Genetic background determines susceptibility to experimental immune-mediated blepharoconjunctivitis: comparison of Balb/c and C57BL/6 mice.

Balb/c and C57BL/6 mice have been reported to be biased towards Th2 and Th1 immune responses, respectively. We investigated which strain is more susceptible to the development of experimental immune-mediated blepharoconjunctivitis (EC), which is predominantly mediated by Th2 immune responses. EC was induced by three different methods in Balb/c and C57BL/6 mice using ragweed (RW) as the antigen. The mice were thus either actively immunized with RW, passively immunized by transfer of RW-primed T cells, or passively immunized by transfer of RW-specific IgE, followed by RW challenge in eye drops.

Differential plague-transmission dynamics determine Yersinia pestis population genetic structure on local, regional, and global scales.

Plague, the disease caused by the bacterium Yersinia pestis, has greatly impacted human civilization. Y. pestis is a successful global pathogen, with active foci on all continents except Australia and Antarctica. Because the Y. pestis genome is highly monomorphic, previous attempts to characterize the population genetic structure within a single focus have been largely unsuccessful. Here we report that highly mutable marker loci allow determination of Y. pestis population genetic structure and tracking of transmission patterns at two spatial scales within a single focus. In addition, we found

A novel method for assessing gastritis in the murine model demonstrates genetically determined variation in response to Helicobacter felis infection.

We reported that melatonin prevents the progression of carbon tetrachloride (CCl4)-induced acute liver injury in rats possibly by attenuating enhanced lipid peroxidation and reduced glutathione depletion. Herein, we examined the effect of melatonin on the changes in hepatic reactive oxygen species (ROS) metabolism in rats with a single intraperitoneal injection of CCl4 (1.6 g/kg body weight); the intent was to clarify the therapeutic mechanism of the indoleamine on CCl4-induced acute liver injury. Rats with and without CCl4 treatment received a single oral dose of melatonin (10, 50 or 100 mg/k

Topography of genetic elements of X-chromosome relative to the cell nucleus and to the chromosome X territory determined for human lymphocytes.

Topography of three genetic elements--dystrophin (dmd) exons 5-7 (E(1)), 46-47 (E(2)), and centromere of chromosome X (N(X)) were studied relative to cell nuclei and to chromosome X territories of spatially fixed human lymphocytes. Repeated three-dimensional (3D) dual color fluorescence in situ hybridization combined with high-resolution cytometry was used. In addition, the nuclear location of fluorescence weight centers (FWC), spatial volume, and maximal area per one section of chromosome-X territories were investigated. The larger (X(L)) and smaller (X(S)) homologous X-chromosomes were disti

Necator americanus in inbred mice: evidence in support of genetically determined differences in the cellular immune response to a primary infection.

The blood and broncho-alveolar leucocyte (BAL) responses, to a primary Necator americanus infection, were studied in male BALB/c and NIH mice. Following percutaneous infection, a significant blood leucocytosis occurred in both the BALB/c and NIH strains. The peak response occurred, in both strains, on day 10 p.i. and this was reflected in the eosinophil response which peaked at the same time-point. The change in blood eosinophil numbers, as a result of infection, was found to be the greatest recorded for any cell type. In BALB/c mice, however, infection elicited a significantly more intense to

Genetic determinants of a nickel-specific transport system are part of the plasmid-encoded hydrogenase gene cluster in Alcaligenes eutrophus.

Nickel-deficient (Nic-) mutants of Alcaligenes eutrophus requiring high levels of nickel ions for autotrophic growth with hydrogen were characterized. The Nic- mutants carried defined deletions in the hydrogenase gene cluster of the indigenous pHG megaplasmid. Nickel deficiency correlated with a low level of the nickel-containing hydrogenase activity, a slow rate of nickel transport, and reduced activity of urease. The Nic+ phenotype was restored by a cloned DNA sequence (hoxN) of a megaplasmid pHG1 DNA library of A. eutrophus H16. hoxN is part of the hydrogenase gene cluster. The nickel requi

Association of genetically determined plasma hepatocyte growth factor with lung cancer and its subtypes: Mendelian randomization and mediation analysis.

Growing scientific evidence suggested that hepatocyte growth factor (HGF) might play a crucial role in the development of lung cancer, which might be influenced by the epidermal growth factor (EGF)/EGF receptor. However, the specific causality behind the association has not been clarified due to potential bias. Thus, a Mendelian randomization (MR) study was conducted to investigate the effects of gene-determined elevated plasma HGF on the risk of lung cancer and its subtypes, as well as the mediating effects of EGF. Thirteen instrumental variants for plasma HGF were derived from a genome-wide

Genetic determinants of disinfectant and antibiotic resistance, and virulence factors in Enterococcus faecalis isolated from a pasteurized crab canning environment.

Blue swimming crab (Portunus pelagicus) is highly commercial Indonesian seafood commodity and exported in the form of pasteurized canned crab product. As a ready-to-eat food industry, the effectiveness of sanitization practices and the level of hygiene in the production environment are critical factors in ensuring food safety. These challenges are heightened by bacteria that form biofilms or carry resistance genes. This study aimed to identify resistance bacterial species and strain, identify disinfectants and antibiotic resistance genes, and virulence genes in the pasteurized crab production

Genetic determinants of gestational diabetes mellitus in thai pregnant women: role of GCKR, CDKAL1, TCF7L2, NEDD1, and CMIP variants.

BACKGROUND: Gestational diabetes mellitus (GDM) has a high global prevalence and arises from complex interactions between genetic predisposition and environmental factors. GDM is associated with metabolic disturbances and chronic low-grade inflammation, both of which contribute to its pathogenesis. This study aimed to investigate the association between GDM and 135 single-nucleotide polymorphisms (SNPs) across 20 genes related to metabolic traits. METHODS: In this case-control study, 152 pregnant women with GDM and 684 pregnant women with normal glucose tolerance (NGT) who underwent antenatal

Antimicrobial Resistance and Associated Genetic Determinants of Aliivibrio Colonizing the Gastrointestinal Tract of Farmed Atlantic Salmon (Salmo salar L.).

BACKGROUND/OBJECTIVES: Currently, readily available data on antimicrobial resistance in bacteria colonizing farmed Atlantic salmon (Salmo salar L.) are limited. Recent data indicate that the gastrointestinal tract mucosa of adult Atlantic salmon farmed in Tasmania (Australia) is consistently colonized by Aliivibrio species. Aliivibrio and other Vibrionaceae may contribute to gut dysbiosis through overgrowth. We investigated Aliivibrio isolates and other Aliivibrio species to link antimicrobial resistance (AMR) phenotypes with the presence of antimicrobial resistance genes (ARGs). METHODS: We

Integrated Genomic, Transcriptomic and Metabolomic Analyses Identify Key Genetic Determinants and Regulatory Networks for Salinity Tolerance in Macrobrachium rosenbergii.

Salinity stress severely restricts the culture and geographical expansion of Macrobrachium rosenbergii. Artificial selective breeding has generated a salt-tolerant (NY) strain that can survive and grow normally under 10‰ salinity, while the commercial Suhu No.1 (SH) strain and commercial population (PT) exhibit weaker salt adaptability. In the present study, we performed whole-genome resequencing, transcriptome and untargeted metabolomic analyses to systematically reveal the molecular basis of salinity tolerance in the NY strain. Population genetic analyses including ADMIXTURE, PCA and phyloge

Genetic variation in IL-4 activated tissue resident macrophages determines strain-specific synergistic responses to LPS epigenetically.

How macrophages in the tissue environment integrate multiple stimuli depends on the genetic background of the host, but this is still poorly understood. We investigate IL-4 activation of male C57BL/6 and BALB/c strain specific in vivo tissue-resident macrophages (TRMs) from the peritoneal cavity. C57BL/6 TRMs are more transcriptionally responsive to IL-4 stimulation, with induced genes associated with more super enhancers, induced enhancers, and topologically associating domains (TAD) boundaries. IL-4-directed epigenomic remodeling reveals C57BL/6 specific enrichment of NF-κB, IRF, and STAT mo

Genetic determinants of blood-cell traits and the risk of cardiometabolic Diseases: Triangulating evidence from Mendelian randomization and observational analyses.

BACKGROUND AND AIMS: Blood cell traits (BCTs) are linked to cardiometabolic diseases (CMDs) through mechanisms such as inflammation, thrombosis, endothelial dysfunction, and lipid metabolism. BCTs could serve as readily available, cost-effective biomarkers to enhance CVD risk prediction and, if proven causal, may also emerge as novel therapeutic targets. This study examined the genetic architecture and tested potential causal relations between 29 BCTs and 20 CMDs. METHODS: We employed univariable Mendelian randomization (UVMR) using inverse-variance weighting as the primary method, MR-Egger a

Disease clusters and their genetic determinants following a diagnosis of depression: analyses based on a novel three-dimensional disease network approach.

Depression is strongly associated with a range of subsequent diseases. To elucidate key mechanistic pathways for targeted interventions, this study aimed to determine the main disease networks associated with depression as well as their underlying genetic determinants. We developed a novel three-dimensional network approach which refines disease association verification by incorporating regularized partial correlations, and facilitates robust identification and visualization of disease clusters (i.e., groups of depression-associated diseases with high within-group connectivity) through both no

Uncommon Pathogens in Common Presentations: Genetic Profiling and Virulence Determinants of Vibrio alginolyticus Isolated from a Case of External Otitis.

Backgrunod/Objectives: Routine identification of common bacterial pathogens is typically efficient, utilizing standardized, cost-effective methods. However, the diagnostic process becomes significantly more complex when dealing with rare or unexpected microorganisms, especially as they can be considered colonizers in many cases. Methods: This study presents diagnostic details of an uncommon pathogen, Vibrio alginolyticus, isolated from auricular discharge in a patient with non-Hodgkin lymphoma diagnosed with persistent otitis externa and explores its identification through both conventional an
