A Novel Chitosan-Gelatin Scaffold and Cell Spray Therapy for Treating Limbal Stem Cell Deficiency.

Limbal stem cell deficiency (LSCD) is a severe ocular condition that impairs vision and causes chronic pain. Current therapeutic approaches, such as stem cell transplantation and biomaterial scaffolds, are under continuous refinement. This study evaluated a novel treatment strategy combining a biocompatible and biodegradable chitosan-gelatin (CS-Gel) electrospun scaffold with a topical limbal stem cell (LSC) spray for treating alkaline-induced LSCD in a rabbit model. The scaffold was fabricated via electrospinning, stabilized with water vapor, and characterized using SEM, ATR-FTIR, contact ang

Tissue engineering for conjunctival reconstruction: established methods and future outlooks.

Reconstruction of the conjunctiva is an essential part of ocular surface regeneration, especially if an extensive area or the whole ocular surface is affected, such as in patients with ocular cicatricial pemphigoid, Stevens-Johnson syndrome, toxic epidermal necrolysis, or chemical/thermal burns. In these situations, corneal reconstruction almost inevitably fails unless the conjunctival surface is first repaired and a deep fornix is restored. The growing field of tissue engineering and advances in stem cell research offer promising new alternatives for these challenges. This article reviews the

Repair of a full thickness corneoscleral defect in a German shepherd dog using porcine small intestinal submucosa.

Porcine small intestinal submucosa (SIS) was used, in conjunction with a conjunctival graft, to repair a full thickness corneoscleral defect resulting from the excision of a limbal melanoma in a German shepherd dog. The SIS was found to provide adequate mechanical support and to act as a suitable physical barrier in place of the excised cornea and sclera. Corneal vascularisation was present distant to the graft by two weeks postoperatively but this was effectively controlled with topical cyclosporin. By six weeks postoperatively, the graft had become incorporated into the cornea and sclera, an
