Natural Antibodies Mediate Protection Against Acinetobacter baumannii Respiratory Infections.

BACKGROUND: Acinetobacter baumannii causes a wide range of dangerous infections due to the emergence of pandrug-resistant strains. Therefore, there is a need for alternative therapeutics to treat these infections, including those targeting the host immune responses. However, immune responses, especially the humoral response against this pathogen, are poorly understood. METHODS: This study investigated the lymphocyte-mediated innate immune resistance to A. baumannii AB5075 pulmonary infection using B- and T-cell-deficient (Rag2-/-) mice, the protective effect of natural antibodies (NAbs), and …

Motility of Acinetobacter baumannii: regulatory systems and controlling strategies.

Acinetobacter baumannii is a Gram-negative opportunistic zoonotic pathogenic bacterium that causes nosocomial infections ranging from minor to life-threatening. The clinical importance of this zoonotic pathogen is rapidly increasing due to the development of multiple resistance mechanisms and the synthesis of numerous virulence factors. Although no flagellum-mediated motility exists, it may move through twitching or surface-associated motility. Twitching motility is a coordinated multicellular movement caused by the extension, attachment, and retraction of type IV pili, which are involved in s…

Acinetobacter baumannii response to cefiderocol challenge in human urine.

Cefiderocol (CFDC) is a novel chlorocatechol-substituted siderophore antibiotic approved to treat complicated urinary tract infections (cUTI) and hospital-acquired and ventilator-acquired pneumonia (HAP/VAP). Previous work determined that albumin-rich human fluids increase the minimum inhibitory concentration (MICs) of Acinetobacter baumannii against CFDC and reduce the expression of genes related to iron uptake systems. This latter effect may contribute to the need for higher concentrations of CFDC to inhibit growth. The presence of human urine (HU), which contains low albumin concentrations,…

Hydrogen Sulfide Sensitizes Acinetobacter baumannii to Killing by Antibiotics.

The production of endogenous hydrogen sulfide (H2S) has been shown to confer antibiotic tolerance in all bacteria studied to date. Therefore, this mediator has been speculated to be a universal defense mechanism against antibiotics in bacteria. This is assuming that all bacteria produce endogenous H2S. In this study, we established that the pathogenic bacteria Acinetobacter baumannii does not produce endogenous H2S, giving us the opportunity to test the effect of exogenous H2S on antibiotic tolerance in a bacterium that does not produce it. By using a H2S-releasing compound to modulate the sul…

Prevalence of Aminoglycoside Resistance Genes in Acinetobacter baumannii Isolates.

BACKGROUND: Acinetobacter baumannii is one of the major causes of nosocomial infections and is resistant to most available antibiotics. Aminoglycosides remain as drugs of choice for treatment of Acinetobacter infections yet resistance to aminoglycosides has increased in the recent years. OBJECTIVES: The present study investigated the prevalence of genes encoding aminoglycoside-modifying enzymes in A. baumannii strains isolated from patients of Tabriz city, northwest of Iran. MATERIALS AND METHODS: A total of 103 Acinetobacter isolates were collected from Imam Reza Hospital of Tabriz Universi…

Crystal structure of carbapenemase OXA-58 from Acinetobacter baumannii.

Class D β-lactamases capable of hydrolyzing last-resort carbapenem antibiotics represent a major challenge for treatment of bacterial infections. Wide dissemination of these enzymes in Acinetobacter baumannii elevated this pathogen to the category of most deadly and difficult to treat. We present here the structure of the OXA-58 β-lactamase, a major class D carbapenemase of A. baumannii, determined to 1.30-Å resolution. Unlike two other Acinetobacter carbapenemases, OXA23 and OXA-24, the OXA-58 enzyme lacks the characteristic hydrophobic bridge over the active site, despite conservation of the…

[Necrotizing fasciitis caused by Acinetobacter baumannii : A case report].

A 42-year-old man developed necrotizing fasciitis on the right leg. A multidrug-resistant Acinetobacter baumannii was cultivated from the deep wound. Following therapy with imipenem and tobramycin as well as extensive debridement, the lesions improved slowly. A. baumannii is today an important cause of nosocomial infections, especially in intensive care units.

Two cases of necrotizing fasciitis due to Acinetobacter baumannii.

Necrotizing fasciitis has conventionally been associated with the streptococci, and when it is caused by other organisms, it is most often the result of a polymicrobial infection. We report on two cases of fatal monomicrobial necrotizing fasciitis due to Acinetobacter baumannii, an unusual finding that may be an indication of enhanced virulence of the organism.

Distinct role of outer membrane protein A in the intrinsic resistance of Acinetobacter baumannii and Acinetobacter nosocomialis.

Acinetobacter baumannii outer membrane protein A (AbOmpA) contributes to the intrinsic resistance of A. baumannii through the OmpA-like domain. The present study investigated the role of Acinetobacter nosocomialis OmpA (AnOmpA) in the intrinsic resistance of A. nosocomialis and compared it with the role of AbOmpA. The minimal inhibitory concentrations (MICs) of antimicrobial agents against wild-type A. nosocomialis ATCC 17903, ∆AnompA mutant, and single-copy AnompA-complemented strains were determined by performing E-test or agar dilution. Single-copy ompA cross-complemented strains were const…

Isolation, characterization and therapeutic evaluation of a new Acinetobacter virus Abgy202141 lysing Acinetobacter baumannii.

Acinetobacter baumannii is an opportunistic pathogen that easily resists currently available antibiotics. Phages are considered alternative therapeutic agents to conventional antibiotics for the treatment of multidrug-resistant bacteria. We isolated an Acinetobacter virus Abgy202141 from underground sewage in a residential area of Guiyang City in China. Transmission electron microscopy (TEM) analysis showed that Acinetobacter virus Abgy202141 has an icosahedral head attached to a tail. This phage infects A. baumannii strain GY-4, and was found to have a short latent period of 5 min and with a …

"In vitro evaluation of sulbactam combination therapies for Acinetobacter baumannii".

BACKGROUND AND OBJECTIVES: Acinetobacter baumannii, a leading cause of hospital-acquired infections in critically ill patients, is increasingly resistant to carbapenems, leading WHO to designate carbapenem-resistant Acinetobacter baumannii (CRAB) as a critical priority pathogen. While the 2024 IDSA guidelines recommend sulbactam-based regimens as the cornerstone of therapy, high-dose sulbactam is effective only against isolates with MICs up to 16-32 μg/mL. This study aimed to describe the sulbactam MIC distribution and to evaluate MIC reduction and synergy when combined with β-lactam antibioti…

The glutathione pathway is required for biofilm formation in Acinetobacter baumannii.

Acinetobacter baumannii is a Gram-negative nosocomial bacterium that is a member of the ESKAPE group of pathogens, notable for its virulence and intrinsic antibiotic resistance. It causes diverse infections, including respiratory and soft tissue disease, that are increasingly difficult to treat. Glutathione (GSH), the major intracellular redox buffer, is known in other bacteria to protect against stress and influence physiological processes such as biofilm formation. To investigate the roles of GSH in A. baumannii, we analyzed transposon mutant strains lacking gshA and gshB genes, which encode…

Genomic epidemiology of carbapenem resistance in Acinetobacter baumannii in Chile.

BACKGROUND: Carbapenem-resistant Acinetobacter baumannii (CRAb) is a World Health Organization (WHO) critical priority pathogen, yet genomic data from Chile are scarce. Here, we investigated the genetic diversity of Chilean isolates, focusing on molecular epidemiology, carbapenemase genes, and mobile genetic elements. METHODS: We analysed 83 A. baumannii genomes (42 newly sequenced, 41 public). Newly sequenced isolates underwent antimicrobial susceptibility testing, and whole-genome sequencing was performed using Illumina, with hybrid assemblies for two CRAb strains. Genomes were assembled an…

Roles of GacSA and DJ41_1407 in Acinetobacter baumannii ATCC 19606.

Two-component systems (TCSs) in bacteria are often involved in the global regulation of various physiological activities and behaviours. This study investigated the GacSA TCS and DJ41_1407 transcriptional sensor adjacent to GacA in Acinetobacter baumannii ATCC 19606. The relationship between GacS, GacA, and DJ41_1407 and their functions and signal transduction mechanisms are described. A. baumannii ATCC 19606 mutants, ∆gacS, ∆gacA, and ∆DJ41_1407, were generated using markerless mutation and cultured in LB medium, then collected for RNA sequencing. It was found that GacS, GacA, and DJ41_1407 r…

Arginine catabolism is essential to polymyxin dependence in Acinetobacter baumannii.

Polymyxins are often the only effective antibiotics against the "Critical" pathogen Acinetobacter baumannii. Worryingly, highly polymyxin-resistant A. baumannii displaying dependence on polymyxins has emerged in the clinic, leading to diagnosis and treatment failures. Here, we report that arginine metabolism is essential for polymyxin-dependent A. baumannii. Specifically, the arginine degradation pathway was significantly altered in polymyxin-dependent strains compared to wild-type strains, with critical metabolites (e.g., L-arginine and L-glutamate) severely depleted and expression of the ast…
