Cutaneous cryptococcosis simulating pyoderma gangrenosum.

A 33-year-old white man with a history of chronic ulcerative colitis presented with multiple cutaneous ulcers and an indurated cellulitic area on his right thigh. Clinically, the ulcers were considered to represent pyoderma gangrenosum. However, tissue biopsy revealed copious yeast forms that were identified as Cryptococcus neoformans. The indurated area on the right thigh later ulcerated, and tissue culture of this area also revealed Cryptococcus. This case illustrates that when pyoderma gangrenosum is diagnosed, the possibility of deep fungal infection should be considered and excluded by ap

[Acute pulmonary histoplasomis. 1st findings in Czechoslovakia].

A 57-year-old man, who for many years was treated for a pleomorphic lymphoma with cytotoxic drugs and x-ray therapy, died from the generalized tumor process. At autopsy, an extraordinary finding was found in form of acute pulmonary mycosts, which histologically appears to be histoplasmosis. Culture was not attempted, but the diagnosis is based on morphology, both of the organism proper and its intracellular location, and on results obtained by immunofluorescence studies. In numerous pulmonary vessels, endothelial pillows of histiocytes with yeast cells are found, typical for histoplasmosis, bu

Saccharomyces cerevisiae 48338 Suppresses Antibiotic-Induced Clostridioides difficile Infection in a Murine Model.

Clostridioides difficile infection (CDI) is a major cause of antibiotic-associated diarrhea and colitis, driven by toxin-mediated epithelial injury and inflammation. While antibiotics such as vancomycin remain the primary treatment, they can further disrupt the gut microbiota and promote recurrence. Probiotics, including yeast strains, have emerged as potential adjunctive therapies for mitigating CDI. In this study, several Saccharomyces cerevisiae strains were evaluated for their probiotic potential, and strain 48338 was identified as the most promising candidate based on its gastrointestinal

Antifungal Effects of Non-Thermal Atmospheric Pressure Plasma In Vitro and Ex Vivo.

BACKGROUND: Non-thermal atmospheric pressure plasma (NTAP) generates reactive oxygen species, reactive nitrogen species, and ultraviolet radiation, which can inactivate microorganisms. Onychomycosis treatment is challenging, and its prognosis is poor owing to mixed infections and dermatophytosis. Although NTAP has shown in vitro antifungal effects against dermatophytes and yeast, its efficacy against non-dermatophyte molds (NDMs) and in clinical or nail model studies remains poorly understood. OBJECTIVE: We evaluated the effects of NTAP on fungi, including NDMs, and infected nail plates. MET

GPATCH11 ortholog Sap34 regulates pre-mRNA splicing by interacting with early spliceosomal complexes in Schizosaccharomyces pombe.

Pre-mRNA splicing is an essential step in gene expression regulation. It is mediated by the spliceosome, a large ribonucleoprotein complex that undergoes dynamic structural and compositional rearrangements during each splicing cycle. Although the mechanisms of splicing and the roles of main spliceosomal components are well defined, the identities and functions of transiently associated spliceosomal proteins remain incompletely understood. Here, we investigated the molecular function of the poorly characterized G-patch domain-containing protein SPAC6F6.19 (herein Sap34, for spliceosome-associat

The transcription factor SbWRKY6 confers cadmium tolerance via activating SbPLAC8-17 expression in sorghum.

Cadmium (Cd) is a widespread environmental pollutant that severely threatens crop productivity and food safety. However, the regulatory mechanisms underlying Cd detoxification and tolerance in sorghum remain largely elusive. Herein, we functionally characterized SbWRKY6, a Cd‑induced WRKY transcription factor that localizes to the nucleus and functions as a transcriptional activator. Stable overexpression of SbWRKY6 significantly enhanced Cd tolerance in sorghum, as evidenced by improved growth performance, mitigated oxidative damage, and decreased Cd concentration in plant tissues, whereas si

The C2H2 zinc-finger protein CaZAT11 positively regulates cold tolerance in pepper and physically interacts with CaSTOP1.

Low temperature is a major environmental constraint that challenges pepper growth and productivity. Although C2H2 zinc-finger transcription factors have been implicated in plant responses to abiotic stresses, their roles in pepper cold tolerance remain poorly understood. In this study, we identified the C2H2 zinc-finger protein CaZAT11 as a novel interacting partner of the C2H2-type zinc-finger protein CaSTOP1 through yeast two-hybrid library screening. This interaction was subsequently confirmed by directed yeast two-hybrid, luciferase complementation imaging, and bimolecular fluorescence com

Sporobolomyces pararoseus: a versatile chassis for circular bioproduction of β-carotene, torulene, and torularhodin.

Recent advances in biomanufacturing have stimulated the exploration of microbial platforms for pigment production, offering alternatives to plant-derived and synthetic colorants. Among these biological solutions, microbial carotenoids, a group of naturally occurring pigments, have emerged as particularly valuable due to their vibrant red, orange, and yellow coloration coupled with demonstrated antioxidant capacity and health-promoting benefits. Sporobolomyces pararoseus, one of the most well-studied oleaginous red yeasts, exhibits a remarkable ability to simultaneously produce three commercial

In vitro bactericidal and fungicidal activities of commercially available low-irritant hand sanitizers.

BACKGROUND: High-concentration alcohol-based hand rubs (≥60% [v/v]) are strongly recommended to prevent healthcare-associated infections; however, their frequent use can aggravate hand eczema and reduce hand hygiene compliance. Therefore, low-irritant hand sanitizers with reduced alcohol content and alcohol-free formulations have been increasingly adopted in Japanese healthcare settings. Because these products typically contain benzalkonium chloride or chlorhexidine gluconate, along with ingredients that may affect bactericidal performance, this study evaluated the in vitro bactericidal and fu

Genome-wide identification of fatty acyl-CoA reductase gene family in cucumber and functional analysis of CsFAR3-like in cuticle wax formation and drought tolerance.

Fatty acyl-CoA reductases (FARs) catalyze the formation of primary alcohols, which are key components of plant cuticular wax. Although a few FAR members have been functionally characterized in Arabidopsis and several other species, their roles remain largely unexplored in Cucurbitaceae crops. Here, we identified five CsFAR genes in the cucumber genome, unevenly distributed across chromosomes 1, 4, and 6. Phylogenetic and collinearity analyses revealed that CsFARs share conserved evolutionary relationships with FARs from Arabidopsis thaliana, wheat, and other Cucurbitaceae species. Promoter cis

ZmGRXCC13, a key factor in coping with drought and saline-alkali stress.

Maize (Zea mays L.) is a paramount global staple crop; however, its production is severely limited by diverse abiotic stresses, particularly drought and saline-alkali stress. Although glutaredoxins (GRXs) are known to regulate plant growth and stress responses, their specific functions and underlying molecular mechanisms in maize under drought and saline-alkali conditions remain largely elusive. In this study, we cloned the maize GRX family member ZmGRXCC13 (Zm00001d037757) and characterized its biological functions in abiotic stress tolerance. Partial bypass of frataxin deficiency by ISCU M141I restores cytosolic and nuclear Fe-S cluster assembly.

Iron-sulfur (Fe-S) clusters are essential cofactors required for the activity of numerous proteins involved in fundamental cellular processes, including DNA replication, metabolism and mitochondrial respiration. In eukaryotes, Fe-S cluster biogenesis is initiated in mitochondria by the ISC machinery, which assembles iron and sulfur, delivered by a cysteine desulfurase, onto the scaffold protein ISCU. Frataxin (FXN), a key regulator of this pathway, enhances Fe-S production by accelerating persulfide transfer to ISCU. FXN is essential in eukaryotes, and its loss results in a "petite" phenotype

Extraction, structural characterization, and liposome encapsulation of alcohol dehydrogenase from pig liver: Potential applications in alcohol detoxification.

Alcohol dehydrogenase (ADH) is a key enzyme in ethanol metabolism and is abundant in pig liver, an underutilized animal by-product. Here, ADH was isolated from pig liver using aqueous two-phase extraction, and its enzymatic properties, structural characteristics, and biological relevance were systematically evaluated. The extracted ADH showed optimal activity at 45°C and pH 8.5, and its activity was enhanced by K+ and Mn2+. Spectroscopic analyses indicated overall structural similarity to yeast-derived ADH, while revealing a more ordered secondary structure with higher α-helix content. Kinetic

Ethanol production from rice straw: Comparative study of single and combined use of CaO/Ag nanocatalyst with Saccharomyces cerevisiae and Aspergillus terreus.

This research investigates a sustainable method for bioethanol synthesis from ammonia-pretreated rice straw utilizing a CaO/Ag nanocatalyst. The addition of ammonia elevated the available cellulose level of rice straw from 37.7% to 55.87%, thereby improving its suitability for enzymatic hydrolysis. The fungal isolate Aspergillus terreus (At PP590607) and the yeast Saccharomyces cerevisiae (Sc OR668931) were utilized both separately and in conjunction, with or without the CaO/Ag nanocatalyst. Antimicrobial testing revealed MICs of CaO/Ag nanoparticles ranging from 1 to 10 µg/ml, with sub-MIC do

Subcellular sirtuin signaling networks: pivotal regulators of cardiovascular homeostasis and remodeling.

Sirtuins represent a family of highly conserved enzymes, initially identified as Silent Information Regulator 2 (Sir2) in yeast, where they serve as fundamental determinants of longevity. Overexpression of Sir2 in yeast significantly extends lifespan, while its deletion leads to shortened longevity. In mammals, sirtuins (SIRT1-7) represent a conserved family of NAD+-dependent deacylases with diverse catalytic activities. While most members primarily function as deacetylases, SIRT4 exhibits mono-ADP-ribosylation activity, and SIRT5 uniquely targets negatively charged acyl groups, including lysi

Bi-allelic ATG12 variants impair autophagy and cause a neurodevelopmental disorder.

Autophagy is an essential developmental and homeostatic process, defined by the endolysosomal degradation of intracellular components and pathogens. Dysfunctional autophagy is implicated in complex human disease, yet reports of congenital autophagy disorders were considered exceedingly rare until the recent report of several unrelated families with bi-allelic variants in the core autophagy effector ATG7, complementing the report of two individuals harboring ATG5 variants. We now report six affected individuals from five families with bi-allelic ATG12 variants with complex neurological phenotyp
