CB1 receptor antagonist rimonabant protects against chronic intermittent hypoxia-induced renal injury in rats.

BACKGROUND: Obstructive sleep apnoea (OSA) induced chronic kidney disease is mainly caused by chronic intermittent hypoxia (CIH). Our study investigate the mechanism underlying CIH-induced renal damage and whether the cannabinoid receptor 1 (CB1R) antagonist rimonabant (Ri) alleviates CIH-induced renal injury. METHODS: Male Sprague-Dawley rats were randomly divided into five groups: one normal control (NC) group, two chronic intermittent hypoxia (CIH) groups, and two CIH + Ri groups. Rats in the NC groups were exposed to room air, while the CIH groups were exposed to a CIH environment for 4 w

Impact of lateral cortical notching on biomechanical performance in cephalomedullary nailing for unstable pertrochanteric fractures.

PURPOSE: In pertrochanteric femur fractures the risk for fracture healing complications increases with the complexity of the fracture. In addition to dynamization along the lag screw, successful fracture healing may also be facilitated by further dynamization along the shaft axis. The aim of this study was to investigate the mechanical stability of additional axial notch dynamization compared to the standard treatment in an unstable pertrochanteric femur fracture treated with cephalomedullary nailing. METHODS: In 14 human cadaver femora, an unstable pertrochanteric fracture was stabilized wit

APH Inhibitors that Reverse Aminoglycoside Resistance in Enterococcus casseliflavus.

Aminoglycoside-phosphotransferases (APHs) are a class of bacterial enzymes that mediate acquired resistance to aminoglycoside antibiotics. Here we report the identification of small molecules counteracting aminoglycoside resistance in Enterococcus casseliflavus. Molecular dynamics simulations were performed to identify an allosteric pocket in three APH enzymes belonging to 3' and 2'' subfamilies in which we then screened, in silico, 12,000 small molecules. From a subset of only 14 high-scored molecules tested in vitro, we identified a compound, named here EK3, able to non-competitively inhibit

Phosphopantetheinyl transferase binding and inhibition by amidino-urea and hydroxypyrimidinethione compounds.

Owing to their role in activating enzymes essential for bacterial viability and pathogenicity, phosphopantetheinyl transferases represent novel and attractive drug targets. In this work, we examined the inhibitory effect of the aminido-urea 8918 compound against the phosphopantetheinyl transferases PptAb from Mycobacterium abscessus and PcpS from Pseudomonas aeruginosa, two pathogenic bacteria associated with cystic fibrosis and bronchiectasis, respectively. Compound 8918 exhibits inhibitory activity against PptAb but displays no activity against PcpS in vitro, while no antimicrobial activity

Activity feedback and hyperedge overlap shape bistability in higher-order networks.

Behavioral feedback plays a crucial role in shaping epidemic dynamics. In this work, a contagion framework that combines higher-order interactions with a dynamic activity-feedback mechanism is developed and analyzed. An evolution equation describing the temporal dynamics of the activity rate is formulated under the assumption that group behavior is regulated by the current prevalence. Two different bistable states are identified, and the conditions for their appearance and disappearance are clarified. To assess the generality of these findings, we numerically simulated the framework on network

NMR Characterization of the Papain-like Protease from SARS-CoV-2 Identifies the Conformational Heterogeneity in Its Inhibitor-Binding Site.

Papain-like protease (PLpro) from severe acute respiratory syndrome Coronavirus 2 (SARS-CoV-2) is a prime target for the development of antivirals for Coronavirus disease 2019 (COVID-19). However, drugs that target the PLpro protein have not yet been approved. In order to gain insights into the development of a PLpro inhibitor, conformational dynamics of PLpro in complex with GRL0617, the most well-characterized PLpro inhibitor, were investigated using nuclear magnetic resonance (NMR) spectroscopy in solution. Although mutational analyses demonstrated that the L162 sidechain interaction is res

Dynamics of CRISPR loci in microevolutionary process of Yersinia pestis strains.

The potential use of CRISPR loci genotyping to elucidate population dynamics and microevolution of 146 Yersinia pestis strains from different biovars and locations was investigated in this work. The majority of strains from the Orientalis biovar presented specific spacer arrays, allowing for the establishment of a CRISPR signature for their respective isolates. Twenty-one new spacers were found in the Y. pestis strains from plague foci in Brazil. Ninety-three (64%) strains were grouped in the G1 genotype, whereas the others were distributed in 35 genotypes. This study allowed observing a micro

Ultraviolet Excitation Dynamics of Nitrobenzenes.

Time-resolved photoelectron imaging was used to investigate nonadiabatic processes operating in the excited electronic states of nitrobenzene and three methyl-substituted derivatives: 3,5-, 2,6-, and 2,4-dimethylnitrobenzene. The primary goal was evaluating the dynamical impact of the torsional angle between the NO2 group and the benzene ring plane-something previously implicated in mediating the propensity for branching into different photodissociation pathways (NO vs NO2 elimination). Targeted, photoinitiated release of NO radicals is of interest for clinical medicine applications, and there
