[Use of polymerase chain reaction in the diagnosis of tuberculosis in children with diseases of the bronchopulmonary system].

A comprehensive study was conducted to determine the activity of tuberculous infection in 115 patients who formed 3 groups: 1) patients with primary forms of tuberculosis; 2) those who were infected with Mycobacterium tuberculosis (MBT); and 3) those who were infected with bronchopulmonary diseases. The detection of MBT DNA in the blood by polymerase chain reaction (PCR) using a highly sensitive four-primed diagnostic kit established that 80 patients with tuberculosis had positive results and their decrease as the patient had a course of tuberculostatic therapy. The fact that a third of patien…

Comparison of four polymerase chain reaction assays for specificity in the identification of Streptococcus pneumoniae.

We determined the usefulness of 4 conventional polymerase chain reaction (PCR) assays, lytA, psaA, and two primer sets from the ply gene, for accuracy in the discrimination of nontypeable (NT) Streptococcus pneumoniae from closely related atypical streptococci. The study used 100 strains. We compared the PCR results with laboratory tests that included optochin (ethylhydrocupreine hydrochloride) sensitivity, bile solubility, the Quellung reaction, and AccuProbe (Gen-Probe Inc., San Diego, CA). These latter tests did not discriminate the atypical streptococci from the NT pneumococci. All PCR pri…

[Detection of plague microbe in the fleas by polymerase chain reaction by using magnetic immunosorbents].

The polymerase chain reaction (PCR) with and without pre-treatment of the samples of the fleas Citellophilus tesquorum with magnoimmunosorbents was used to detect the plague microbe in the samples containing 142, 168, 193 or more bacterial cells. PCR analysis reduced the time of tests of ectoparasites for plague to 6 hours. The use of a combination of PCR and magnoimmunosorbent simplified the most time-consuming and longest stage of isolation of plague microbial DNA from the suspensions of the fleas, which allows the time of a test to be reduced to 3 hours. The administration of an affine sorb…

An interlaboratory comparison for the detection of Mycoplasma pneumoniae in respiratory samples by the polymerase chain reaction.

A panel of 78 respiratory samples collected from 43 patients was analyzed in three different Centers for the presence of Mycoplasma pneumoniae DNA by polymerase chain reaction (PCR). One Center collected the samples and extracted the DNA by two different methods. DNA extracted according to the first method was amplified using primers targetting the 16 S rRNA gene. DNA extracted according to the second method was amplified using the same primers in a semi-nested format and was sent to the two other Centers. The latter Centers both used the same primers targetting the P1 gene but with a differen…

Molecular probes and the polymerase chain reaction for detection and typing of Leishmania species in Mexico.

Leishmaniasis in Mexico is a public health problem because all the clinical forms have been recorded in most Mexican states. We studied patients showing clinical symptoms of any form of leishmaniasis, from several endemic areas. Bone marrow samples, aspirates or skin biopsies were taken and deoxyribonucleic acid (DNA) was extracted and amplified by the polymerase chain reaction (PCR) with universal primers AJS1 and DeB8, specific for the Leishmania subgenus Leishmania. The PCR products were then hybridized by dot- or Southern blotting and probed with probe 9.2, specific for the L. mexicana com…

Diagnosis by polymerase chain reaction of pneumonia caused by Legionella pneumophila in an immunocompetent child.

A 10-year-old, previously healthy child with pneumonia caused by Legionella pneumophila diagnosed by polymerase chain reaction (PCR) of serum is presented. Diagnostic methods were PCR of serum using two different primer sets, and the detection of specific antibody in paired sera using an indirect immunofluorescence assay. Legionella DNA was amplified from serum obtained before and on day 6, but not after completion of a 14-day course of oral clarithromycin. The etiologic role of L. pneumophila was confirmed by seroconversion. The report illustrates that L. pneumophila PCR of serum may contribu…
