Transradial access for flow diversion of intracranial aneurysms: Case series.

BACKGROUND: Transradial access for neurointerventions offers advantages of fewer access-site complications, reduced procedure time, and greater patient comfort over transfemoral access. Data about transradial access for flow diversion are limited. We share our early experience with transradial access for flow diversion in a relatively large case series. METHODS: Consecutive patients who underwent Pipeline embolization device (Medtronic) deployment via transradial access were included in the study, irrespective of location and laterality of the intracranial aneurysm. The cases were performed b

TRK Fusions Are Enriched in Cancers with Uncommon Histologies and the Absence of Canonical Driver Mutations.

PURPOSE: TRK inhibitors achieve marked tumor-agnostic efficacy in TRK fusion-positive cancers and consequently are now an established standard of care. Little is known, however, about the demographics, outcomes, response to alternative standard therapies, or genomic characteristics of TRK fusion-positive cancers. EXPERIMENTAL DESIGN: Utilizing a center-wide screening program involving more than 26,000 prospectively sequenced patients, genomic and clinical data from all cases with TRK fusions were extracted. An integrated analysis was performed of genomic, therapeutic, and phenomic outcomes.

Genomic sites hypersensitive to ultraviolet radiation.

If the genome contains outlier sequences extraordinarily sensitive to environmental agents, these would be sentinels for monitoring personal carcinogen exposure and might drive direct changes in cell physiology rather than acting through rare mutations. New methods, adductSeq and freqSeq, provided statistical resolution to quantify rare lesions at single-base resolution across the genome. Primary human melanocytes, but not fibroblasts, carried spontaneous apurinic sites and TG sequence lesions more frequent than ultraviolet (UV)-induced cyclobutane pyrimidine dimers (CPDs). UV exposure reveale

Advanced Genetic Approaches in Discovery and Characterization of Genes Involved With Osteoporosis in Mouse and Human.

Osteoporosis is a complex condition with contributions from, and interactions between, multiple genetic loci and environmental factors. This review summarizes key advances in the application of genetic approaches for the identification of osteoporosis susceptibility genes. Genome-wide linkage analysis (GWLA) is the classical approach for identification of genes that cause monogenic diseases; however, it has shown limited success for complex diseases like osteoporosis. In contrast, genome-wide association studies (GWAS) have successfully identified over 200 osteoporosis susceptibility loci with

Heterogeneity of asthma and the risk of celiac disease in children.

BACKGROUND: Although human leukocyte antigen (HLA)-DR and HLA-DQ genes and gluten play crucial roles in developing celiac disease (CD), most patients with these risk factors still do not develop CD, which indicates additional unrecognized risk factors. OBJECTIVE: To determine the association between asthma and the risk of CD in children. METHODS: We conducted a population-based retrospective case-control study in children who resided in Olmsted County, Minnesota. We identified children with CD (cases) between January 1, 1997, and December 31, 2014, and compared these with children without CD

Genetic evidence for a causal relationship between melatonin metabolism and depression.

To investigate the causal relevance of melatonin metabolism, which provides the biological basis for circulating melatonin levels, to specific depression symptom subtypes, we performed a targeted systematic review of melatonin metabolism pathways in the human brain and liver. Using two-sample Mendelian randomization (MR), we assessed the causal effects of metabolism pathways and/or individual genes on major depressive disorder (MDD) and nine symptom subtypes derived from Patient Health Questionnaire-9 (PHQ-9). Instrumental variables (IVs) were expression quantitative trait loci (eQTL) for eigh

Circulating Plasma Proteins Influence the Risk of Aortic Dissection via Blood Pressure: A Network Mendelian Randomization and Multi-Omics Study.

BACKGROUND: The causal link between plasma proteins and aortic dissection (AD), and the role of blood pressure (BP) in this relationship, is not well understood. METHODS: We applied an integrated network Mendelian randomization (MR) and multi-omics framework to investigate the causal effects of plasma proteins on AD and the mediating role of BP. Genetic instruments for plasma proteins were derived from deCODE genetics (n = 35,559), and genome-wide association studies (GWAS) data for BP and AD were sourced from UK Biobank (n = 458,855), Million Veteran Program (n = 449,042) and FinnGen (n = 46

Shared genetic architecture of psychoactive substance use and pan-cancer: insights from a large‑scale genome‑wide cross‑trait analysis.

BACKGROUND: Psychoactive substance use (PSU) and cancer are frequently observed comorbidities that have reciprocal influences and shared behavioral traits of the affected patients. While, e.g., nicotine and alcohol are major carcinogens in the etiology of lung and head and neck cancers, little is known about a shared overarching genetic architecture of PSU and cancer that may predispose individuals to both illnesses. METHODS: Large-scale genome-wide association study (GWAS) summary data revealed shared genetic architecture between cancer and PSU, including alcohol use dependence (AlcUD) and n

Integrative multi-omics analysis identifies GNAZ and SLC5A4 as critical regulators of gestation length in Large White pigs.

BACKGROUND: As a key index of reproductive performance in pigs, gestation length (GL) exerts a direct influence on litter size and the survival rate of piglets. Phenotypic variability in GL has been observed across Large White pig populations. To elucidate the genetic mechanisms underlying GL, this study analyzed 22,783 reproductive records from 9,057 pigs across five Large White populations. We employed a combination of diverse analytical strategies, encompassing genome-wide association studies (GWAS), GWAS meta-analysis, Bayesian fine-mapping (BFM), transcriptome-wide association studies (TW

Single-cell transcriptome-wide Mendelian randomization and colocalization reveal cell-specific mechanisms in systemic lupus erythematosus.

OBJECTIVE: SLE is a multifactorial autoimmune disease with complex genetic architecture and immune cell involvement. While genome-wide association studies (GWAS) have identified numerous risk loci, most are non-coding, making it challenging to pinpoint causal eGenes [genes with expression quantitative trait loci (eQTLs)] and therapeutic targets. METHODS: We integrated single-cell expression quantitative trait loci (sc-eQTL) data from 14 human immune cell types with Mendelian randomization (MR) and Bayesian colocalization analyses to identify eGenes causally associated with SLE. We applied phe

The first genome-wide association study on pediatric obesity in Taiwan.

BACKGROUND: The global prevalence of pediatric obesity has risen sharply, increasing from 0.7% to 5.6% in girls and from 0.9% to 7.8% in boys between 1975 and 2016. Genetic factors contribute an estimated 50-80% to obesity risk. In 2007, FTO was identified as the first obesity-associated gene through a genome-wide association study (GWAS), but research in Asian pediatric populations remains limited. This study investigated the genetic architecture of pediatric obesity in Taiwanese children. METHODS: Data were obtained from the China Medical University Hospital Biobank. Obese cases were define

Mapping the Immune cell-specific gene regulatory network in bipolar disorder: A framework from scTWMR to exploratory drug-target annotation.

BACKGROUND: Although the involvement of the immune system in the genetic susceptibility of bipolar disorder (BD) is widely acknowledged, the causal relationship between gene expression in specific immune cell subtypes and BD requires systematic elucidation. METHODS: We implemented an analytical framework integrating single-cell transcriptome-wide Mendelian randomization (scTWMR) with colocalization analysis. This approach utilized cis-expression quantitative trait loci (cis-eQTLs) derived from 14 distinct immune cell types as instrumental variables to interrogate BD genome-wide association st

Identification of patients at risk for pancreatic cancer in a 3-year timeframe based on machine learning algorithms.

Early detection of pancreatic cancer (PC) remains challenging largely due to the low population incidence and few known risk factors. However, screening in at-risk populations and detection of early cancer has the potential to significantly alter survival. In this study, we aim to develop a predictive model to identify patients at risk for developing new-onset PC at two and a half to three year time frame. We used the Electronic Health Records (EHR) of a large medical system from 2000 to 2021 (N = 537,410). The EHR data analyzed in this work consists of patients' demographic information, diagn

Transdiagnostic and Disorder-Level Genome-Wide Association Studies Enhance Precision of Substance Use and Psychiatric Genetic Risk Profiles in African and European Ancestries.

BACKGROUND: Substance use disorders (SUDs) and psychiatric disorders frequently co-occur, and their etiology likely reflects both transdiagnostic (i.e., common/shared) and disorder-level (i.e., independent/nonshared) genetic influences. Understanding the genetic influences that are shared and those that operate independently of the shared risk could enhance precision in diagnosis, prevention, and treatment, but this remains underexplored, particularly in non-European ancestry groups. METHODS: We applied genomic structural equation modeling to examine the common and independent genetic archite

Multi-ancestry genome-wide association study of topiramate's effects on heavy alcohol use.

BACKGROUND: Topiramate reduces alcohol consumption in individuals who drink heavily. Candidate gene studies aimed at identifying genetic variants that predict topiramate's effects on drinking have yielded inconsistent findings. To identify genetic variation associated with treatment response, we conducted a genome-wide association study (GWAS) among participants in the Million Veteran Program (MVP) who initiated topiramate treatment. METHODS: Using electronic health records, we identified individuals who were dispensed topiramate for at least 60 days for any indication (i.e., the index event)
