Directional transit of the ribosome along the messenger RNA (mRNA) template is a key determinant of the rate and processivity of protein synthesis. Imaging of the multistep translocation mechanism using single-molecule FRET has led to the hypothesis that substrate movements relative to the ribosome resolve through relatively long-lived late intermediates wherein peptidyl-tRNA enters the P site of the small ribosomal subunit via reversible, swivel-like motions of the small subunit head domain within the elongation factor G (GDP)-bound ribosome complex. Consistent with translocation being rate-l

Fluoropharmacology integrates pharmacokinetics/pharmacodynamics with real-time visualization by employing intrinsically fluorescent drugs that combine therapy and luminescence in a single entity. Herein, we report the systematic characterization that clinically approved quinolone antibiotics exhibit intrinsic aggregation-induced emission (AIE) characteristics. Mechanistic studies reveal that the AIE behavior originates from restricted intramolecular motion, thereby establishing the "drug-probe unity" principle that a single molecular architecture inherently encodes pharmacological activity, fl

AMPA receptors (AMPARs) mediate synaptic transmission and plasticity during learning, development, and disease. Mechanisms determining subsynaptic receptor position are poorly understood but are key determinants of quantal size. We used a series of live-cell, high-resolution imaging approaches to measure protein organization within single postsynaptic densities in rat hippocampal neurons. By photobleaching receptors in synapse subdomains, we found that most AMPARs do not freely diffuse within the synapse, indicating they are embedded in a matrix that determines their subsynaptic position. Scintillators, the luminescent materials that convert high-energy radiation into photons with lower energy, act widely as the key component in X-ray imaging systems. The scintillators would typically have high light yield, low detection limit, as well as high environmental stability, which enables them to work in outdoor harsh environments. Herein, we report a novel one-dimensional hybrid copper(I) halide, N,N,N',N'-tetramethylethylenediamine copper(I) iodide ([TMEDA]Cu2I4). The bulk single crystals are obtained by solvent evaporation. The sample shows broadband blue emission under UV excitati

Nitric oxide (NO) and methylglyoxal (MGO) play a vital part in maintaining redox homeostasis, modulating substances, and signal transduction. Fluctuations in these signaling molecules are closely associated with various pathological processes. However, due to the absence of appropriate multifunctional fluorescent sensors, concurrent identification of NO and MGO has not been achieved in inflammatory diseases. Herein, a dual-responsive near-infrared window II fluorescent probe (TPA-4T) was rationally developed to monitor NO and MGO through its distinct emission channels concurrently. Upon NO exp

Polymers are widely applied to drug delivery systems because polymers are generally excreted from the body more slowly than small molecules. Subcutaneous injection is one plausible means of administration. In this study, the in vivo behaviors of subcutaneously injected polymers, linear poly(glutamic acid) (Poly-Glu), acetylated dendrimer (Ac-den) and collagen peptide-conjugated dendrimer (CP-den), were investigated. Single photon emission computed tomography (SPECT) imaging was used to noninvasively monitor the in vivo behaviors. Diethylenetriaminepentaacetic acid (DTPA) was conjugated to thes

Prior studies have largely focused on transcriptional and translational control during stress, but how regulated nuclear mRNA export contributes to the stress response remains unresolved. We show that nuclear mRNA export is progressively inhibited during arsenite and heat stress in human cells. In contrast to previous work largely in yeast that suggests nuclear export of stress-induced transcripts is prioritized through sequence-specific mechanisms, we found that mRNA export is governed by temporal gating, in which the timing of mRNA biogenesis determines the nucleocytoplasmic distribution of

In the sperm nucleus, protamine replaces histones to mediate extreme DNA compaction. The histone-to-protamine transition involves the occurrence of double-strand breaks, and is facilitated by transition proteins including those containing high-mobility-group boxes. Here we use optical tweezers and microscopy to study the actions of HMGB1 and protamine on DNA. Confocal scans of GFP-HMGB1 on overstretched λ-DNA show 2-3 foci that spread on the DNA upon retraction. Spreading of foci coincides with reannealing of ssDNA tracts, confirming their localization at ss-dsDNA junctions. Whereas the force-

IR-780 is a lipophilic heptamethine cyanine dye that exhibits strong absorption and fluorescence emission in the near-infrared (NIR) region (700-900 nm). Similar to other cyanine dyes, IR-780 is emerging as a promising fluorophore for fluorescence-guided surgery (FGS). In addition, due to its high molar extinction coefficient, IR-780 can generate reactive oxygen species (ROS), enable efficient conversion of light into heat, and impart additional phototherapeutic capabilities. Consequently, IR-780 represents a promising "all-in-one" single molecule for FGS and phototherapy applications. However

AIMS: This study aims to resolve the mechanisms underlying Calmodulin (CaM)'s signalling diversity by investigating whether the three CaM genes-Calm1, Calm2, and Calm3-play distinct or redundant roles in cardiac myocytes, focusing on their spatial mRNA localization and interactions with key targets.
METHODS AND RESULTS: We utilized single-molecule mRNA detection and three-dimensional imaging to map the spatial distribution of Calm1, Calm2, and Calm3 mRNAs within ventricular myocytes. These mRNAs were found to be consistently positioned within specific cellular zones, overlapping with their ta

Hydrogen sulfide (H2S) is one of the critical gasotransmitters, which play important roles in regular physiological processes, especially in vital signaling pathways. However, fluctuations in endogenous H2S concentration can be linked to serious health problems, such as neurodegenerative diseases, cancer, diabetes, inflammation, cardiovascular diseases, and hypertension. Thus, it has attracted a great deal of attention in therapeutic applications, specifically in the field of phototherapy. Photodynamic therapy (PDT) and photothermal therapy (PTT) are two subclasses of phototherapy, which utili

Low-power laser irradiation (LPLI) has been shown to exert promotive effects on cell survival and proliferation through activation of various signaling pathways. Estrogen receptors (ERs, ERα, and ERβ) are ligand-activated transcription factors, which regulate target gene expression, promote cell proliferation, and resist apoptosis. However, it is unclear whether LPLI could induce ligand-independent activation of ERs. In the present study, we investigated the subcellular pools, nuclear redistribution, and transcriptional activity of ERs under LPLI (1.2 J/cm(2), 633 nm) treatment using single-mo

BACKGROUND & AIMS: Radiation-induced enteritis develops in cancer patients treated with radiotherapy in the abdominal and pelvic cavity, a condition that impairs their quality of life. Radiation injury depletes proliferative intestinal stem cells; in response to this, the epithelium activates a regenerative program that facilitates the healing of the intestine. However, the mechanisms that induce the activation of the intestinal regenerative program are poorly characterized.
METHODS: In this study, we induced radiation-induced enteritis in mice through abdominal irradiation, mimicking clinica

This issue of the Biomedical Journal highlights regulatory mechanisms that shape aging, disease progression, and biological complexity across molecular, cellular, and systems levels. A central theme is plasminogen activator inhibitor-1 (PAI-1) as a multifunctional regulator linking fibrinolysis, cellular senescence, stem cell dynamics, fibrosis, and tumor biology. Contributions examine structure-guided development of small-molecule PAI-1 inhibitors and their therapeutic potential across thrombotic disease, chronic myeloid leukemia, systemic sclerosis, lung cancer, and skin cancer, emphasizing

Sonodynamic therapy (SDT) utilizes ultrasound to activate sonosensitive agents, generating reactive oxygen species (ROS) within tumor cells to induce oxidative damage. The advantages of ultrasound, such as deep tissue penetration, non-invasiveness, and spatial controllability, make it a promising candidate for cancer treatment. However, its practical application is still limited by factors such as low in vivo delivery efficiency, tumor hypoxia and antioxidant defense mechanisms, insufficient conversion of immunogenic cell death into durable anti-tumor immunity, and the lack of standardized ult

Dimethyl sulfoxide (DMSO) is a polar aprotic solvent used in a wide range of applications, including uses as a drug and in drug delivery, as a solvent for fluorescent dyes, and in enzymatic reactions that process DNA. Consequently, many assays contain low concentrations (≤10%) of DMSO. While it is well known that DMSO lowers the melting temperature of DNA, its effects on DNA conformations and mechanical properties below the melting temperature are unclear. Here, we use complementary single-molecule techniques to probe DNA in the presence of 0-60% DMSO. Magnetic tweezers force-extension measure

Cystic fibrosis (CF) is the most common lethal genetic disease among Caucasians. It is caused by mutations in the CF Transmembrane Conductance Regulator (CFTR) gene, which encodes an apical membrane anion channel that is required for regulating the volume and composition of epithelial secretions. The most common CFTR mutation, present on at least one allele in >90% of CF patients, deletes phenylalanine at position 508 (F508del), which causes the protein to misfold. Endoplasmic reticulum (ER) quality control elicits the degradation of mutant CFTR, compromising its trafficking to the epithelial

MicroRNAs (miRNAs) are a class of small noncoding RNAs that act as pivotal post-transcriptional regulators of gene expression, thus involving in many fundamental cellular processes such as cell proliferation, migration, and canceration. The detection of miRNAs has attracted significant interest, as abnormal miRNA expression is identified to contribute to serious human diseases such as cancers. Particularly, miRNAs in peripheral blood have recently been recognized as important biomarkers potential for liquid biopsy. Furthermore, as miRNAs are expressed heterogeneously in different cells, invest
