The formation of somite compartments in the avian embryo.

The somites develop from the unsegmented paraxial mesoderm that flanks the neural tube. They form in an intrinsic process which lays down the primary segmental pattern of the vertebrate body. We review the processes of somitogenesis and somite differentiation as well as the mechanisms involved in these developmental events. Long before overt differentiation occurs, different compartments of the still epithelial somites give rise to special cell lines and to particular derivatives. By means of isotypic grafting between quail and chick embryos, it is possible to follow the fate of groups of somi…

Zebrafish Cdx1b modulates epithalamic asymmetry by regulating ndr2 and lft1 expression.

Nodal signaling is essential for mesoderm and endoderm formation, as well as neural plate induction and establishment of left-right asymmetry. However, the mechanisms controlling expression of Nodal pathway genes in these contexts are not fully known. Previously, we showed that Cdx1b induces expression of downstream Nodal signaling factors during early endoderm formation. In this study, we show that Cdx1b also regulates epithalamic asymmetry in zebrafish embryos by modulating expression of ndr2 and lft1. We first knocked down cdx1b with translation-blocking and splicing-blocking morpholinos (M…

Upper Sternal Cleft Associated With Congenital Cardiac Defect: Single Stage Surgical Correction.

Sternal cleft is a rare congenital malformation of anterior chest wall resulting from the failure of mesodermal migration to the midline during early embryological development. It may occur as an isolated anomaly or in connection with other congenital defects, most commonly of the cardiovascular system. The authors report the case of a 6-month-old girl with an upper sternal cleft combined with ventricular and atrial septal defects, who underwent single-stage surgical correction. The cosmetic and functional outcomes of the reconstruction were very good, with a stable anterior chest wall. The ch…

Intraneural Hemangioma: Case Report of a Rare Tibial Nerve Lesion.

An intraneural hemangioma is a rare, benign mesodermal lesion. We present a case of a three-year-old female with the inability to straighten her right knee and fullness over the right popliteal fossa for one year. Magnetic resonance imaging (MRI) demonstrated a T2 hyperintense lesion of the popliteal fossa, within the tibial nerve. The patient underwent an uncomplicated right knee excisional biopsy, which confirmed the diagnosis of an intraneural hemangioma. Although rare, an intraneural hemangioma should be considered in the differential diagnosis of a soft tissue lesion located in the expect…

PET/CT demonstration and monitoring of thoracic and abdominal wall mesothelioma.

Mesothelioma is a malignancy arising from the embryonic coelomic mesodermal lining forming the pleura, peritoneum, pericardium, and tunica vaginalis. It is mostly induced by exposure to asbestos. The author presents a 77-year-old woman with an atypical manifestation of epithelioid mesothelioma, which does not follow the expected clinical characteristics of this disease mentioned above. In this case, PET/CT provides useful information concerning the extension of the lesions to thoracic and abdominal walls not fully evaluated by the initial conventional cross-sectional imaging. PET/CT also allow…

The development of the male genitourinary system: III. The formation of the spongiose and glandar urethra.

It is generally agreed that the urethral plate disintegrates, resulting in the urethral groove. This is subsequently transformed into the urethra by fusion of the urethral folds, which flank its sides. Recently, the existence of such a groove and folds has been denied and this challenge to the long accepted existence of such folds is significant since hypospadias is considered to result from failure of their fusion. The present studies indicate that mesodermal fold formation and its subsequent subepithelial fusion across the midline plays an essential role in urethral tube formation. Disruptio…

Transverse vaginal septum. A case report.

A case occurred of imperforate, complete transverse vaginal septum in the lower third of the vagina. The patient presented at 16 years of age with primary amenorrhea and was found to have hematocolpos. The clinical impression was an imperforate hymen because of the very low position of the septum in the vagina. After excision the diagnosis of transverse vaginal septum was made histologically because of the presence of müllerian duct (mesodermal origin) tissue in the septum. The patient also had syndactyly of the second and third toes on her right foot. The association of these two congenital a…

Two-year-old boy with Proteus syndrome and fatal pulmonary thromboembolism.

A 2-year-old boy with a relatively mild form of Proteus syndrome (PS) died suddenly during a previously uncomplicated postsurgical convalescence. Autopsy demonstrated massive acute pulmonary embolism, which has not been previously reported in PS. In addition, clinically occult mesodermal hamartomata, predominantly hemolymphangiomata, were found to be widespread in the pelvoabdominal viscera, including spleen, appendix, kidneys, adrenal gland, liver, and retroperitoneal soft tissues. Such lesions may well be common in PS patients but may not be detected by conventional radiographic imaging tech…

[Cystic lymphangioma of the retroperitoneum. A case report].

Cystic lymphangioma of the retroperitoneum is a primary mesodermal derived benign neoplasm, arising from retroperitoneal lymphatics. The Authors report a case of cystic lymphangioma of the retroperitoneum recently observed, and a review of the international literature registering 181 cases from 1958 up to nowadays. Retroperitoneal localization, sub-clinic evolution and the impossibility to preoperatively define the histological type of the tumour, in spite of the modern diagnostic techniques, explain the difficulties to reach a correct differential diagnosis. The authors conclude that surgery …

[Early amnion rupture or amniotic band syndrome].

Early rupture of the amnion with resultant oligohydramnios and subsequent development of mesodermal bands between the chorion and the fetus can cause various deformities. The degree of deformity depending on the extent of the band formation and the gestational age at which this occurs. The condition may involve all parts of the fetus and comprizes a spectrum ranging from clinically insignificant small constricting bands on the extremities to large craniofacial and visceral defects which may be fatal and cause abortion. Prenatal diagnosis relies mainly on ultrasonic investigation, and the treat…

[Monomelic melorheostosis of the hand].

Melorheostosis is a rare sclerosing bone dysplasia affecting children and adults. Concerning the etiology it seems to be a metameric abnormality of the tissue of mesodermal origin, which is initiated early in embryonic life prior to the formation of the extremity buds. The flowing hyperostosis is usually associated with limb deformity, soft tissue contractures, severe pain and stiffness of joints. Bone scanning complements standard radiologic studies and permits evaluation of the extent and activity of the disease. Conservative treatment is disappointing. Thus we recommend the excision of the …

Two cases of megalourethra are presented, the ninth and tenth cases to be documented in the literature. Both patients exhibited mesodermal abnormalities which justify categorization among the minor forms of prune belly syndrome. One patient, the youngest to have undergone surgical correction, presented with azotemia, dilated posterior urethra, megacystis, and megaureters. The second patient with incomplete or scaphoid form of megalourethra also exhibited undescended testis, corrected by orchiopexy at the time of urethoplasty. In all instances the goal of treatment is preservation of renal func…

Human placentation: foundations and implications for reproductive endocrinology and infertility.

Human fetal development requires sustenance via the placenta, which mediates molecular transport between maternal and fetal circulations. Placental formation begins as cells of the trophoblast lineage differentiate and the extraembryonic mesoderm becomes vascularized, assembling a unique organ de novo that facilitates nutrient and gas exchange, waste removal, hormone production and immune modulation. We describe how placentation is orchestrated to keep pace with fetal growth, but is vulnerable to disruption by medical interventions for infertility. Initially, trophoblast stem cells differentia…

ETV2 and VEZF1 interaction and regulation of the hematoendothelial lineage during embryogenesis.

Ets variant 2 (Etv2), a member of the Ets factor family, has an essential role in the formation of endothelial and hematopoietic cell lineages during embryonic development. The functional role of ETS transcription factors is, in part, dependent on the interacting proteins. There are relatively few studies exploring the coordinated interplay between ETV2 and its interacting proteins that regulate mesodermal lineage determination. In order to identify novel ETV2 interacting partners, a yeast two-hybrid analysis was performed and the C2H2 zinc finger transcription factor VEZF1 (vascular endotheli…

Stemness enhancement of human neural stem cells following bone marrow MSC coculture.

Rapid loss of stemness capacity in purified prototype neural stem cells (NSCs) remains a serious challenge to basic and clinical studies aiming to repair the central nervous system. Based on the essential role of mesodermal guidance in the process of neurulation, we hypothesized that coculture of human NSCs (hNSCs) with human bone marrow-derived mesenchymal stromal stem cells (hMSCs) could enhance the stemness of hNSCs through Notch-1 signaling. We have now tested the hypothesis by assessing behaviors of hNSCs and hMSCs under systematically designed coculture conditions relative to monoculture…
