CD43 expressed in Sertoli, Leydig and germ cells inhibits testicular function and represents a potential target to treat male infertility or effect male contraception.

CD43 is a heavily glycosylated transmembrane molecule that plays critical roles in leukocyte activation and adhesion. Previously, only hematopoietic cells were thought to normally express CD43. However, our immunohistochemical analysis of normal human testes demonstrates that the N-terminal domain of CD43 is expressed in the cytoplasm of Sertoli and Leydig cells while the C-terminal domain is expressed separately in the nucleus of Sertoli and germ cells. The observation that normal testicular cells express CD43 is entirely novel and indicates that testes function is controlled in ways not prev…

An updated review of male subfertility and leydig cell senescence under oxidative stress: Pathophysiology, mechanisms, and emerging antioxidant and nanotherapeutic approaches.

Leydig cells (LCs) are primary for testosterone production and the preservation of male reproductive function; however, their activity is highly susceptible to oxidative stress (OS)-induced damage. Accumulating evidence indicates that excessive reactive oxygen species (ROS) disrupt redox homeostasis, impair mitochondrial function, and interfere with key steroidogenic processes, ultimately promoting cellular senescence in LCs. This review provides an updated synthesis of the molecular mechanisms underlying OS-induced LC dysfunction, with particular emphasis on mitochondrial impairment, DNA dama…

Multimodal Ultrasound Characterization of a Testicular Leydig Cell Tumor With Emphasis on Microvascular Flow Imaging Findings: A Case Report and Literature Review.

We retrospectively analyzed the clinical and imaging data of a patient with pathologically confirmed testicular Leydig cell tumor (LCT). During routine preconception screening 1 year ago, a right testicular mass was detected. The patient underwent multimodal ultrasound, including two-dimensional ultrasound, color Doppler flow imaging (CDFI), elastography, and Microvascular Flow Imaging (MVFI). A distinctive feature was the visualization of both peripheral annular vascularity and intralesional blood flow from MVFI. Postoperative histopathology and immunohistochemistry confirmed the diagnosis of

Steering clear of incorrect laboratory reference ranges in Leydig cell testicular tumor-induced peripheral precocious puberty: A case report.

Leydig cell tumors are rare pediatric testicular neoplasms. We report a case of an 8-year-old boy who presented with early pubertal progression, including pubic hair and voice deepening, yet with biochemical results suggesting a prepubertal state. Normal testosterone levels and a flat Gonadotropin Releasing Hormone (GnRH) stimulation test response, bone age was advanced. Careful examination revealed asymmetric testicular size, and ultrasound identified right intratesticular lesion. Histopathology following unilateral orchiectomy confirmed a LCT. This case illustrates the diagnostic trap of rel

Ethylene dimethanesulfonate effects on gene promoter activities related to the endocrine function of immortalized Leydig cell lines R2C and MA-10.

Ethylene dimethanesulfonate (EDS) is a molecule with known selective cytotoxicity on adult Leydig cells. A single intraperitoneal injection in rats but not mice, leads to male androgen deprivation and infertility. In vitro studies using rat and mouse immortalized Leydig cell lines, showed similar effects of cell death promoted by EDS in rat cells as seen in vivo, and suggest that EDS affects gene transcription, which could firstly compromise steroidogenesis before the apoptosis process. Using gene reporter assay, this study aimed to investigate EDS effects on the promoter activity of genes imp…

Fatty acids suppress the steroidogenesis of the MA-10 mouse Leydig cell line by downregulating CYP11A1 and inhibiting late-stage autophagy.

Obese men have lower circulating testosterone than men with an optimal body mass index. Elevated fatty acids (FAs) caused by obesity have been reported to suppress the steroidogenesis of Leydig cells. Recent studies have demonstrated that autophagy regulates steroidogenesis in endocrine cells; however, few studies have investigated the molecular mechanisms of FA-impaired steroidogenesis. To study FA regulation in the steroidogenesis of Leydig cells, MA-10 cells were treated with an FA mixture and co-treated with 8-Br-cAMP to stimulate the steroidogenesis capacity. We showed that FAs led to cel…

Inhibition of progesterone biosynthesis induced by deca-brominated diphenyl ether (BDE-209) in mouse Leydig tumor cell (MLTC-1).

Polybrominated Diphenyl Ethers (PBDEs) have been extensively applied as flame retardants in different polymeric materials since the 1970s, which have become a group of long-lasting environmental pollutants. They have been reported from previous studies to accumulate and then disrupt the endocrine system in humans. However, the mechanisms are still little known. In the present study, mouse Leydig tumor cells were utilized to investigate steroidogenic activity influenced by deca-brominated diphenyl ether (BDE-209). Our data showed that BDE-209 did not change intracellular cAMP level in the prese

Confounder factors masking a Leydig-cell ovarian tumor in a post-menopausal woman treated for androgen-positive receptor breast cancer.

Post-menopause hyperandrogenism is a condition that needs careful evaluation. Aromatase inhibitors (AI), which are important in the management of positive estrogen breast cancer, and chronic kidney disease (CKD) can puzzle the evaluation of this condition. A postmenopause female with type-2 diabetes and advanced CKD was attended due to progressive virilization, which has started after the introduction of an AI for breast cancer 5 years earlier. Clinical and radiological investigation has confirmed a pure Leydig cell tumor as source of hyperandrogenism. Re-evaluation of the breast tumor immunoh

Role of Heat Shock Factor 1 in Conserving Cholesterol Transportation in Leydig Cell Steroidogenesis via Steroidogenic Acute Regulatory Protein.

Testicular testosterone synthesis begins with cholesterol transport into mitochondria via steroidogenic acute regulatory (StAR) protein in Leydig cells. Acute heat stress is known to obstruct testicular steroidogenesis by transcriptional repression of StAR. In contrast, chronic heat stress such as cryptorchidism or varicocele generally does not affect testicular steroidogenesis, suggesting that Leydig cells adapt to heat stress and retain their steroid synthesis ability. However, the mechanisms of the stress response in steroid-producing cells are unclear. We examined the relationship between …

A Rare Case of Intra-Endometrial Leiomyoma of Uterus Simulating Degenerated Submucosal Leiomyoma Accompanied by a Large Sertoli-Leydig Cell Tumor.

A 50-year-old peri-menopausal woman presented with hard palpable mass on her lower abdomen and anemia from heavy menstrual bleeding. Ultrasonography showed a 13×12 cm sized hypoechoic solid mass in pelvis and a 2.5×2 cm hypoechoic cystic mass in uterine endometrium. Abdomino-pelvic computed tomography revealed a hypodense pelvic mass without enhancement, suggesting a leiomyoma of intraligamentary type or sex cord tumor of right ovary with submucosal myoma of uterus. Laparoscopy revealed a large Sertoli-Leydig cell tumor of right ovary with a very rare entity of intra-endometrial uterine leiomy

Perfluorooctane Sulfonate Concentrations in Amniotic Fluid, Biomarkers of Fetal Leydig Cell Function, and Cryptorchidism and Hypospadias in Danish Boys (1980-1996).

BACKGROUND: Exposure to perfluorooctane sulfonate (PFOS) may potentially disturb fetal Leydig cell hormone production and male genital development. OBJECTIVES: We aimed to study the associations between levels of amniotic fluid PFOS, fetal steroid hormone, and insulin-like factor 3 (INSL3) and the prevalence of cryptorchidism and hypospadias. METHODS: Using the Danish National Patient Registry, we selected 270 cryptorchidism cases, 75 hypospadias cases, and 300 controls with stored maternal amniotic fluid samples available in a Danish pregnancy-screening biobank (1980-1996). We used mass spe

Poorly differentiated ovarian sertoli-leydig cell tumor in a 16-year-old single woman: a case report and literature review.

Sertoli-Leydig cell tumor (SLCT) of ovary is an exceedingly unusual neoplasm that belongs to a group of sex cord-stromal tumors of ovary and accounts for less than 0.5% of all primary ovarian neoplasms. Very few case reports have been documented in the literature so far. Herein, we report a case of primary poorly differentiated ovarian Sertoli-Leydig cell tumor (SLCT) involving the left ovary in a 16-year-old single woman who presented with a 3-month history of a pelviabdominal mass, acne, hirsutism, and menstrual irregularities. In addition, a literature review on ovarian SLCTs is provided.

Cyclic GMP signaling in rat urinary bladder, prostate, and epididymis: tissue-specific changes with aging and in response to Leydig cell depletion.

Aging of the male reproductive system leads to changes in endocrine signaling and is frequently associated with the emergence of prostate hyperplasia and bladder dysfunctions. Recent reports highlight prostate and bladder as promising targets for therapeutic interventions with inhibitors of the cyclic GMP (cGMP)-degrading phosphodiesterase 5 (PDE5). However, the cGMP signaling system in these organs is as yet poorly characterized, and the possibility of age-related alterations has not been addressed. This study investigates key proteins of cGMP pathways in bladder, prostate, and epididymis of

High levels of cholesteryl esters, progesterone and estradiol in the testis of aging male Fischer 344 rats: feminizing Leydig cell tumors.

The levels and fatty acid composition of cholesteryl esters (CEs), and the levels of steroid hormones in the testis of aging Fischer 344 (F344) rats were studied in comparison with those of 6-month-old rats without Leydig cell tumors (controls). The total lipid content in the testis increased as Leydig cell tumors developed: in 23-month-old rats, the content increased to five times higher than that of the controls. The CE level reached about 300 times higher than that of the controls and comprised 57.5% of the total lipid of the testis. The fatty acid composition of the CEs in the tumor compar
