[The positive chronotropic effects of salsolinol on the isolated rat heart].

Salsolinol (6,7-dioxy-1-methyl-1,2,3,4-tetrahydroisochinoline), a product of nonenzymatic condensation of dopamine and acetaldehyde, produced a dose-dependent positive chronotropic effect on the rat isolated perfused heart: EC = 5.6 X 10(-6) M. Inderal, a beta-adrenoreceptor antagonist (10(-5) g/ml), blocked and naloxone, an opioid receptor antagonist (3.1 X 10(-5) M), potentiated this effect. Apart from tachycardia, salsolinol induced arrhythmias which were inhibited by naloxone. Salsolinol seems to play an important role in regulating the heart performance.

[Alcoholic myopathy].

Six cases of alcoholic myopathy (two acute and four chronic) are reported. Histologically, cases of acute alcoholic myopathy showed dystrophic changes of the sarcoplasm (especially in the muscles of lower extremities), ending with total rhabdomyolysis. In chronic cases, histology revealed selective atrophy of muscle fibers of the 2nd type. Both direct and indirect effects of alcohol are discussed in the pathogenesis as well as the metabolic disturbances caused by acetaldehyde, a metabolic product of ethanol. Possible predisposition factors are taken into consideration and the differential diag…

New method for reducing the binding power of sweet white wines.

Sulfur dioxide is now considered to be a toxic chemical by most world health authorities. However, it remains an irreplaceable additive in e … ain carbonyl compounds responsible for the SO 2 binding power of sweet wines were removed, that is, acetaldehyde, pyruvic acid, 2-oxoglutaric acid, and 5-oxofructose. The method retained was selective liquid-solid removal, using phenylsulfonylhydrazine as a scavenging agent. The scavenging function was grafted on different classes of porous polymer supports, a …

Reactions of the HCN-tetramer with aldehydes.

The HCN-tetramer, a 'classic' of the prebiotic chemistry of HCN, is shown to undergo a remarkable reaction with acetaldehyde in slightly basic or neutral aqueous solution at room temperature. The reaction consists in an aldolization-type C,C-bond formation, accompanied by a (presumably aldehyde-catalyzed) hydration of one of the two nitrile groups and the formation of two cyclic aminal-type groupings, each of the latter incorporating an additional molecule of the aldehyde. Should this so far unexplored type of chemistry of the HCN-tetramer prove to have some generality, the finding might add a…

Optimization of solid-phase microextraction conditions for gas chromatographic determination of ethanol and other volatile compounds in blood.

A procedure for the determination of acetaldehyde, acetone, methanol, ethanol, 1-propanol and 2-propanol in blood was developed. Separation of analytes was carried out on DB-wax capillary column (l = 30 m, I.D. = 0.32 mm, dF = 0.5 microm) at 40 degrees C, hydrogen was used as a carrier gas (at 30 kPa) and FID as a detector. Quantification was performed with the use of 2-butanol as an internal standard. Headspace solid-phase microextraction was applied as the sample preparation technique. The usefulness of most commercially available fiber coatings was checked and 65 microm Carbowax/DVB proved …

Immunologic abnormalities in alcoholic hepatitis.

There are many unanswered questions in the possible autoimmune pathogenesis of alcoholic liver disease. Why do only 15-20% of chronic alcoholic patients develop alcoholic hepatitis and cirrhosis? What factors make a patient susceptible to immune events in alcoholism? What initiates immune reactivity in susceptible patients? What permits accumulation of alcoholic hyalin in patients with alcoholic hepatitis? Is alcoholic hyalin central to the pathogenesis of alcoholic hepatitis? If adducts play a role, why don't more alcoholics develop liver disease, as adducts probably form in all patients? Unf

Disulfiram-containing polymeric nanocapsules with anticancer activity for cancer treatment.

Disulfiram, a medication traditionally used to treat alcohol addiction, has gained attention as a potential cancer treatment in recent years. Disulfiram works by inhibiting the enzyme aldehyde dehydrogenase, involved in the breakdown of acetaldehyde, a by-product of alcohol metabolism. This results in the build up of acetaldehyde in the body leading to unpleasant side effects such as nausea and vomiting when alcohol is consumed while taking the drug. With cancer treatment, disulfiram has been found to have several mechanisms of action. It has been shown to inhibit cancer cell growth and metast…

Density functional theory study of the mechanisms of oxidation of ethylene by chromyl chloride.

The mechanistic pathways for the formation of epoxide, 1,2-dichloroethane, 1,2-chlorohydrin, acetaldehyde, and vinyl alcohol precursors in the oxidation of ethylene by chromyl chloride has been studied using hybrid density functional theory at the B3LYP/LACVP* level of theory. The formation of the epoxide precursor (Cl(2)(O)Cr-OC(2)H(4)) was found to take place via initial [2 + 2] addition of ethylene across the Cr=O bonds of CrO(2)Cl(2) to form a chromaoxetane intermediate. The pathway involving initial [3 + 2] addition of ethylene to the oxygen and chlorine atoms of CrO(2)Cl(2), which has no…

Decreased in vivo rate of ethanol metabolism in the suckLing rat.

Blood concentrations of ethanol and acetaldehyde were determined in suckling rats after a single oral ethanol gavage. These results were compared with the hepatic activities of alcohol and aldehyde dehydrogenase. After intragastric administration of 3 g/kg body weight of ethanol, ethanol concentrations were much higher in suckling rats than in adult animals, especially at 90, 120, and 180 min after its administration. In addition, acetaldehyde concentrations were undetectable in suckling rats as opposed to adult rats, in whom micromolar concentrations were detected. Thus, 5- to 30-day-old rats…

Vanillin production using Escherichia coli cells over-expressing isoeugenol monooxygenase of Pseudomonas putida.

The isoeugenol monooxygenase gene of Pseudomonas putida IE27 was inserted into an expression vector, pET21a, under the control of the T7 promoter. The recombinant plasmid was introduced into Escherichia coli BL21(DE3) cells, containing no vanillin-degrading activity. The transformed E. coli BL21(DE3) cells produced 28.3 g vanillin/l from 230 mM isoeugenol, with a molar conversion yield of 81% at 20 degrees C after 6 h. In the reaction system, no accumulation of undesired by-products, such as vanillic acid or acetaldehyde, was observed.

Effect of tremorogenic and antitumor agents on enzymes regulating a minor metabolic pathway of biogenic amines: an hypothesis for the underlying mechanism.

The effects of some tremor-producing and antitremor agents on the enzymes sequentially metabolizing ethanol and acetaldehyde in addition to biogenic-amine-derived aldehyde intermediates were studied in mouse liver preparations. Aldehyde but not alcohol dehydrogenase was differentially stimulated by the compounds studied in vitro. This effect was confined to the mitochondrial enzyme and was isoenzyme-dependent. The results indicate difference between in vitro and in vivo effects and suggest a role for mitochondrial aldehyde dehydrogenase in metabolic detoxification of endogenous biogenic aldehy…

A study of cyanide-insensitive respiration in the genus Dekkera and Brettanomyces.

Dekkera intermedia and Brettanomyces custersii were shown to have a respiratory pathway resistant to cyanide, antimycin A, and azide. This respiration remained sensitive to salicylhydroxamic acid (SHAM). The "cyanide-resistant" respiration was induced mainly at the end of the growth phase and could reach 50% of total respiratory capacity. The mitochondrial "petite colony" mutation had no effect on this oxidation pathway. The presence of this respiration pathway in these strains constitutes a compensation mechanism for the reducing activity of acetaldehyde dehydrogenase. This alternate pathway …
