Effects of commercial genetic selection on gene expression in the developing neuroendocrine system of broilers.

Selective breeding of broilers has significantly improved growth rates, muscle mass, and feed efficiency and may have influenced the neuroendocrine systems that regulate growth and metabolism. Embryonic development represents one-third of the life of a modern broiler. To assess the impact of genetic selection on the neuroendocrine regulation of growth and metabolism during embryonic development, we examined mRNA expression of growth-related genes in the hypothalamus and anterior pituitary of two chicken breeds: the modern Ross 708 broiler and the Athens Canadian Random Bred (ACRB) line, the ol

Feedback Signaling between Cholangiopathies, Ductular Reaction, and Non-Alcoholic Fatty Liver Disease.

Fatty liver diseases, such as non-alcoholic fatty liver disease (NAFLD), are global health disparities, particularly in the United States, as a result of cultural eating habits and lifestyle. Pathological studies on NAFLD have been mostly focused on hepatocytes and other inflammatory cell types; however, the impact of other biliary epithelial cells (i.e., cholangiocytes) in the promotion of NAFLD is growing. This review article will discuss how cholestatic injury and cholangiocyte activity/ductular reaction influence NAFLD progression. Furthermore, this review will provide informative details

Critical Illness and the Frailty Syndrome: Mechanisms and Potential Therapeutic Targets.

Frailty is a syndrome characterized by decreased reserves across multiple physiologic systems resulting in functional limitations and vulnerability to new stressors. Physical frailty develops over years in community-dwelling older adults but presents or worsens within days in the intensive care unit (ICU) because common mechanisms governing age-related physical frailty are often exacerbated by critical illness. The hallmark of physical frailty is a combined loss of muscle mass, force, and endurance. About one-third of ICU patients have frailty before hospitalization, which increases their risk

Application of Proteomic Techniques for Improved Stratification and Treatment of Schizophrenia Patients.

For major psychiatric disorders such as schizophrenia, there have been shortcomings in the translation of scientific findings into new treatments and this has led to diminished interest for large pharmaceutical companies. This chapter describes how incorporation of proteomic approaches into the clinical pipeline can lead to identification and implementation of biomarker tests for improved patient characterization, prediction of treatment response and monitoring treatment effects to help revitalize efforts in this important area. In addition, the construction of specific biomarker tests for dis

[Neuroendocrine disturbances after acquired brain damage].

Hypopituitarism is not a rare disease and its clinical signs and symptoms deserve the attention of the clinically practising neurologist. Next to the classical cause of hypopituitarism mediated by tumours of the hypothalamo-pituitary region, a number of recent articles have highlighted the high frequency of central endocrine disturbances in patients with brain damage, i. e. not only after traumatic brain injury and subarachnoid haemorrhage but also as a consequence of the treatment of childhood brain tumours. This article provides an overview of the clinical symptomatology and pathophysiology

Axelrod, the pineal and the melatonin hypothesis: lessons of 50 years to shape chronodisruption research.

With key work in the 1950s and 1960s, the 1970 Nobel laureate Julius Axelrod made major contributions to the development of pineal science. Looking back at some of his accomplishments in and for the field, we feel that lessons can be derived for future work regarding impairments of the pineal gland's and melatonin's many functions for promoting health and preventing disease in man.

Pregnancy-induced adaptation in the neuroendocrine control of prolactin secretion.

During pregnancy, neuroendocrine control of prolactin secretion is markedly altered to allow a state of hyperprolactinaemia to develop. Prolactin secretion is normally tightly regulated by a short-loop negative-feedback mechanism, whereby prolactin stimulates activity of tuberoinfundibular dopamine (TIDA) neurones to increase dopamine secretion into the pituitary portal blood. Dopamine inhibits prolactin secretion, thus reducing prolactin concentrations in the circulation back to the normal low level. Activation of this feedback secretion by placental lactogen during pregnancy maintains relati

Neuroendocrine modulation induced by selective blockade of TNF-alpha in rheumatoid arthritis.

Tumor necrosis factor-alpha (TNFalpha) is a main actor in the pathogenesis of rheumatoid arthritis (RA), interacting with other molecules in complex mechanisms. The neuroendocrine system is known to be involved and Chromogranin A (CHGA) serum levels are elevated in patients with RA. We evaluated the effect of the selective blockade of TNF-alpha, induced by treatment with anti-TNF-alpha monoclonal antibodies (mAbs), on the serum levels of CHGA and on its correlation with TNF-alpha and TNF-alpha receptors (TNFRs) serum levels. Seven patients with RA have been treated with the anti-TNF-alpha mAb,

Does the type II glucocorticoid receptor mediate cortisol-induced suppression in pituitary responsiveness to gonadotropin-releasing hormone?

Stress-like elevations in plasma cortisol suppress LH pulse amplitude in ovariectomized ewes by inhibiting pituitary responsiveness to GnRH. Here we sought to identify the receptor mediating this effect. In a preliminary experiment GnRH and LH pulses were monitored in ovariectomized ewes treated with cortisol plus spironolactone, which antagonizes the type I mineralocorticoid receptor (MR), or with cortisol plus RU486, which antagonizes both the type II glucocorticoid receptor (GR) and the progesterone receptor (PR). Cortisol alone reduced LH pulse amplitude, but not pulsatile GnRH secretion,

Prolonged neurogenesis during early development of gonadotropin-releasing hormone neurones in sheep (Ovis Aries): in vivo and in vitro studies.

Gonadotropin-releasing hormone (GnRH) neurons involved in controlling the reproductive function in vertebrates are derived from the olfactory placode. However, in the sheep and the rat species, GnRH-immunoreactive (GnRH-IR) neurons could not be detected in the olfactory region during the earliest phase of GnRH system development. Using in situ hybridization (ISH) and immunohistochemistry (IHC) in sheep embryos ranging from 26 to 53 days' gestational age (G26-G53), the present work confirmed that GnRH expression could not be detected during the earliest steps of migration. The first ISH+ cells

Melatonin serum levels in rheumatoid arthritis.

The pineal hormone melatonin (MLT) exerts a variety of effects on the immune system. MLT activates immune cells and enhances inflammatory cytokine and nitric oxide production. Cytokines are strongly involved in the synovial immune and inflammatory response in rheumatoid arthritis (RA) and reach the peak of concentration in the early morning, when MLT serum level is higher. Nocturnal MLT serum levels were evaluated in 10 RA patients and in 6 healthy controls. Blood samples were obtained at 8 and 12 p.m., as well as at 2, 4, 6, and 8 a.m. MLT serum levels at 8 p.m. and 8 a.m. were found to be hi

The pigmentotropic hormone [His(7)]-corazonin, absent in a Locusta migratoria albino strain, occurs in an albino strain of Schistocerca gregaria.

[His(7)]-corazonin has recently been identified in the corpora cardiaca (CC) of two locust species, the migratory locust, Locusta migratoria and the desert locust, Schistocerca gregaria, as the dark colour inducing neurohormone. Here, we investigate whether [His(7)]-corazonin occurs in the brain-CC axis of a Schistocerca albino strain. From data obtained by immunocytochemistry, injection experiments, chromatographic and mass spectrometric analysis of brain and CC tissues, it could be concluded that an albino strain of S. gregaria from Denmark contains authentic [His(7)]-corazonin. This was une

Musculoskeletal adaptations to weightlessness and development of effective countermeasures.

A Research Roundtable, organized by the American College of Sports Medicine with sponsorship from the National Aeronautics and Space Administration, met in November 1995 to define research strategies for effective exercise countermeasures to weightlessness. Exercise was considered both independently of, and in conjunction with, other therapeutic modalities (e.g., pharmacological nutritional, hormonal, and growth-related factors) that could prevent or minimize the structural and functional deficits involving skeletal muscle and bone in response to chronic exposure to weightlessness, as well as

[The neuroendocrine mechanisms of the development of the anovulatory hyperandrogenic syndrome in rats].

The anovulatory syndrome in adult female rats occurred after subcutaneous implantation of silastic capsules with testosterone. Acceleration of testosterone conversion into estradiol in the hypothalamus, suppression of pituitary LH responsiveness to LHRH, increase of blood plasma bioactive LH, estrous cycle disorder as well as morphological changes in ovaries demonstrate the alterations in neuroendocrine control of ovulation.

Dissociation between in vivo hippocampal norepinephrine response and behavioral/neuroendocrine responses to noise stress in rats.

The behavioral and extracellular hippocampal norepinephrine responses to audiogenic stress were concomitantly characterized in freely moving rats using in vivo microdialysis. Noise stimulation produced a rapid, but short-lived increase in norepinephrine release from the hippocampus during the first 20 min of noise presentation that declined to baseline levels for the duration of the noise stimulation and following noise offset. In contrast, the behavioral response persisted throughout the duration of the noise stimulation. In a separate group of similarly treated animals, neuroendocrine indice

Oral administration of estradiol to young C57BL/6J mice induces age-like neuroendocrine dysfunctions in the regulation of estrous cycles.

Exogenous estradiol (E2) can accelerate the onset of acyclicity and other age-related neuroendocrine changes in rodents. The present study demonstrates that chronic oral administration of E2 (850 micrograms/kg body wt/day) induces premature acyclicity in intact C57BL/6J mice. After 6 wk of E2, mice regained cyclicity but ceased cycling prematurely, whereas 12 wk of E2 caused permanent acyclicity in all mice. The acyclicity after 12 wk of E2 was not reversed by ovarian replacement from young donors, which implies extra-ovarian (neuroendocrine) lesions. The above studies with intact mice can not

Electron microscopic localization of chromogranin A in osmium-fixed neuroendocrine cells with a protein A-gold technique.

An antibody (LK2H10) to chromogranin A has been recommended for use in ultrastructural identification of neuroendocrine secretory granules. Previous studies have demonstrated immunoreactive chromogranin A in specimens prepared for electron microscopy by glutaraldehyde fixation only. In this study, the effect of specimen post-fixation by osmium tetroxide on post-embedding localization of chromogranin A was evaluated. Human tissues from benign endocrine glands, neuroendocrine tumors, and non-neuroendocrine tumors were post-fixed in osmium, embedded in epoxy resin, and the sample thin sections im

Dose-related stimulation of feeding by systemic injections of monosodium glutamate.

Monosodium glutamate (MSG) is an excitotoxin capable of both stimulating and lesioning neurons in circumventricular organs (CVOs) after systemic administration. In this study, MSG and equiosmotic concentrations of NaCl were administered subcutaneously to adult rats in order to observe the effects on food and water intake. MSG (0.5, 1, 2 and 6 g/kg), but not NaCl, stimulated feeding. The magnitude of the feeding was dose-related. After the highest dose, rats consumed 4.4 g of pelleted food. Since MSG does not cross the blood-brain barrier, we conclude that feeding was stimulated by an action of

Nutrition and reproduction.

Adequate nutrition is essential for normal reproductive function in man. Menarche occurs at a critical level of 'fatness' and it appears that the hypothalamic events leading to pubertal development and the achievement of reproductive competence may be triggered by metabolic/endocrinological changes due to an increase in fat. The attainment and maintenance of ovulatory cycles demands a minimum degree of body fat (about 22%). Undernutrition results in weight loss and a reduction in fat tissue. This alteration in body composition in turn precipitates the hypothalamic changes that cause impaired g
