Optical microsensing reveals spatiotemporal oxygen dynamics in cornea wounds that affect healing via reactive oxygen species.

Oxygen (O2) metabolism plays a critical role in cornea wound healing, regeneration, and homeostasis; however, the underlying spatiotemporal mechanisms are poorly understood. Here we used an optical sensor to profile O2 flux in intact and wounded corneas of mouse eyes. Intact corneas have unique centrifugal O2 influx profiles, smallest flux at the cornea center, and highest at the limbus. Following cornea injury, the O2 influx profile presents three distinct consecutive phases: a "decreasing" phase from 0 to 6 h, a "recovering" phase from 12 to 48 h, and a 'peak' phase from 48 to 72 h, congruen

Transplantation with cultured stem cells derived from the human amniotic membrane for corneal alkali burns: an experimental study.

Amniotic membranes (AM) have been used in a wide range of clinical applications. We successfully extracted mesenchymal stem cells (MSCs) fro … rization within corneal alkali burns. Furthermore, dark blue-stained particles were detected in the limbus corneae at day 28. These results demonstrated the ability of hAM-dMSCs to enhance epithelial healing and reduce corneal opacification and neovascularization in corneal alkali wounds.

[Inhibition of corneal neovascularization by liposomes mediated plasmid encoding human endostatin].

OBJECTIVE: To evaluate the effect of liposomes mediated plasmid encoding endostatin (ES) for inhibiting experimental corneal neovascularization (CNV). METHODS: Thirty New Zealand albino rabbits were sutured on the superior cornea. Animals were randomly divided into 3 groups. Different reagents were injected at each group:liposomes and plasmid encoding human ES complex in the group 1, liposomes and carrier plasmid complex in the group 2 and saline in the group 3. The occurrence and development of CNV were observed by slit-lamp microscope 3, 7, 14, 21 and 28 days after suturing, the size of CNV

Angiopoietin-1 is regulated by miR-204 and contributes to corneal neovascularization in KLEIP-deficient mice.

PURPOSE: Corneal neovascularization can cause loss of vision. The introduction of anti-VEGF therapy has been a major improvement in therapeutic options. Recently, we established Kelch-like Ect2-interacting protein (KLEIP/KLHL20) knockout mice as a model of spontaneous corneal neovascular dystrophy. The aim of the present study was to characterize corneal neovascularization in progressive corneal dystrophy in KLEIP(-/-) mice, to evaluate the efficacy of anti-VEGF therapy, and to identify novel molecular regulators in this experimental model. METHODS: Corneal neovascularization was assessed by

Bilateral and massive lipoidal infiltration of the cornea (secondary lipoidal degeneration).

A 41-year-old white man presented with bilateral white infiltration of the cornea from limbus to limbus. Extensive examination revealed no manifestations of disorders of lipid metabolism and the patient gave no history of previous ocular disease. VDRL and other serological tests were negative. The corneal button removed by penetrating keratoplasty from the right eye was studied by light microscopy, histochemistry and electron microscopy. The light microscopic appearance was consistent with lipoidal degeneration of the cornea associated with stromal vascularization and chronic keratitis, more l

Identification of Two Distinct Stem Cell Clusters, Lrig1-Derived and Wnt/CD44-Dependent, in Corneal Epithelium.

We previously showed that selective suppression of CD44 in the corneal epithelium leads to structural abnormalities in the mouse cornea. Our comparative studies of young and aged ocular biopsies revealed that CD44 expression is downregulated in aged corneas, while leucine-rich repeats and immunoglobulin-like domain 1 (Lrig1+) stem cells remain preserved in the peripheral limbus. These findings suggest an age-related shift in the corneal stem cell compartmentalization, characterized by impaired CD44 expression in the central cornea and preservation of Lrig1+ stem cells in the limbus, which beco

Corneal abnormalities in Ehlers-Danlos syndrome type VI.

Eleven patients with blue sclera, limbus-to-limbus corneal thinning, hypermobile joints, and consanguineous parents were examined between January 1983 and September 1991. The clinical diagnosis was consistent with the Ehlers-Danlos syndrome type VI phenotype in all patients. A "halo" sign at the limbus was present in all patients. Corneal rupture occurred in seven patients (nine eyes) either spontaneously or following minimal trauma. Acute hydrops occurred in three patients. Bilateral microcornea was present in one patient and two patients had a unilateral increased corneal diameter as a resul

Suppurative interstitial ring keratitis due to streptococcus.

Three cases of interstitial ring abscess of the cornea caused by different strains of streptococcus are described. These infections occurred in three different clinical settings: after penetrating trauma at the limbus, after a cataract operation, and in conjunction with keratoconjunctivitis sicca. The acute appearance of a light, interstitial ring not extending to the limbus in the post-traumatized eye should alert the clinician to the possibility that a streptococcus may be responsible. The ring frequently progresses rapidly to a total ring abscess with intraocular extension. The outcome was

Plasticity of ocular surface epithelia: Using a murine model of limbal stem cell deficiency to delineate metaplasia and transdifferentiation.

Maintaining corneal health and transparency are necessary pre-requisites for exquisite vision, a function ascribed to stem cells (SCs) nestled within the limbus. Perturbations to this site or depletion of its SCs results in limbal SC deficiency. While characterizing a murine model of this disease, we discovered unusual transformation phenomena on the corneal surface including goblet cell metaplasia (GCM), conjunctival transdifferentiation, and squamous metaplasia (SQM). GCM arose from K8+ differentiated conjunctival epithelial cells when the limbus was breached and was exacerbated by neovascul

Rabbit model of phlyctenulosis and catarrhal infiltrates.

Phlyctenules and catarrhal infiltrates of the human cornea have been described in association with staphylococcal blepharitis. Rabbits immunized and boosted with phenol-inactivated Staphylococcus aureus had a fourfold or greater increase in antibody titer to S aureus and delayed hypersensitivity to S aureus. After topical challenge with viable S aureus, the rabbits in this model had vascularized, elevated, nodular infiltrates of the cornea resembling phylctenules in humans and peripheral corneal infiltrates running parallel to the limbus and separated from it by a lucid interval resembling cat

Corneal goblet cells and their niche: implications for corneal stem cell deficiency.

Goblet cells are terminally differentiated cells secreting mucins and antibacterial peptides that play an important role in maintaining the health of the cornea. In corneal stem cell deficiency, the progenitor cells giving rise to goblet cells on the cornea are presumed to arise from differentiation of cells that migrate onto the cornea from the neighboring conjunctiva. This occurs in response to the inability of corneal epithelial progenitor cells at the limbus to maintain an intact corneal epithelium. This study characterizes clusters of cells we refer to as compound niches at the limbal:cor

Primary Corneal Squamous Cell Carcinoma Confined to the Cornea: An Unusual Presentation of Ocular Surface Squamous Neoplasia Without Limbal Involvement.

Ocular surface squamous neoplasia manifests primarily on the limbus, with occasional extension onto the cornea. Primary corneal squamous cell carcinoma (SCC) devoid of limbal involvement is an exceptionally rare occurrence. The present case report details a 58-year-old male painter who experienced progressive blurred vision for three months and was diagnosed with SCC strictly confined to the corneal epithelium in his right eye, with no extension to the limbus. Clinical examination revealed significantly reduced visual acuity (0.2 or 20/100 with correction) and diffuse corneal epithelial opacit

Influence of corneal shape on limbal light focusing.

PURPOSE: Light incident at the temporal cornea is focused by the peripheral anterior eye to the nasal limbus, the usual site of pterygium formation. Parameters that may contribute to observed individual variations in the degree of limbal light focusing were assessed. METHODS: Computer-assisted optical ray tracing techniques were applied to a human anterior segment model. The angle of incident light (theta, 95 degrees to 108 degrees posterior to the sagittal plane), corneal central radius of curvature (ro, 7.2 to 8.4 mm), and shape factor (p) were varied, and the effect on distal limbal intens
