Optimized DNA extraction and purification method for characterization of bacterial and fungal communities in lung tissue samples.

Human lungs harbor a scarce microbial community, requiring to develop methods to enhance the recovery of nucleic acids from bacteria and fungi, leading to a more efficient analysis of the lung tissue microbiota. Here we describe five extraction protocols including pre-treatment, bead-beating and/or Phenol:Chloroform:Isoamyl alcohol steps, applied to lung tissue samples from autopsied individuals. The resulting total DNA yield and quality, bacterial and fungal DNA amount and the microbial community structure were analyzed by qPCR and Illumina sequencing of bacterial 16S rRNA and fungal ITS gene

Investigating the shared genetics of non-syndromic cleft lip/palate and facial morphology.

There is increasing evidence that genetic risk variants for non-syndromic cleft lip/palate (nsCL/P) are also associated with normal-range variation in facial morphology. However, previous analyses are mostly limited to candidate SNPs and findings have not been consistently replicated. Here, we used polygenic risk scores (PRS) to test for genetic overlap between nsCL/P and seven biologically relevant facial phenotypes. Where evidence was found of genetic overlap, we used bidirectional Mendelian randomization (MR) to test the hypothesis that genetic liability to nsCL/P is causally related to imp

Enantiospecific determination of naftopidil by RRLC-MS/MS reveals stereoselective pharmacokinetics and tissue distributions in rats.

Naftopidil (NAF) is used as a racemate to treat benign prostatic hyperplasia (BPH) and to prevent prostate cancer. However, racemic NAF has low bioavailability; therefore, it is commonly administered at higher clinical dosages compared to other therapeutic BPH drugs. Differences in interactions between individual enantiomers and biological macromolecules may result in variations in pharmacokinetics and dispositions. This study aimed to investigate the pharmacokinetics, bioavailability and tissue distributions of NAF enantiomers in rats after intragastric administration of the individual enanti

Molecular and in silico analysis of BRCA1 and BRCA2 variants.

Germline mutations of high penetrant BRCA1 and BRCA2 genes have been associated to hereditary breast cancer risk, while polymorphic variants of the two genes still have an unknown role in breast pathogenesis. The aim of our study was to characterize BRCA1 and BRCA2 genes polymorphic variants in familial breast cancer. 110 patients affected by familial breast and/or ovarian cancer have been consecutively enrolled according to family history and BRCA mutation risk. All of them have been screened for BRCA1 and BRCA2 pathogenetic mutations, SNPs and intronic variants. In silico analysis have been

Autonomic reactivity and clinical severity in children with sickle cell disease.

Individual differences in autonomic nervous system reactivity have been studied in relation to physical and mental health outcomes, but rarely among children with chronic disease. The purpose of this study was to examine the associations among autonomic reactivity, clinical severity, family stressors, and mental health symptoms in children with homozygous sickle cell disease. Nineteen children with homozygous sickle cell disease participated in a cross-sectional study involving parent-completed measures, medical record reviews and laboratory-based measures of autonomic nervous system responses

Genistein in polycystic ovary syndrome: mechanisms, preclinical evidence, and translational potential.

Polycystic ovary syndrome (PCOS) is a prevalent reproductive-endocrine disorder in reproductive-aged women. Due to the lack of radical cure, multi-target therapeutic strategies are urgently needed. Genistein, a soy-derived phytoestrogen, exhibits pleiotropic biological activities and is a promising adjuvant candidate for PCOS. Accumulating studies suggests that genistein may improves insulin sensitivity, regulates reproductive hormones, promotes follicular development, and alleviates oxidative stress and chronic inflammation, thereby ameliorating PCOS symptoms. This review summarizes the thera

Modeling Alzheimer's disease with human multi-omics profiles: Promise to personalized medicine.

Alzheimer's disease (AD) presents considerable heterogeneity in disease risk and outcomes, posing a major challenge for effective therapeutic development. Recent advances in multi-omics profiling are revolutionizing our understanding of the complex AD pathogenesis. Leveraging long-read sequencing and large-scale multi-ancestry genetic studies, researchers are unraveling the complex polygenic architecture of AD. Additionally, advances in multi-omics techniques unlock the power of molecular profiling of human biological samples, particularly spatial omics of human postmortem brain tissues and mo

Implications of gene × environment interactions in post-traumatic stress disorder risk and treatment.

Exposure to traumatic stress is common in the general population. Variation in the brain's molecular encoding of stress potentially contributes to the heterogeneous clinical outcomes in response to traumatic experiences. For instance, only a minority of those exposed to trauma will develop post-traumatic stress disorder (PTSD). Risk for PTSD is at least partially heritable, with a growing number of genetic factors identified through GWAS. A major limitation of genetic studies is that they capture only the genetic component of risk, whereas PTSD by definition requires an environmental traumatic

A multimodal vision knowledge graph of cardiovascular disease.

Understanding gene-disease associations is important for uncovering pathological mechanisms and identifying potential therapeutic targets. Knowledge graphs can represent and integrate data from multiple biomedical sources, but lack individual-level information on target organ structure and function. Here we develop CardioKG, a knowledge graph that integrates over 200,000 computer vision-derived cardiovascular phenotypes from biomedical images with data extracted from 18 biological databases to model over a million relationships. We used a variational graph auto-encoder to generate node embeddi

Associations of self-identified race and ethnicity and genetic ancestry with mortality among cancer survivors.

Self-identified race and ethnicity (SIRE) and genetic ancestry are potentially associated with disparities in health outcomes; however, independent effects of SIRE and genetic ancestry on mortality in cancer survivors including when adjusting for multiple risk factors are understudied. Among 23 445 cancer survivors in the Prostate, Lung, Colorectal, and Ovarian Screening Trial, SIRE was associated with mortality among prostate, colorectal, lung, ovarian, and breast cancer survivors; genetic ancestry was associated with mortality among prostate, colorectal, and breast cancer survivors. The menopause transition: endocrine changes and clinical symptoms.

Several biological changes take place during the menopause transition. The number of oocytes declines progressively from before birth but reaches a critically low level by the time of the menopause. The regular pattern of the menstrual cycle becomes disrupted and the frequency of normal ovulatory cycles declines. Rising gonadotrophin levels, particularly of follicle stimulating hormones (FSH), and declining estrogen levels are thought to characterize the menopausal transition. It now appears that declining levels of inhibin may play an important role in maintaining estrogen levels until just b

Artificial intelligence and machine learning in smart vaginal formulation development.

Vaginal drug delivery in women's health remains underutilized and insufficiently studied, largely due to the complexity and dynamic nature of the vaginal microenvironment. Variations in vaginal pH, hormonal levels, and microbiota composition introduce significant biological variability, complicating formulation design and contributing to inconsistent therapeutic outcomes and poor patient adherence. Conventional vaginal formulations often fail to account for these individual differences, highlighting the need for more adaptive and predictive approaches. Emerging advances in artificial intellige

Potential factors contributing to extreme longevity in the Greenland shark.

The Greenland shark (Somniosus microcephalus) is a deep-sea vertebrate inhabiting the cold waters of the North Atlantic and Arctic Ocean and is renowned for its exceptional longevity, with individuals estimated to live for more than 400 years. It has also been proposed as a candidate species exhibiting negligible senescence. This narrative review synthesizes current knowledge on the biological mechanisms that may contribute to this phenotype. Potential contributing factors include its extreme environment, low metabolic rate and remarkably late sexual maturation, all of which may reduce cumulat

Burden of negative energy balance in anorexia nervosa: Role of circadian rhythm, mitochondrial dynamics, microbiota-gut-brain axis and their interactions.

Anorexia nervosa (AN) is a chronic eating disorder that often requires prolonged hospitalisation for effective treatment. As a multifactorial disease, many environmental, psychiatric, and biological factors impede recovery, creating a vicious circle of anorexia, gastrointestinal disorders, and anxiety-depressive disorders. Individuals with AN often exhibit inappropriate physical activity, sleep disturbances, alterations in circadian hormonal profiles, disruption of the gut barrier, dysbiosis of the gut microbiota, oxidative stress, and alterations in brain function. The aim of this review is t

A saturated map of common genetic variants associated with human height.

Common single-nucleotide polymorphisms (SNPs) are predicted to collectively explain 40-50% of phenotypic variation in human height, but identifying the specific variants and associated regions requires huge sample sizes1. Here, using data from a genome-wide association study of 5.4 million individuals of diverse ancestries, we show that 12,111 independent SNPs that are significantly associated with height account for nearly all of the common SNP-based heritability. These SNPs are clustered within 7,209 non-overlapping genomic segments with a mean size of around 90 kb, covering about 21% of the

Comparison of markers of coagulation activation and thrombin generation test in uncomplicated pregnancies.

INTRODUCTION: Pregnancy is a well-established risk factor for venous thromboembolism, and is associated with a state of hypercoagulability or parameters of thrombin generation. Currently, there is a lack of consensual data on thrombin generation during pregnancy. This study aimed to find a sensitive and specific biological marker of coagulation activation and to identify parameters of thrombin generation. PATIENTS AND METHODS: The population included 101 women with uncomplicated pregnancies. The objective of this study was to correlate thrombin generation test (measured at 5pM tissue factor,

T-cell lymphomas in South america and europe.

Peripheral T-cell lymphomas are a group of rare neoplasms originating from clonal proliferation of mature post-thymic lymphocytes with different entities having specific biological characteristics and clinical features. As natural killer cells are closely related to T-cells, natural killer-cell lymphomas are also part of the group. The current World Health Organization classification recognizes four categories of T/natural killer-cell lymphomas with respect to their presentation: disseminated (leukemic), nodal, extranodal and cutaneous. Geographic variations in the distribution of these diseas

Bone apposition to plasma-sprayed cobalt-chromium alloy.

The use of porous metallic coatings for fixation of total joint prostheses by bone ingrowth has become a widespread alternative to fixation with PMMA bone cement. However, concerns about such coatings include long-term effects of metal ion release, potential coating loss, and decreased substrate fatigue strength. The biological fixation capability of a nonporous, high-integrity plasma-sprayed CoCr coating with low surface area was compared to a conventional sintered bead coating in goat cortical and cancellous bone sites after 8 and 16 weeks of implantation. Histological evaluation showed subs

Partner HIV Serostatus Impacts Viral Load, Genital HIV Shedding, and Immune Activation in HIV-Infected Individuals.

Studies of seronegative individuals in HIV discordant relationships provide important insights into the effects of HIV exposure on the seronegative partner, but few have examined the impact of partner serostatus on disease progression in seropositive individuals. We investigated the impact of HIV serostatus on clinical and biological factors influencing HIV disease progression in 337 HIV-infected heterosexual individuals in stable long-term HIV-seroconcordant or HIV-serodiscordant relationships. Seroconcordant individuals had significantly higher plasma viral loads (pVLs) than HIV-infected par

Skeletal Age Evaluation with BAUSportTM Ultrasound in Young Slovak Female Athletes and Non-Athletes-A Cross-Sectional Study.

Background: Biological maturity assessment is increasingly discussed in youth sport because chronological age alone does not reflect inter-individual variation in growth and pubertal timing. Ultrasound-based systems such as BAUSport™ provide a radiation-free alternative to radiographic skeletal age assessment. This exploratory cross-sectional study examined skeletal age (SA) in Slovak female volleyball players, track-and-field athletes, and non-athletes using BAUSport™, while acknowledging substantial age differences between groups. Methods: Eighty girls (55 non-athletes, 13 volleyball players
