Single-cell profiling reveals clonal CD8+T cell-derived IFNγ drives Sjögren's disease-related dacryoadenitis.

BACKGROUND: Sjögren's disease (SjD) is a chronic progressive autoimmune disorder primarily affecting the exocrine glands. Dacryoadenitis represents one of the typical clinical manifestations of SjD, yet its underlying pathogenic mechanisms remain poorly understood. This study investigated the immune microenvironment of the lacrimal gland to elucidate the pathogenic cell subset and key molecular factors driving SjD-related dacryoadenitis. METHODS: Aire-/- mice, a well-established SjD-related dacryoadenitis model, were employed in this study. Lacrimal gland infiltrating immune cells were profil

Alternative BCG delivery strategies improve protection against Mycobacterium tuberculosis in non-human primates: Protection associated with mycobacterial antigen-specific CD4 effector memory T-cell populations.

Intradermal (ID) BCG injection provides incomplete protection against TB in humans and experimental models. Alternative BCG vaccination strategies may improve protection in model species, including rhesus macaques. This study compares the immunogenicity and efficacy of BCG administered by ID and intravenous (IV) injection, or as an intratracheal mucosal boost (ID + IT), against aerosol challenge with Mycobacterium tuberculosis Erdman strain. Disease pathology was significantly reduced, and survival improved, by each BCG vaccination strategy, relative to unvaccinated animals. However, IV induce

[Immunological status of people inoculated with various types of inactivated vaccine against tick-borne encephalitis].

As compared with cellular immunity tests, serological tests more frequently detect virus-specific immune response in humans immunized with concentrated purified vaccine (CV) against tick-borne encephalitis. Combined determination of cell reactions and antibody production after a single injection of the vaccine increases the rate of detection of specifically responding persons. The duration of the inactivated vaccine-induced immunity is determined not only by the level of serum antibodies but also by the state of immunological memory detectable by the capacity of the host to produce antibodies

Characterization of a helper T lymphocyte defect in vitamin A-deficient mice.

Vitamin A-deficient mice exhibited sharply decreased IgG1 responses to protein Ag in vivo and in vitro. We traced the cellular basis for these impaired responses to a functional defect in A-deficient Th. Ag-presenting cells and B cells from A-deficient mice showed no functional defects. The A-deficient T cells proliferated normally in response to Ag but failed to provide the B cell stimulus for Ag-specific IgG1 responses. The A-deficient mice had fewer Th by limiting dilution analysis, and retinyl acetate supplementation in vitro restored Th frequency to control levels. Because added retinyl a

RD antigen based nanovaccine imparts long term protection by inducing memory response against experimental murine tuberculosis.

BACKGROUND: The absence of certain genomic loci that are present in most of the virulent strains of Mycobacterium tuberculosis as well as lack of lasting memory responses are some of the major causes attributed to the non effectiveness of Bacille Calmette-Gue'rin (BCG) vaccine. Immunization schedules addressing these issues can offer better strategy for protection against tuberculosis. METHODS: The immunological responses evoked upon administration of archaeosome based antigen delivery system comprising T cell antigen, Rv3619c (an ESAT-6 family protein), has been assessed against experimental

GATHeR: graph-based accurate tool for immunoglobulin heavy- and light-chain reconstruction.

Recovering full-length, paired B-cell receptor (BCR) sequences from scRNA-seq reads remains difficult, especially in naive and memory B cells where immunoglobulin transcripts are sparse. Incomplete constant-region coverage in current methods limits isoform, subclass, and allele resolution. Here we present GATHeR, an open-source tool that assembles and annotates paired heavy- and light-chain BCR sequences and extends assembled sequences into constant regions. This enables confident subclass and allele assignment and recovery of membrane-bound isoforms, including the transmembrane segment and cy

Identifying and validating immunological biomarkers in obstructive sleep apnea through bioinformatics analysis.

Obstructive sleep apnea (OSA) is a prevalent sleep disorder characterized by disrupted breathing patterns and dysfunctions in multiple organ systems. Although studies support a close correlation between OSA and immune function, the broader implications and specific manifestations remain unclear. Therefore, it is pressingly needed to identify potential immune-related markers and elucidate underlying immunological mechanisms of OSA. OSA-related datasets (GSE38792) and immune-related genes were downloaded from the GEO and ImmPort databases and intersected to obtain differentially expressed immune

Sequential Immunization With Heterologous Viruses Does Not Result in Attrition of the B Cell Memory in Rainbow Trout.

Long-term immunity is of great importance for protection against pathogens and has been extensively studied in mammals. Successive heterologous infections can affect the maintenance of immune memory, inducing attrition of T memory cells and diminishing B cell mediated protection. In fish, the basis of immune memory and the mechanisms of immunization to heterologous pathogens remain poorly understood. We sequentially immunized isogenic rainbow trout with two immunologically distinct viruses, VHSV and IPNV, either with one virus only or in combination, and analyzed the antibody responses and rep

Vitamin A status and immunoglobulin G subclasses in rats immunized with tetanus toxoid.

Vitamin A deficiency severely compromises the magnitude of the primary and secondary antibody response to tetanus toxoid (TT) but does not impair the development of immunologic memory. To further characterize this immunodeficiency in antibody production, we have quantified the immunoglobulin G (IgG) subclasses (IgG1, IgG2a, IgG2b, and IgG2c) during the primary and secondary response to TT in normal, vitamin A-deficient, and retinol-repleted rats. In the primary response in normal rats, anti-TT IgG1 and IgG2b predominated. In vitamin A-deficient rats the production of anti-TT IgG2b was severely

Distribution and phenotype of immune cells in normal human gingiva: active immune response versus unresponsiveness.

The oral cavity, and particularly the gingival mucosa, is continuously exposed to numerous food and bacterial plaque antigens, though evident immunologic reactions are uncommon. It is therefore possible that the mucosal associated lymphoid tissue (MALT) of this region is preferentially biased towards unresponsiveness, rather than immune cell activation. The distribution and phenotype of immune cells in normal human gingiva were examined. Their distribution varied, and high and low cellularity areas could be distinguished in the same specimen. The number of CD3 positive (CD3+) T lymphocytes was

Comprehensive and advanced T cell cluster analysis for discriminating seropositive and seronegative rheumatoid arthritis.

OBJECTIVE: Rheumatoid arthritis (RA) is classified into seropositive (SP-RA) and seronegative (SN-RA) types, reflecting distinct immunological profiles. This study aimed to identify the T cell phenotypes associated with each type, thereby enhancing our understanding of their unique pathophysiological mechanisms. METHODS: We analyzed peripheral blood T cells from 50 participants, including 16 patients with untreated SP-RA, 17 patients with SN-RA, and 17 healthy controls, utilizing 25 T cell markers. For initial analysis, a dataset was established through manual T cell subset gating analysis. F

Revealing the immune landscape of menstrual blood: unlocking insights into activation, exhaustion, and mitochondrial mass for reproductive health.

Reproductive disorders such as endometriosis and polycystic ovary syndrome (PCOS) are increasingly recognized as immune-mediated conditions, yet their immunopathology remains poorly understood. Menstrual blood, a noninvasive and biologically relevant sample, offers a unique window into reproductive tract immunity but has been underutilized in this context. We optimized Cytek's® 25-color high-dimensional flow cytometry panel by incorporating a mitochondrial dye to investigate immune cell profiles in menstrual mononuclear cells (MMCs) from healthy individuals, and those with endometriosis or PCO

Distinct in vivo dynamics of donor-derived stem cell memory CAR T cells post-allogeneic HSCT relapse.

Donor-derived CD19-CAR T cells offer a therapeutic option for B cell malignancies relapsing after allogeneic hematopoietic stem cell transplantation but are often constrained by poor engraftment, expansion, and persistence. In a first-in-human study (NCT01087294), we found that CAR-modified stem-cell memory T (TSCM) cells exhibited greater expansion and persistence than standard CAR T cells, enabling complete responses at low doses in the absence of lymphodepletion. CAR TSCM cells induced mild cytokine-release syndrome, dominated by IFN-γ. Both products differentiated into effectors; however,

Human hypofunctional NCF1 variant aggravates salivary gland immunopathology in Sjögren's disease by promoting switched memory B-cell recruitment and long-lived plasma cell differentiation.

OBJECTIVES: We assessed the role of a systemic lupus erythematosus causal, hypofunctional variant, neutrophil cytosolic factor 1 (NCF1)-p.Arg90His (p.R90H) substitution, in Sjögren's disease (SjD). METHODS: Association between NCF1-H90 and SjD was assessed in case-control cohorts. Peripheral blood mononuclear cells (PBMCs) and minor salivary gland (MSG) from patients with Sjögren's Syndrome type A antigen (SSA)+ SjD were assessed using cytometry by time-of-flight and single-cell RNA sequencing, respectively. RESULTS: The NCF1-H90 allele was associated with increased risk for SjD in Chinese a

Circulating memory B-cell receptor repertoire analysis identifies novel candidate antibodies against metastatic melanoma in immunotherapy-responsive patients.

BACKGROUND: By examining the B-cell receptor (BCR) repertoire of metastatic melanoma (MM) patients with favorable treatment outcomes, it is now possible to identify unique patterns of immune responses, with the potential of discovering novel antitumor antibodies. METHODS: Here, we isolated CD27-positive circulating memory B cells from non responders, partial responders, and complete responders MM patients to first-line therapy with anti-PD-1 immune checkpoint inhibitor (ICI) nivolumab, to perform a BCR repertoire sequencing analysis. We looked for complementarity-determining region 3 (CDR3) s

Anti-cancer immune priming with β-radioligand therapy using a novel high affinity antibody selectively targeting the 4Ig-Isoform of B7-H3.

RATIONALE: Radioligand therapy (RLT) is an emerging oncologic strategy that uses molecularly targeted therapeutic radioisotopes to reduce tumor burden and improve survival in patients with advanced cancers. Expanding RLT to new targets and understanding its systemic immune effects could enhance its clinical impact. B7-H3 (CD276) is an attractive target due to its high differential expression in solid tumors compared to normal tissues. However, the presence of two isoforms, 4Ig-B7-H3 (tumor-associated) and 2Ig-B7-H3 (soluble decoy), poses challenges for selective targeting, especially in the co

Vaccination against rapidly evolving pathogens and the entanglements of memory.

Immune memory determines infection risk and responses to future infections and vaccinations over potentially decades of life. Despite its centrality, the dynamics of memory to antigenically variable pathogens remains poorly understood. This Review examines how past exposures shape B cell responses to vaccinations with influenza and SARS-CoV-2. An overriding feature of vaccinations with these pathogens is the recall of primary responses, often termed 'imprinting' or 'original antigenic sin'. These recalled responses can inhibit the generation of new responses unless some incompletely defined co

Staphylococcus aureus specific lung resident memory CD4+ Th1 cells attenuate the severity of influenza virus induced secondary bacterial pneumonia.

Staphylococcus aureus is a major cause of severe pulmonary infections. The evolution of multi-drug resistant strains limits antibiotic treatment options. To date, all candidate vaccines tested have failed, highlighting the need for an increased understanding of the immunological requirements for effective S. aureus immunity. Using an S. aureus strain engineered to express a trackable CD4+ T cell epitope and a murine model of S. aureus pneumonia, we show strategies that lodge Th1 polarised bacterium specific CD4+ tissue resident memory T cells (Trm) in the lung can significantly attenuate the s

Clonotypic structure of the human CD4+ memory T cell response to cytomegalovirus.

High steady-state frequencies of CMV-specific CD4(+) memory T cells are maintained in CMV-exposed subjects, and these cells are thought to play a key role in the immunologic control of this permanent infection. However, the essential components of this response are poorly defined. Here, we report the use of a step-wise application of flow cytometric and molecular techniques to determine the number and size of the TCR Vbeta-defined clonotypes within freshly obtained CMV-specific CD4(+) memory T cell populations of four healthy, CMV-exposed human subjects. This analysis revealed a stable clonoty
