UPLC-Q-TOF/MS-Based Metabolomics and 16S rRNA Profiling Reveal that Corosolic Acid Ameliorates High-Fat Diet-Induced MASLD by Modulating the Gut-Liver Axis to Inhibit the cGAS-STING Pathway.

Metabolic-associated steatohepatitis liver disease (MASLD) is characterized by abnormal hepatic fat accumulation and liver injury. Corosolic acid (CA) has proven lipid-lowering and hepatoprotective effects, yet the underlying mechanism by which CA mitigates MASLD remains unclear. In this study, mice were fed a high-fat diet for 8 weeks to induce MASLD, followed by 8 weeks of CA intervention. We found that CA significantly suppressed weight gain, reduced serum lipid levels, and improved liver function in the HFD-fed mice. Fecal metabolomic analysis showed that CA regulated multiple metabolic pa

Mechanism of swertiamarin and novel nitrogen-containing metabolites (R)-Gentiandiol and (S)-Gentiandiol in treating non-alcoholic fatty liver disease in rats: an untargeted metabolomics study based on UPLC-Q-TOF/MS.

Swertiamarin, a predominant iridoid glycoside from hepatoprotective Swertia herbs, is biotransformed in vivo into nitrogen-containing metabolites (R)-gentiandiol and (S)-gentiandiol. These metabolites may be the real active hepatoprotective agents. A high-fat diet-fed rat model was treated for 12 weeks with swertiamarin, (R)-gentiandiol, (S)-gentiandiol and silybin, and the therapeutic effects on non-alcoholic fatty liver disease (NAFLD) were systematically evaluated through biochemical indices and histopathological observations. Swertiamarin and (R)-gentiandiol reversed high-fat diet-induced

Analysis by transcriptomics and metabolomics for the proliferation inhibition and dysfunction through redox imbalance-mediated DNA damage response and ferroptosis in male reproduction of mice and TM4 Sertoli cells exposed to PM2.5.

Sertoli cells play a pivotal role in the complex spermatogenesis process. This study aimed to investigate the effects of PM2.5 on Sertoli cells using the TM4 cell line and a real time whole-body PM2.5 exposure mouse model, and further explore the underlying mechanisms through the application of metabolomics and transcriptomics. The results in vivo and in vitro showed that PM2.5 reduced Sertoli cells number in seminiferous tubules and inhibited cell proliferation. PM2.5 exposure also induced Sertoli cell dysfunction by increasing androgen binding protein (ABP) concentration, reducing the blood-

Relationship between neonatal glutamine supplementation and amino acid profiles, protein synthesis in splanchnic tissues and the plasma metabolome in 5-day-old piglets with low birth weight.

The incidence of low birthweight (LBiW) piglets, which show impaired growth compared to normal birthweight (NBiW) littermates, has increased with litter size. We previously reported that glutamine increased the bodyweight and plasma glutamine concentrations in LBiW piglets, with no effect on jejunal glutamine concentrations or glutamine transport and metabolism mRNA abundance. In this study, we investigated the effect of glutamine supplementation on proteinogenic free amino acid (FAA), protein-bound AA (PBAA) and AA-metabolite profiles, mRNA abundance, fractional protein synthesis (FPSR) and t

Comprehensive exploration and comparison of chemical constituents of Paris polyphylla var. chinensis and Paris polyphylla var. yunnanensis through two-dimensional chromatography, metabolomic analysis and furostanosides quantitation.

Paris polyphylla var. chinensis (PPC) and Paris polyphylla var. yunnanensis (PPY) were recognized as two legal species of Paridis Rhizoma in the Chinese Pharmacopoeia, exhibiting distinct chemical profiles primarily in terms of spirostanosides content. However, the chemical differences of other types of steroidal saponins remained unclear, comprehensive component comparison is lacking. In this research, an offline two-dimensional separation system (comprising HILIC and C18) was established to thoroughly explore and compare the chemical components between PPC and PPY. Additionally, a nontargete

Intrauterine growth restriction, defined by an elevated brain-to-liver weight ratio, affects faecal microbiota composition and, to a lesser extent, plasma metabolome profile at different ages in pigs.

BACKGROUND: Intrauterine growth restriction (IUGR) affects up to 30% of piglets in a litter. Piglets exposed to IUGR prioritize brain development during gestation, resulting in a higher brain-to-liver weight ratio (BrW/LW) at birth. IUGR is associated with increased mortality, compromised metabolism, and gut health. However, the dynamic metabolic and microbial shifts in IUGR-affected pigs remain poorly understood. This study aimed to investigate the longitudinal effects of IUGR, defined by a high BrW/LW, on the composition of faecal microbiota and plasma metabolome in pigs from birth to slaugh
