somatotopy have been conducted independently, primarily utilizing body stimulations or movements. To date, however, no study has probed the somatotopic arrangement of cerebro-spinal functional connectionsin vivoin humans. In this study, we used simultaneous brain and cervical spinal cord functional magnetic resonance imaging (fMRI) to demonstrate how the coordinated a
