Pechiche (Vitex Cymosa Berteo ex Speng), a Nontraditional Fruit from Ecuador, is a Dietary Source of Phenolic Acids and Nutrient Minerals, in Addition to Efficiently Counteracting the Oxidative-Induced Damage in Human Dermal Fibroblasts.

Pechiche fruits (Vitex cymosa Berteo ex Speng) from Ecuador were studied to determine their phenolic acid profile, nutrient minerals and capacity to protect primary human dermal fibroblasts (HDFa) against oxidative-induced damage. Up to five phenolic acids were identified, with homovanillic acid as the main one. Vitamin C, β-carotene and lutein were also determined. Phosphorus and potassium were the main macrominerals, while iron was the principal micromineral. HDFa were preincubated with a crude pechiche extract (PCext) and then subjected to oxidative stress. The activity of five antioxidant

Long-term ethanol intake causes morphological changes in Schistosoma mansoni adult worms in mice.

Schistosoma mansoni adult worms are extensively challenged by reactive oxygen species from intrinsic sources. However, the effects of extrinsic sources such as ethanol have not been looked at in schistosomes. We examined adult worms recovered from ethanol-consuming mice by light (LM), confocal (CM) and scanning electron microscopy (SEM) to address this question. Schistosomiasis-infected mice were orally gavaged with 18% (v/v) ethanol from 35 to 63 days post-infection, when they were euthanized. CM examination revealed reduced germ cells density (-36%, p = 0.0001) and sperm density (-58%, p = 0

Role of Aldehyde Dehydrogenases in Physiopathological Processes.

Many different diseases are associated with oxidative stress. One of the main consequences of oxidative stress at the cellular level is lipid peroxidation, from which toxic aldehydes may be generated. Below their toxicity thresholds, some aldehydes are involved in signaling processes, while others are intermediaries in the metabolism of lipids, amino acids, neurotransmitters, and carbohydrates. Some aldehydes ubiquitously distributed in the environment, such as acrolein or formaldehyde, are extremely toxic to the cell. On the other hand, aldehyde dehydrogenases (ALDHs) are able to detoxify a w

Identification of platelet-activating factor as the inflammatory lipid mediator in CCl4-metabolizing rat liver.

Unmitigated oxidative stress is deleterious, as epitomized by CCl4 intoxication. In this well-characterized model of free radical-initiated damage, liver metabolism of CCl4 to CCl3. causes lipid peroxidation, F-ring isoprostane formation, and pathologic leukocyte activation. The nature of the mediator that couples oxidation to the hepatotoxic inflammatory response is uncharacterized. We found that oxidatively modified phosphatidylcholines were present in the livers of CCl4-exposed rats and not in livers from control animals, that CCl4 metabolism generated lipids that activated 293 cells stably

Development of AuxCuyPdz Nanocomposites as Therapeutic Agents: Enhancing Cancer Treatment through Autophagy Modulation and Immune-Associated Effects.

The development of multimetallic nanoparticles for cancer treatment represents a significant advancement in the field of nanomedicine. We introduce a Cu-templated synthesis method to create AuxCuyPdz hollow nanomicrostructures, wherein gold atoms stabilize copper (Cu) and facilitate the incorporation of palladium (Pd) through oxidation and coreduction processes. These ternary nanocomposites demonstrate enhanced cellular uptake via the copper transporter CTR1/2-mediated pathway and exhibit superior catalytic activity for the reaction of hydrogen peroxide to generate hydroxyl radicals. The prese

The Efficacy of Ultraviolet LED Cold Light Detoxification Technology in Removing Aflatoxin B1 from Peanut Oil and Its Application.

Peanut oil is prone to contamination with aflatoxin B1 (AFB1). Traditional ultraviolet (UV) irradiation methods demonstrate limited effectiveness in detoxification and may compromise the oil quality. To address this, this study developed a novel UV LED cold light source temperature control system, offering a new solution for AFB1 detoxification in peanut oil production. On the basis of this system, peanut oil samples spiked with varying concentrations of AFB1 were prepared and treated under different operational conditions. Then, the AFB1 content, acid value, and peroxide value of the decontam

Acidic plasma-activated povidone-iodine induces copper-dependent oxidative death in oral squamous cell carcinoma cells.

Oral squamous cell carcinoma (OSCC) requires therapeutic strategies that preserve oral function while providing effective tumor control. Here we demonstrate that non-thermal plasma (NTP) irradiation of a clinically established povidone-iodine solution generates a potent cytotoxic formulation termed plasma-activated povidone-iodine (PAI). PAI selectively reduced viability of OSCC cell lines (SAS, HSC-2, HSC-4) compared with normal fibroblasts. Mechanistically, PAI activity strictly depended on its acidic pH (3.0 or below) and hydrogen peroxide (H2O2) generated during NTP irradiation. Neutraliza

Tapinanthus globiferus mitigates arsenic-induced oxidative stress and genetic dysregulation In vivo.

Environmental toxins such as sodium arsenite induce oxidative stress, disrupt redox homeostasis, and trigger oncogenic signaling, providing a model for studying natural redox modulators. This study investigated the protective effects of Tapinanthus globiferus fractions (butanol, methanol, and ethyl acetate) on sodium arsenite-exposed Drosophila melanogaster. Biochemical, behavioral, and molecular assays were employed to assess oxidative stress markers, enzyme activities, locomotor performance, cell viability, and gene expression. Sodium arsenite exposure significantly decreased acetylcholinest

Biological Consequences of Single and Combined Exposure to Magnetite-Chitosan Nanocomposite with Adsorbed Cobalt (II) in Danio rerio.

Magnetic nanocomposite sorbents are increasingly explored for the remediation of metal-contaminated waters; however, high abiotic removal efficiency may not always translate into biological safety. The present study evaluated the single and combined effects of dissolved cobalt (II) ions and magnetite-chitosan nanocomposites (MCN) in zebrafish (Danio rerio) embryos and larvae. MCN (30 wt.% Fe3O4) were synthesized via co-precipitation and crosslinking and physiochemically characterized. Adsorption experiments conducted in fish incubation medium demonstrated the efficacy of divalent Co removal an

Metabolic sovereignty through oxidative hostility: a mechanistic perspective on how cancer engineers stromal dependency via ROS-mediated lysosomal reprogramming.

Cancer cells orchestrate a profound remodeling of their microenvironment to suppress immune surveillance and create metabolic dependency in surrounding stroma. We propose a mechanistic hypothesis in which this transformation is driven by a coordinated reactive oxygen species (ROS) signaling cascade. Cancer cells generate superoxide (O2•-) through NADPH oxidase (NOX) upregulation and mitochondrial respiration. Superoxide is rapidly converted to hydrogen peroxide (H2O2), a stable, membrane-diffusible ROS species that crosses stromal cell membranes via aquaporin channels. Within cancer-associated

Provoking Potent Cellular Ferroptosis in the Thermo-Sensitive S-Phase via CPT-11 Loaded Janus Nanomedicine for Enhanced Cancer Therapy.

Ferroptosis is a promising anticancer strategy, yet its efficacy is limited by insufficient cellular hydrogen peroxide (H2O2) levels and intrinsic antioxidant defenses. Meanwhile, mild photothermal therapy (mPTT) enhances ferroptosis by accelerating lipid peroxidation. Herein, we designed a Janus nanoplatform that specifically arrests cells in thermo-vulnerable S-phase to maximize the ferroptosis-sensitizing capacity of mPTT. A Janus architecture with a unique morphology was engineered to incorporate three functional modules: (1) A photothermal half-bowl composed of polydopamine (PDA) that gen

Reducing agents for induction and maintenance therapy achieve long-term remission of refractory ulcerative colitis: A case report and review of literature.

BACKGROUND: Ulcerative colitis (UC) is a chronic and debilitating inflammatory bowel disease. Cumulative evidence indicates that excess hydrogen peroxide, a potent neutrophilic chemotactic agent, produced by colonic epithelial cells has a causal role leading to infiltration of neutrophils into the colonic mucosa and subsequent development of UC. This evidence-based mechanism identifies hydrogen peroxide as a therapeutic target for reducing agents in the treatment of UC. CASE SUMMARY: Presented is a 41-year-old female with a 26-year history of refractory UC. Having developed steroid dependence

Near-infrared fluorescence probe achieves the early diagnosis and efficacy assessment of pesticide-mediated liver injury by monitoring H2O2.

Recently, pesticide residues have become a serious environmental hazard due to the widespread employment of pesticides in agricultural production. With the continuous accumulation of pesticide in vivo, liver injury caused by pesticide metabolism has become a rapidly developing disease. Although pesticide-mediated liver injury (PMLI) has attracted extensive attention, its diagnosis and treatment still face serious challenges. Previous studies have indicated that PMLI is closely related to oxidative stress caused by pesticides, in which the abnormal level of hydrogen peroxide (H2O2) is a critica

UV-B Radiation Triggers Phenolic Accumulation and Oxidative Stress Response in Lamiaceae Species: From Plant Defense to Green Dye Remediation.

The present study explores the impact of UV-B radiation on biochemical traits and phenolic profile of in vitro cultures (IC) and micropropagated plants (MP) of Lavandula viridis L'Hér and Thymus lotocephalus G. López and R. Morales. Two UV-B treatments were applied: a single 4 h exposure (UV-B 1) and repeated exposure over four consecutive days (UV-B 4). Additionally, the potential of phenolic-rich extracts loaded into alginate-based hydrogels for dye removal was also evaluated. UV-B exposure triggered oxidative stress in both species, particularly in MP, increasing hydrogen peroxide levels an

Mechanism of oxidative stress and neurotoxicity associated with heme and copper-Aβ relevant to Alzheimer's disease.

Alzheimer's disease (AD) is a neurodegenerative disorder with a complex pathophysiology involving oxidative stress, amyloid β (Aβ) aggregation and dysregulation of metal ions, particularly copper and heme. The overproduction of reactive oxygen species (ROS) plays a crucial role in the early stages of AD, leading to lipid peroxidation, protein oxidation, nucleic acid damage and neurotransmitter oxidation. These oxidative processes are further catalysed by the accumulation of Aβ peptides, which increase ROS production, creating a self-perpetuating cycle that accelerates disease progression. This

Iron single atom enzyme-mediated hydrogen sulfide delivery amplifies reactive oxygen species cascade to induce ferroptosis susceptibility.

Lipid peroxidation (LPO) represents one of the most deleterious processes contributing to ferroptosis susceptibility. However, the tumor microenvironment is often characterized by an overproduction of endogenous glutathione (GSH) and reactive oxygen species (ROS)-scavenging enzymes, which limit the ferroptosis susceptibility. Herein, we introduce an iron single-atom enzyme (Fe/SAE) nanoplatform, termed Fe/SAE@A, that acts as hydrogen sulfide (H2S) donor anethole trithione (ADT) delivery system to amplify LPO-mediated ferroptosis vulnerability. Upon internalization by cancer cells, Fe/SAE, feat

Ascorbic acid regulates in vitro and in vivo toxicogenetic effects of hydroxyurea on eukaryotic cells.

Hydroxyurea (HU) exerts unique and diverse biological effects as an anti-leukemic agent, irradiation sensitizer, and HbS inducer in patients with sickle cell anemia. Herein, we assessed the potential toxicogenic and/or oxidant effects of hydroxyurea associated with ascorbic acid by in vivo examinations in Allium cepa and human cancer cells and systemically on mice tissues. Growing A. cepa roots and HCT-116 colorectal tumor cells were examined after HU and HU plus ascorbic acid exposure. DNA damage and antioxidant enzymatic activity were quantified in peripheral blood mononuclear cells (PBMC),

Vapours from plant essential oils to manage tomato grey mould caused by Botrytiscinerea.

Tomato grey mould has been a great concern during tomato production. The in vitro antifungal activity of vapours emitted from four plant essential oils (EOs) (cinnamon oil, fennel oil, origanum oil, and thyme oil) were evaluated during in vitro conidial germination and mycelial growth of Botrytis cinerea, the causal agent of grey mould. Cinnamon oil vapour was the most effective in suppressing conidial germination, whereas the four EOs showed similar activities regarding inhibiting mycelial growth in dose-dependent manners. The in planta protection effect of the four EO vapours was also invest
