Effect of extracellular hyperosmolality on sweat rate during metaboreflex activation in passively heated young men.

Metaboreflex activation augments sweating during mild-to-moderate hyperthermia in euhydrated (isosmotic isovolemic) individuals. Recent work indicates that extracellular hyperosmolality may augment metaboreflex-mediated elevations in sympathetic nervous activity. Our primary objective was, therefore, to test the hypothesis that extracellular hyperosmolality would exacerbate metaboreflex-mediated increases in sweat rate. On two separate occasions, 12 young men [means (SD): 25 (5) yr] received a 90-min intravenous infusion of either 0.9% saline (isosmotic condition, ISO) or 3.0% saline (hyperosm…

Integrated Transcriptomic and Proteomic Analysis of Human Eccrine Sweat Glands Identifies Missing and Novel Proteins.

The eccrine sweat gland is an exocrine gland that is involved in the secretion of sweat for control of temperature. Malfunction of the sweat glands can result in disorders such as miliaria, hyperhidrosis and bromhidrosis. Understanding the transcriptome and proteome of sweat glands is important for understanding their physiology and role in diseases. However, no systematic transcriptome or proteome analysis of sweat glands has yet been reported. Here, we isolated eccrine sweat glands from human skin by microdissection and performed RNA-seq and proteome analysis. In total, ∼138,000 transcripts …

The Sweat Metabolome of Screen-Positive Cystic Fibrosis Infants: Revealing Mechanisms beyond Impaired Chloride Transport.

The sweat chloride test remains the gold standard for confirmatory diagnosis of cystic fibrosis (CF) in support of universal newborn screening programs. However, it provides ambiguous results for intermediate sweat chloride cases while not reflecting disease progression when classifying the complex CF disease spectrum given the pleiotropic effects of gene modifiers and environment. Herein we report the first characterization of the sweat metabolome from screen-positive CF infants and identify metabolites associated with disease status that complement sweat chloride testing. Pilocarpine-stimula…

Immunohistological Expression of p16INK4a is Commonly Present Both in Benign and Malignant Sweat Gland Neoplasias.

The expression of p16INK4a has been reported to be a significant marker for malignant transformation of epidermal tumors. However, little is known about sweat gland tumors. We examined the immunohistological expression of p16INK4a in benign and malignant sweat gland tumors. The ductal and acrosyringial portion of normal eccrine glands were positively stained with p16INK4a while it was negative in the normal epidermis. Moderate to strong expression of p16INK4a was found in 16 of 17 eccrine poromas, 4 of 5 hidradenomas, 3 of 3 syringocystadenoma papilliferums, 2 of 2 mixed tumors, and 3 of 3 syr…

Managing a Rare Malignant Sweat Gland Tumor Invading the Brain: Case Report and Literature Review.

BACKGROUND: Malignant sweat gland adnexal tumors are rare with an incidence of 0.001%. Of these, clear cell hidradenocarcinoma is an extremely uncommon subtype that accounts for 6% of malignant eccrine sweat gland tumors. They occur commonly in the head, neck, and extremities. Although they have a propensity for local recurrence, intracranial extension with brain invasion is extremely rare. CASE DESCRIPTION: We report a 76-year-old man with a large, recurring, ulcerated, fungating scalp swelling of 14 years who presented with focal seizures and drowsiness. Neuroimaging revealed a massive tumo…

A manufactu(RED) ethics: labor, HIV, and the body in Lesotho's "sweat-free" garment industry.

Employing mostly women and producing for major U.S. labels, Lesotho's primarily foreign-owned garment industry undertook efforts to become "sweat-free" in 2006; simultaneously, it also began producing for the Product(RED) campaign. This article explores the parameters and ethical challenges of an industry-wide, public-private partnership providing HIV prevention and treatment services in this industry. Here, HIV services are intimately bound up in emerging patterns of humanitarian consumption and the production of an ethical industry. Within this ethical production zone, all is not what it see…

Apocrine sweat gland carcinoma: initial evaluation, staging, and response monitoring using 18F-FDG PET/CT.

Primary apocrine sweat gland carcinomas (PASGCs) are rare tumors, commonly located in the axilla. Metastases are common and confer poor prognosis. Given the rarity of these tumors, there is limited knowledge regarding its diagnosis and management. Here we show 18F-FDG PET/CT images of a 61-year-old man with PASGCs of the left axilla. PET/CT confirmed the diagnosis as primary axillary malignancy with nodal, pulmonary, and skeletal metastases. Another interesting finding in this case was the presence of FDG-avid calcified metastatic lymph nodes during the initial evaluation. Follow-up PET/CT sho…

Diagnostic utility of direct immunofluorescence findings around hair follicles and sweat glands in immunobullous disease.

BACKGROUND: Inherent to some immunobullous disorders is potential for intraepidermal or dermal-epidermal junction fragility, a phenomenon that may compromise biopsy specimen integrity and direct immunofluorescence (DIF) interpretation. In these situations, cutaneous adnexal structures (e.g. hair follicles, sweat apparatus) usually remain intact. Whether periadnexal DIF findings are reliable in diagnosing immunobullous conditions is unknown. METHODS: We evaluated 56 cutaneous specimens with diagnostic immunoglobulin (Ig) deposition patterns that contained adnexal structures. In a corollary stu…

Dissolution of cemented carbide powders in artificial sweat: implications for cobalt sensitization and contact dermatitis.

Skin exposure to cobalt-containing materials can cause systemic immune sensitization and upon repeat contact, elicitation of allergic contact dermatitis (ACD). Data on cobalt dissolution rates are needed to calculate uptake through skin and for development of models to understand risk of sensitization or dermatitis. The purpose of this research was to measure the dissolution kinetics of feedstock and process-sampled powders encountered in the production of hard metal alloys using artificial sweat. The physicochemical properties of each material were characterized prior to evaluation of dissolu…

Determination of zinc diethyldithiocarbamate released into artificial sweat from natural rubber latex vulcanizates by HPLC.

A simple high-performance liquid chromatography method is adopted in order to quantitate the amount of zinc diethyldithiocarbamate (ZDEC) released into artificial sweat from natural rubber latex vulcanizates. The artificial sweat is extracted with dichloromethane, and the residue is recovered and re-dissolved in a known quantity of dichloromethane. ZDEC is quantitated as its copper complex by reacting with copper(II) sulphate. A reversed-phase C18 column and detection wavelength of 435 nm are used to measure the copper-dithiocarbamate complex. The procedure is repeated with cobalt(II) chloride…

Glycoconjugates in sweat glands and other structures of skin from normal and cystic fibrosis subjects.

In this study, glycoconjugates of human skin varied in structure between cell types in an individual and often between individuals for a given cell type. Stored secretory material in dark cells of the sweat gland coil contained complex carbohydrate with terminal alpha-galactose, N-acetylgalactosamine, fucose, or sialic acid. The plasmalemma of clear cells in the secretory coil stained conspicuously for terminal alpha-N-acetylgalactosamine, and the cytosol of clear cells contained lectin-reactive glycogen in the majority of specimens. Superficial and deep cells of the sweat duct evidenced plasm…

Engrailed-1 in eccrine sweat gland development and regeneration: from evolutionary innovation to therapeutic perspectives.

Functional regeneration of eccrine sweat glands (ESGs) remains a major unmet need in burns and trauma, where extensive skin loss leads to permanent anhidrosis, life-threatening hyperthermia, and impaired quality of life. Here we synthesize evidence pointing to the homeodomain transcription factor Engrailed-1 (EN1) as a central regulator that coordinates ESG development across four interconnected biological functions: (1) establishing a permissive ventral limb field by repressing the WNT7A-LMX1B dorsalization pathway; (2) executing a binary fate switch that suppresses the hair follicle program …

An intelligent electronic-textile system with adaptive sweat-regulation for continuous exercise-induced fatigue monitoring.

Characterizing exercise-induced fatigue is challenging due to difficulties in monitoring biochemical and electrophysiological signals consistently. While wearable sensors able to monitor both signals are available, they struggle to maintain stable signal acquisition across exercise-rest scenarios because of significant sweat variation. Here we describe the development of a system, FatigueVisual, combining an adaptive sweat-regulating hybrid electronic textile (e-ASRHT) patch with artificial intelligence (AI) analytics to improve fatigue management. The key innovation lies in the precise regula…

Interlaced 2D cellulose networks with molecular enrichment capability for sensitive SERS detection of sweat biomarkers.

Efficient localization of analytes near plasmonic hotspots remains a major challenge for surface-enhanced Raman scattering (SERS) detection in complex liquid-phase systems. Herein, an interlaced cellulose-based porous membrane composed of dissolving pulp fibers (DPFs), mechanically ground nanofibers (MGNFs), sodium alginate (SA), and Ag nanoparticles was developed as a molecular enrichment-assisted SERS platform for sweat biomarker determination. The hierarchical porous architecture provided interconnected transport channels and abundant interfacial adsorption sites, facilitating analyte reten…

Proteomic Profiling of Human Sweat Reveals Molecular Adaptations to Heat Acclimatization Induced by Isothermic Conditioning.

INTRODUCTION: Prolonged physical exertion in hot and/or humid environments can induce physiological strain and increase the risk of heat exhaustion or exertional heat stroke (EHS). Heat acclimatization (HA) optimizes an individual's ability to tolerate heat and is therefore widely adopted to improve exercise performance in thermally challenging environments. Sweat is a key thermoregulatory indicator reflective of physiological adaptations during HA and hence is regarded as a valuable resource for monitoring HA. However, knowledge of molecular-level changes and adaptations in sweat associated w…

Optimized starch-iodine method for quantification of sweat volume and pore activation in diabetic neuropathy.

Early detection of Diabetic Neuropathy (DN) relies on quantifying Sweat Glands (SGs) function through non-invasive measures of sweat volume and pore activation. Gravimetry, the most common method, measures sweat mass but cannot assess pore activation and is limited by inter- and intra-patient variability. To address this, an optimized starch-iodine test with a stamper-like mechanism was developed to simultaneously quantify sweat volume and pore activation. An algorithm incorporating Structural Similarity Index Measure (SSIM) and its related correction on estimated volume was employed to enhanc…

Remote exercise-induced sweat chloride measurements using a wearable microfluidic sticker in cystic fibrosis patients.

Sweat parameters such as volume and chloride concentration may offer invaluable clinical insights for people with CF (PwCF). Pilocarpine-induced sweat collection for chloridometry measurement is the gold-standard for sweat chloride, but this technique is cumbersome and not suitable for remote settings. We have previously reported the utility of a skin-interfaced microfluidic device (CF Patch) in conjunction with a smartphone image processing platform that enables real-time measurement of sweating rates and sodium chloride loss in laboratory and remote settings. Here we conducted clinical studi…

TiO₂/PANI-based sensing electrode for ethanol detection in human sweat to monitor liver functioning.

Wearable sensors for detecting ethanol volatile organic compounds (VOCs) in sweat offer a non-invasive approach to monitor liver health. In line with this goal, a titanium dioxide/polyaniline (TiO2/PANI) nanocomposite-based sensor electrode was developed for real-time ethanol detection under standard conditions (300 K, 1 atm). The thin film, deposited on a conductive fabric, was characterized for phase, structure, and morphology. Electrochemical tests showed high sensitivity, with a detection limit of 23.62 ppb and quantification of 71.59 ppb. The sensor demonstrated excellent reproducibility,…
