Novel anisotropic engineered cardiac tissues: studies of electrical propagation.

The goal of this study was to engineer cardiac tissue constructs with uniformly anisotropic architecture, and to evaluate their electrical function using multi-site optical mapping of cell membrane potentials. Anisotropic polymer scaffolds made by leaching of aligned sucrose templates were seeded with neonatal rat cardiac cells and cultured in rotating bioreactors for 6-14 days. Cells aligned and interconnected inside the scaffolds and when stimulated by a point electrode, supported macroscopically continuous, anisotropic impulse propagation. By culture day 14, the ratio of conduction velociti

Production of extracellular matrix components in tissue-engineered blood vessels.

Morphology and compliance of tissue-engineered blood vessels (TEBV) are dependent on the culture period and production of extracellular matrix (ECM) components in order to increase the strength of the developing tissue. The aim of the present study was to evaluate the potential of TEBVs to produce an ECM similar to native arteries and veins. Human smooth muscle cells (SMC) were seeded onto the poly(glycolic acid) (PGA) scaffold and placed in bioreactors filled with DMEM supplemented with growth factors. After 6 weeks, the vessels were harvested from the bioreactors and seeded with human endoth

FT-IR imaging of native and tissue-engineered bone and cartilage.

Fourier transform infrared (FT-IR) imaging and microspectroscopy have been extensively applied to the analyses of tissues in health and disease. Spatially resolved mid-IR data has provided insights into molecular changes that occur in diseases of connective or collagen-based tissues, including, osteoporosis, osteogenesis imperfecta, osteopetrosis and pathologic calcifications. These techniques have also been used to probe chemical changes associated with load, disuse, and micro-damage in bone, and with degradation and repair in cartilage. This review summarizes the applications of FT-IR micros

Determination of oxygen gradients in engineered tissue using a fluorescent sensor.

Nutrient and oxygen supply of cells are crucial to tissue engineering in general. If a sufficient supply cannot be maintained, the development of the tissue will slow down or even fail completely. Previous studies on oxygen supply have focused on measurement of oxygen partial pressures (pO(2)) in culture media or described the use of invasive techniques with spatially limited resolution. The experimental setup described here allows for continuous, noninvasive, high-resolution pO(2) measurements over the cross-section of cultivated tissues. Applying a recently developed technique for time-resol…

[Clinical application of tissue-engineered autologous oral mucosa transplants].

BACKGROUND: Tissue engineering of autogenous oral mucosa is a significant enrichment for various indications in oral and maxillofacial surgery. Besides preprosthetic surgery and prelamination of microvascular fasciocutaneous flaps, interdisciplinary work such as reconstruction procedures in cases of hypospadias and epispadias are the main indications for tissue-engineered oral mucosa. RESULTS: One main advantage of this technique is the reduction of second surgical procedures. The morbidity is decreased and the quality of life may be significantly increased. The manufacture of tissue-engineer…

Peripheral nerve injury: a review and approach to tissue engineered constructs.

Eleven thousand Americans each year are affected by paralysis, a devastating injury that possesses associated annual costs of $7 billion (American Paralysis Association, 1997). Currently, there is no effective treatment for damage to the central nervous system (CNS), and acute spinal cord injury has been extraordinarily resistant to treatment. Compared to spinal cord injury, damage to peripheral nerves is considerably more common. In 1995, there were in excess of 50,000 peripheral nerve repair procedures performed. (National Center for Health Statistics based on Classification of Diseases, 9th

Multi-Tissue Integrated Tissue-Engineered Trachea Regeneration Based on 3D Printed Bioelastomer Scaffolds.

Functional segmental trachea reconstruction is a critical concern in thoracic surgery, and tissue-engineered trachea (TET) holds promise as a potential solution. However, current TET falls short in fully restoring physiological function due to the lack of the intricate multi-tissue structure found in natural trachea. In this research, a multi-tissue integrated tissue-engineered trachea (MI-TET) is successfully developed by orderly assembling various cells (chondrocytes, fibroblasts and epithelial cells) on 3D-printed PGS bioelastomer scaffolds. The MI-TET closely resembles the complex structur

Use of tissue engineering strategies to repair joint tissues in osteoarthritis: viral gene transfer approaches.

Osteoarthritis (OA) is a major chronic disease of the joints, affecting mostly the articular cartilage but also all the surrounding tissues including the subchondral bone, synovium, meniscus, tendons, and ligaments. Despite the availability in the clinic of a variety of therapeutic approaches, there is crucial need for improved treatment to protect and regenerate the cartilage with full integrity and function. In this regard, combining gene, cell, and tissue engineering-based procedures is an attractive concept for novel, effective therapy against AO, a slow, progressive, and irreversible dise…

Development of transcatheter implantable autologous tissue-engineered pulmonary valves using in-body tissue architecture.

Transcatheter pulmonary valve implantation (TPVI) is a minimally invasive procedure used to treat pulmonary valve dysfunction in congenital heart disease. However, the limited durability of xenogeneic bioprosthetic valves is a significant concern. Tissue-engineered heart valves (TEHVs) have emerged as a promising alternative. We developed an autologous TEHV, Biovalve, using an in-body tissue architecture technology based on tissue encapsulation. In this study, we aimed to develop stent-integrated Biovalves (stent Biovalves) that can be applied to TPVI. We designed an asymmetric hourglass-shape

A pilot study to evaluate a tissue-engineered bilayered cell therapy as an alternative to tissue from the palate.

BACKGROUND: This study evaluated the safety and effectiveness of a tissue-engineered skin product composed of viable neonatal keratinocytes and fibroblasts and compared it to a free gingival graft (FGG) in a procedure to enhance keratinized tissue (KT) and wound healing around teeth that do not require root coverage. METHODS: Twenty-five subjects were enrolled who had at least two non-adjacent teeth in contralateral quadrants exhibiting an insufficient zone of attached gingiva requiring soft tissue grafting where root coverage was not desired. One tooth was randomized to receive an FGG, and t

Enzymatically triggered glucose-releasing scaffolds as nutritional supports for millimeter-scale tissue engineering.

Construction of millimeter-sized tissues to mimic real human tissue or organ is still challenging, as oxygen and nutrients have a diffusion limit of only 100-200 μm from blood vessels. Almost all mammalian cells except red blood cells and platelets need a glucose supply to support their metabolic activities and functions. Inspired by the properties of blood vessels, we developed an alginate scaffold that is capable to release glucose by hydrolysis of glycogen via enzymes such as amyloglucosidase in this study. The scaffold can be flexibly shaped into glucose-releasing capsules (Glc-RCs) and gl

Research progress on the application of piezoelectric hydrogel materials in bone tissue engineering.

Piezoelectric hydrogels have emerged as a class of biomaterials that have garnered significant attention in bone tissue engineering in recent years. Their unique property lies in their ability to generate electrical charges under mechanical deformation. This piezoelectric effect is key to enhancing bone regeneration by mimicking the natural mechanical forces that stimulate osteogenesisin vivo. With their high water content, elasticity, biocompatibility, and capacity to modulate cellular responses through electrical stimulation (ES), they present an ideal choice for bone defect repair. Recent s…

In Vitro Platforms in Bone Tissue Engineering: From Biological Foundations to Advanced Models.

Large bone defects and related diseases affect millions worldwide, while conventional treatments such as autologous grafting remain limited by donor supply and high complication rates. Bone tissue engineering (BTE) has therefore emerged as a potential strategy to complement existing therapies. Over the past four decades, BTE has progressed from simple scaffold-based methods toward advanced bone-on-a-chip (BOC) systems. Despite substantial advances, current in vitro models still fail to fully capture the complexity of the bone microenvironment, hindering preclinical future translation. This stu…

Anisotropic ice-templated collagen scaffolds for soft tissue engineering: a mechanical characterisation toolkit.

Collagen-based soft tissues are often anisotropic in structure, and in regenerative medicine, it is important that scaffolds are designed to closely mimic their architecture and mechanical behaviour. Existing testing standards are not directly applicable to anisotropic structures in physiologically relevant conditions. The challenge is therefore to systematically quantify mechanical anisotropy, nonlinear tensile behaviour, and both in vitro degradation and fatigue in a way appropriate for soft, porous, natural macromolecular structures. In this study, we fabricated collagen scaffolds with elon
