Interferon-beta expressed by a rabies virus-based HIV-1 vaccine vector serves as a molecular adjuvant and decreases pathogenicity.

Type I interferon is important in anti-viral responses and in coordinating the innate immune response. Here we explore the use of interferon-beta to adjuvant the response to a rabies virus (RV) vaccine vector expressing both HIV-1 Gag and IFN-beta. Viral load and immune responses of immunized mice were analyzed over time. Our results indicate that the RV expressing IFN-beta (IFN+) is highly attenuated when compared to control RV and demonstrate that the expression of IFN-beta reduces viral replication approximately 100-fold. Despite the decrease in replication, those mice immunized with the IF

Phenotypic and genotypic relationships between ethanol tolerance and sensitivity in mice selectively bred for initial sensitivity to ethanol (SS and LS) or development of acute tolerance (HAFT and LAFT).

BACKGROUND: Genetically based risk for development of alcoholism in humans seems to be related to initial sensitivity and/or acute tolerance to ethanol. The genetic basis for the development of tolerance has received less attention than other ethanol-related behaviors. We have selected lines of mice, according to genetics, which are differentially sensitive to the initial hypnotic effect of ethanol (Short Sleep and Long Sleep, SS and LS) and other lines that differentially develop acute functional tolerance to ethanol (High and Low Acute Functional Tolerance, HAFT and LAFT). We review reports

Recombinant Antigen for Newcastle Disease Virus Vaccine Focusing on Currently Circulating Virus Variants.

Newcastle disease virus (NDV) causes a highly contagious avian infection and is characterised by its high genetic diversity and mutation rate. Currently applied vaccines are based on NDV genotypes I and II, which are considerably distinct from the circulating pathogenic genotypes, of which genotype VII is predominant, and therefore, they may not provide complete protection for birds or stop virus transmission. This work is a proof-of-concept study aiming to develop a genotype VII-focused recombinant antigen for a vaccine targeting relevant NDV strains. The research primarily focuses on novel a

Xylt2 Knockout Eliminates O-Xylosylation and Enhances Bone Morphogenetic Protein 2 Production in Chinese Hamster Ovary Cells.

O-xylosylation of glycine-serine (GS) linkers in multispecific antibodies or fusion proteins introduces product heterogeneity, posing critical challenges for biomanufacturing quality control and elevating potential immunogenicity risks. This post-translational modification is primarily catalyzed by xylosyltransferase 2 (Xylt2) in chinese hamster ovary (CHO) cells. To address this, we generated Xylt2-deficient CHO cells via both clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) and zinc finger nuclease (ZFN)-mediated gene knockout, which resul

Lysin Motif Effectors Facilitate the Endophytic Colonization of Wheat by Sclerotinia sclerotiorum via Modulating Plant Immunity.

Sclerotinia sclerotiorum is a necrotrophic fungus that causes severe plant diseases but can also colonize wheat as a symptomless endophyte. However, the mechanism by which its effector proteins regulate this dual lifestyle remains unclear. This study aimed to identify key LysM effectors involved in S. sclerotiorum-plant interactions and elucidate their specific functions. Among seven SsLysM genes, SsLysM6 and SsLysM7 were transcriptionally upregulated during both the pathogenic infection of rapeseed and the endophytic colonization of wheat. The ΔSsLysM6 and ΔSsLysM7 mutants exhibited significa

A domestication-selected CIPK-MYB-HAK module improves salt tolerance in modern maize.

Domestication has profoundly reshaped the maize genome, yet its impact on abiotic stress resilience remains largely unresolved. Here we show that modern maize exhibits markedly greater salt tolerance than its wild progenitor, teosinte. Using a maize-teosinte (W22-T8759) recombinant inbred line population, we identify the transcriptional repressor MYB28 as a major genetic determinant underlying this divergence. MYB28 negatively regulates salt tolerance by impairing shoot Na+ exclusion. In modern maize (W22), however, salt stress activates CIPK20, which phosphorylates MYB28 to relieve its repres

Vaccination against tumour endothelial marker Robo4 inhibits tumour growth.

INTRODUCTION: Targeting tumour antigens is a major challenge in cancer-immunotherapy. We use active vaccination to induce antibodies targeting self-antigen Robo4, which is selectively expressed on tumour vascular endothelium, and supports vascular development. Our previous work showed that a conjugate of Robo4 with a foreign carrier protein induced autoantibodies specific to Robo4, which inhibited angiogenesis and tumour growth. METHODS: To translate the vaccine protocol to exploit a carrier protein used in routine human vaccination schedules, the well-characterized, non-toxic fragment C of t

A reverse genetics-based NS1-truncated live attenuated vaccine confers broad heterologous protection against swine influenza viruses.

Swine influenza A viruses (swIAVs) undergo frequent reassortment and evolution, posing significant threats to both animal and human health. In this study, we employed reverse genetics to generate five recombinant viruses and evaluated their potential as vaccine candidates. Through pathogenicity evaluation in mice, we identified two promising candidates: an inactivated vaccine (rPR8-JGL16+2) and a live-attenuated vaccine (rPR8-JGL15+3). Both vaccines induced robust hemagglutination inhibition (HI) antibody responses and specific IgG levels against homologous strains. Notably, the attenuated rPR

Evaluation of an E2-based indirect ELISA for the serological differentiation of ovine Italy pestivirus from classical swine fever virus in pigs.

The genus Pestivirus includes several species responsible for economically important diseases in domestic and wild animals. In 2017, a novel ovine pestivirus (ovIT PeV) was isolated from aborted lamb fetuses and molecular analysis showed a genetic distinction from known Pestivirus species with the closest phylogenetic relationship to classical swine fever virus (CSFV, Pestivirus suis). This close similarity is concerning because CSF is a notifiable disease to the World Organization for Animal Health (WOAH) and is subject to eradication campaigns. Moreover, ovIT PeV can infect pigs and induce a

The toxin from the PemIK-Sa1 toxin-antitoxin system decreases Staphylococcus aureus resistance to antibiotics.

Toxin-antitoxin (TA) systems are bacteria-specific components involved in maintaining mobile genetic elements that may carry antibiotic resistance determinants (ARDs), among other functions. PemIK-Sa1 is a plasmid-encoded TA system found in Staphylococcus aureus, a dangerous pathogen affecting humans and animals. The PemK-Sa1 toxin is a sequence-specific ribonuclease inhibited by its cognate antitoxin, PemI-Sa1. Using recombinant multidrug-resistant strains, we demonstrated that expression of the toxin reduces resistance to antibiotics from various classes, including β-lactams, fluoroquinolone

Germline variants analysis of Chinese breast cancer patients reveals numerous alterations in homologous recombination genes.

PURPOSE: We aimed to identify the pathogenic variants of homologous recombination (HR) genes and analyze the correlation between the pathogenic variants and clinical characteristics in Chinese breast cancer patients. METHODS: A cohort of 178 breast cancer patients participated in this study. We assessed genomic alterations using a 23-gene panel, which includes most of the HR-related genes and DNA mismatch repair (MMR) gene, through next-generation sequencing. The pathogenicity of variants was determined based on the American College of Medical Genetics and Genomics standards and guidelines. T

Recent Advances in Therapeutics and Manufacturing Processes of Recombinant Adeno-Associated Virus for the Treatment of Lung Diseases.

Developing delivery vectors capable of transducing genetic material across the lung epithelia and mucus barrier is a major challenge and of great interest to enable gene therapies to treat pulmonary diseases. Recombinant Adeno-associated Viruses (rAAVs) have emerged as attractive candidates among viral and non-viral vectors due to their broad tissue tropism, ability to transduce dividing and quiescent cells, and their safety profile in current human applications. While rAAVs have demonstrated safety in earlier clinical trials for lung disease applications, there are still some limitations rega

Clinically approved immunotoxins targeting hematological cancers: "the best of both worlds".

Hematological malignancies contribute significantly to the overall cancer burden. Certain subtypes, such as hairy cell leukemia (HCL), are chronic and characterized by residual disease after first-line therapy, while others, such as blastic plasmacytoid dendritic cell neoplasm (BPDCN), are aggressive and associated with poor prognosis. Although cornerstone interventions such as radiation and chemotherapy are efficiently used to treat some malignant blood neoplasms, these treatments are often limited by resistance, relapse, lack of enduring disease-free survival/complete remission, and systemic

mRNA based in vivo MG1113 antibody production as a potential therapeutic approach for hemophilia.

Hemophilia is a genetic bleeding disorder caused by deficiencies in coagulation factor VIII or IX. Although current therapies-including factor replacement, bispecific antibodies, and gene therapy-have improved disease management, they are limited by short half-lives, immunogenicity, complex manufacturing processes, and the need for repeated administration. Here, we report a lipid nanoparticle (LNP)- encapsulated messenger RNA (mRNA) encoding MG1113, a humanized monoclonal antibody that targets tissue factor pathway inhibitor (TFPI), enabling sustained in vivo antibody production as a factor-un

Influenza Neuraminidase Virus-Like Particle-Based Nanocarriers as a New Platform for the Delivery of Small-Peptide Antigens.

A new and simple platform to produce a nanocarrier for small-peptide antigen delivery was developed. Virus-like particles (VLPs) were of interest due to their good cell-penetrating properties and ability to protect target molecules from degradation. In this study, the VLP that was entirely formed by influenza neuraminidase (NA), NA-VLPs, was employed. The platform construction includes the genetic engineering of target peptides into the NA structure immediately above its stalk, at the bottom of the NA head, by an overlap extension PCR. The resulting chimeric gene is next expressed in stably tr

The role of recombinant LH in ovarian stimulation: what's new?

It is widely recognized that luteinizing hormone (LH) activity is pivotal during folliculogenesis. Nonetheless, the use of LH during ovarian stimulation remains a matter of debate. Indeed, women with good LH function are able to sustain follicle growth and maturation during ovarian stimulation carried out with regimens based on follicle-stimulating hormone (FSH) alone. However, evidence exists that LH activity could be necessary in specific infertile subgroups undergoing assisted reproduction treatment (ART) who are characterized by a functional or constitutive LH deficiency. For instance, wom

The ubiquitin E3 ligase BFAR promotes degradation of PNPLA3.

A missense variant in patatin-like phospholipase domain-containing protein 3 [PNPLA3(I148M)] is the most impactful genetic risk factor for fatty liver disease (FLD). We previously showed that PNPLA3 is ubiquitylated and subsequently degraded by proteasomes and autophagosomes and that the PNPLA3(148M) variant interferes with this process. To define the machinery responsible for PNPLA3 turnover, we used small interfering (si)RNAs to inactivate components of the ubiquitin proteasome system. Inactivation of bifunctional apoptosis regulator (BFAR), a membrane-bound E3 ubiquitin ligase, reproducibly

Medical semiology of patients with monogenic obesity: A systematic review.

Patients with monogenic obesity display numerous medical features on top of hyperphagic obesity, but no study to date has provided an exhaustive description of their semiology. Two reviewers independently conducted a systematic review of MEDLINE, Embase, and Web of Science Core Collection databases from inception to January 2022 to identify studies that described symptoms of patients carrying pathogenic mutations in at least one of eight monogenic obesity genes (ADCY3, LEP, LEPR, MC3R, MC4R, MRAP2, PCSK1, and POMC). Of 5207 identified references, 269 were deemed eligible after title and abstra
