Optimization of citrulline production from a Bacillus subtilis BH-01 isolated from raw buffalo milk.

The main purpose of this study was to optimize the L-citrulline production process using Plackett-Burman and Box-Behnken designs. L-citrulline-producing bacterium BH-01 was isolated from raw buffalo milk. The isolate was tested for probiotic activities such as tolerance to simulated gastric and intestinal juices, antagonistic activity against six antibiotic-resistant bacteria, and temperature tolerance. L-citrulline production and arginine deiminase (ADI) activity were optimized using statistical designs. The bacterial isolate was molecularly identified as Bacillus subtilis strain AUMC B-498 (

Melatonin and hemin co-application reprograms lipolysis, oleosin accumulation and H2S metabolism in NaCl-stressed sunflower seedlings.

Salt stress adversely affects seed germination and lipid mobilization in germinating sunflower seedlings. Notably, recovery in the rate of lipolysis is crucial to promote stress tolerance, thereby imparting longevity to the seedlings. Melatonin has been previously reported to regulate lipolysis in NaCl-stressed sunflower seedling cotyledons. While hemin (oxidized form of heme) is known to function as a metabolic regulator in plants, no substantial reports are available to understand the combined impact of melatonin and hemin application in the regulation of lipid mobilization during seed germi

Integrated Multi-Omics Exploration of the Molecular Mechanisms Underlying Differential Responses to Saline-Alkaline Stress in Leaves of Populus simonii × P. nigra.

Populus simonii × P. nigra is a valuable model for studying woody plant responses to saline-alkaline stress, however the molecular mechanisms underlying its adaptation remain poorly understood. Here, integrated transcriptomic and metabolomic analyses were used to examine the responses in leaves to low (50 mmol/L) and high (200 mmol/L) alkaline salt stress over 7 and 14 days. Physiological assessments revealed that high-concentration stress inhibited growth, induced oxidative damage, elevated malondialdehyde (MDA) content, enhanced antioxidant enzyme activity (SOD, POD, CAT), and promoted osmor

Comprehensive Profiling of the Rice OsEPF/EPFL Gene Family Under Biotic and Abiotic Stresses Reveals the Involvement of OsEPF2 in Disease Resistance.

Members of the EPIDERMAL PATTERNING FACTOR (EPF) and EPF-Like (EPFL) families perform diverse regulatory functions in plant tissue morphogenesis, controlling the development of stomata, awns, shoot apical meristems (SAMs), and inflorescences. Nevertheless, the biological functions of OsEPF/EPFL family members in mediating responses to biotic/abiotic stresses are not yet widely characterized. Here, we demonstrated abundant cis-acting elements in the putative promoters of OsEPF/EPFL genes, including those associated with dehydration-, MeJA-, MYB binding site for drought, stress-, and ABA-respons

Characterization of three mitogen-activated protein kinase kinase-like proteins in Beauveria bassiana.

Pbs2, Mkk1 and Ste7 orthologs are three mitogen-activated protein kinase (MAPK) kinases (MAPKKs) acting as checkpoints of the Hog1, Slt2 and Fus3 MAPK cascades that constitute major parts of fungal signaling network. Here, we show that three other MAPKK-like proteins (Mkk4/5/6) exist in Beauveria bassiana and other entomopathogenic or non-entomopathogenic fungi but lack in yeasts and aspergilli, and elucidate how they function in the fungal insect pathogen. Based on phenotypic defects of single-, double- and triple-deletion mutants, Mkk4, Mkk5 and Mkk6 played collaborative or independent roles

Central antihypertensive effects of chronic treatment with RB150: an orally active aminopeptidase A inhibitor in deoxycorticosterone acetate-salt rats.

BACKGROUND AND OBJECTIVE: Hyperactivity of the brain renin-angiotensin (Ang) system has been implicated in the development and maintenance of hypertension. AngIII, one of the main effector peptides of the brain renin-Ang system, exerts a tonic stimulatory control over blood pressure (BP) in hypertensive rats. Aminopeptidase A (APA), the enzyme generating brain AngIII, represents a new therapeutic target for the treatment of hypertension. We developed RB150, a prodrug of the specific and selective APA inhibitor, EC33. When given orally in acute treatment in hypertensive rats, RB150 crosses the

Delineation of a bacterial starvation stress response network which can mediate antibiotic tolerance development.

This study aimed at elucidating the physiological basis of bacterial antibiotic tolerance. By use of a combined phenotypic and gene knockout approach, exogenous nutrient composition was identified as a crucial environmental factor which could mediate progressive development of tolerance with markedly varied drug specificity and sustainability. Deprivation of amino acids was a prerequisite for tolerance formation, conferring condition-specific phenotypes against inhibitors of cell wall synthesis and DNA replication (ampicillin and ofloxacin, respectively), according to the relative abundances o

Rapid and Scalable Synthesis of Oxazoles Directly from Carboxylic Acids.

A highly efficient and expedient method for the synthesis of 4,5-disubstituted oxazoles has been developed directly from carboxylic acids, employing a stoichiometric amount of the easy-to-access and stable triflylpyridinium reagent. The overall transformation proceeds through the formation of an in situ generated acylpyridinium salt followed by trapping with isocyanoacetates and tosylmethyl isocyanide. This transformation has a broad substrate scope with good functional group tolerance (including hindered and less reactive substrates or those containing sensitive functional groups). The versat

Salicylic Acid's Impact on Growth, Photosynthesis, and Antioxidant Enzyme Activity of Triticum aestivum When Exposed to Salt.

Recently, the application of salicylic acid (SA) for improving a plant's resistance to abiotic stresses has increased. A large part of the irrigated land (2.1% out of 19.5%) is severely affected by salinity stress worldwide. In 2020, total production of wheat (Triticum aestivum) was 761 million tons, representing the second most produced cereal after maize; therefore, research on its salinity tolerance is of world concern. Photosynthetic attributes such as net photosynthetic rate (PN), stomatal conductance (gs), intercellular CO2 concentration (Ci), and transpiration rate (E) were increased si

Characterization, evolution, and expression profiling of cotton DUF677 protein family and their potential role in drought and salt stress response.

BACKGROUND: The membrane-bound proteins belonging to DUF677 (domain of unknown function 677) are found mainly in green plants. The function of the DUF677 gene (AT14A) has been investigated in Arabidopsis and tomato in relation to drought stress tolerance. Overexpression of AT14A improves drought tolerance in tomato, promotes growth in Arabidopsis during drought stress, and confers tolerance against oxidative damage caused by drought stress in suspension-cultured A. thaliana. However, the role of the DUF677 gene family has not yet been reported in cotton. RESULTS: We identified 148 DUF677 gene

Investigating genetic differentiation between brackish and fresh water collections of the arboviral vector Aedes aegypti.

BACKGROUND: Aedes aegypti is typically regarded as a freshwater mosquito; however, recent studies have documented its development in brackish water habitats in coastal regions, including Sri Lanka's Jaffna Peninsula. Compared with freshwater populations, brackish water samples in Jaffna display enhanced salt tolerance throughout larval-to-adult development, along with differences in gene expression, cuticle morphology and composition, and insecticide susceptibility. METHODS: To explore the genetic basis of these differences, we performed a comparative genomic analysis using 5135 genome-wide s…

Kinetic analysis of high-concentration isopropanol biodegradation by a solvent-tolerant mixed microbial culture.

The ability of a previously enriched microbial population to utilize isopropanol (IPA) as the sole carbon source within a minimal salts medium is studied. The advantage of prior enrichment procedures for the improvement of IPA biodegradation performance is demonstrated for an IPA concentration of up to 24 g L(-1). Results showing the interrelationship between temperature and substrate utilization and inhibition levels at temperatures of between 2 degrees C and 45 degrees C are examined. Models of inhibition based on enzyme kinetics are assessed via nonlinear analysis, in order to accurately re

Design of Experiments for Diverse Carbon Isotope Labeling with Aryl Thianthrenium Salts.

A method for accessing structurally diverse carbon-isotope-labeled products from aryl thianthrenium salts has been established. The use of near-stoichiometric amounts of ex situ-generated carbon monoxide along with simple, air-stable palladium catalysts allows for the synthesis and isotope labeling of a broad range of carbonylation products. Design of Experiments enabled rapid optimization and gain of key mechanistic insight. The reaction tolerates electronically diverse electrophiles and could be used for the synthesis and late-stage isotope labeling of several pharmaceutical molecules.

C-5 selective chlorination of 8-aminoquinoline amides using dichloromethane.

An oxidant-free electrochemical regioselective chlorination of 8-aminoquinoline amides at ambient temperature in batch and continuous-flow was achieved. Inert DCM was used as the chlorinating reagent. Owing to the continuous-flow setup, the reaction scale up can be achieved conveniently with higher productivity. Moreover, this method has good position-control, and water and air tolerance. Costly quaternary ammonium salts were avoided. Radical-trapping, H/D exchange, KIE and cyclic voltammetry experiments were conducted to gain insight into the reaction mechanism.

Chemical structure and properties of midazolam compared with other benzodiazepines.

A short review is given of the basic chemical development in the field of 'classical' and 'annelated' benzodiazepines, distinguishing between pro-drugs and directly acting compounds. Some properties of midazolam that are of special interest for its practical use are discussed, such as: the basicity of its imidazole ring nitrogen, which allows water-soluble salts and well-tolerated aqueous injectable solutions to be prepared; its stability to hydrolytic degradation; its rapid metabolic inactivation, which is mainly determined by the methyl group on the imidazole ring, and which is much faster t

Modulation of nitric oxide mediated by phytoglobin1 plays a role in salinity tolerance via reduced nitro-oxidative stress in Arabidopsis.

Salinity is one of the major abiotic stresses that induces nitro-oxidative stress, which severely diminishes plant growth, development, and survival by altering various metabolic pathways. Phytoglobin (Pgb) is a nitric oxide (NO) scavenger that plays an important role in various stresses. However, the role of differential levels of phytoglobin1 in the regulation of salinity stress induced nitro-oxidative stress in plants is not known. Here we characterized the role of Pgb-mediated NO in salinity tolerance by regulation of nitro-oxidative stress using Pgb1 overexpressing (Pgb1-OE) and silencing
