Comparison of methods for whole-organ decellularization in tissue engineering of bioartificial organs.

Organ transplantation is now a well-established procedure for the treatment of end-stage organ failure due to various causes, but is a victim of its own success in that there is a growing disparity in numbers between the donor organ pool available for transplantation and the patients eligible for such a procedure; hence, an alternative solution to the limited donor organ pool is both desirable and necessary. Tissue engineering is an interdisciplinary field that applies the principles of engineering and life sciences toward the development of functional replacement tissues for clinical use. A r…

Tissue engineering and the use of stem/progenitor cells for airway epithelium repair.

Stem/progenitor cells can be used to repair defects in the airway wall, resulting from e.g., tumors, trauma, tissue reactions following long-time intubations, or diseases that are associated with epithelial damage. Several potential sources of cells for airway epithelium have been identified. These can be divided into two groups. The first group consists of endogenous progenitor cells present in the respiratory tract. This group can be subdivided according to location into (a) a ductal cell type in the submucosal glands of the proximal trachea, (b) basal cells in the intercartilaginous zones o

Wet chemical synthesis of chitosan hydrogel-hydroxyapatite composite membranes for tissue engineering applications.

Chitosan, a deacetylated derivative of chitin is a commonly studied biomaterial for tissue-engineering applications due to its biocompatibility, biodegradability, low toxicity, antibacterial activity, wound healing ability and haemostatic properties. However, chitosan has poor mechanical strength due to which its applications in orthopedics are limited. Hydroxyapatite (HAp) is a natural inorganic component of bone and teeth and has mechanical strength and osteoconductive property. In this work, HAp was deposited on the surface of chitosan hydrogel membranes by a wet chemical synthesis method b

Guided cell adhesion and outgrowth in peptide-modified channels for neural tissue engineering.

A hydrogel scaffold of well-defined geometry was created and modified with laminin-derived peptides in an aqueous solution, thereby maintaining the geometry of the scaffold while introducing bioactive peptides that enhance cell adhesion and neurite outgrowth. By combining a fiber templating technique to create longitudinal channels with peptide modification, we were able to synthesize a scaffold that guided cell adhesion and neurite outgrowth of primary neurons. Scaffolds were designed to have numerous longitudinally oriented channels with an average channel diameter of 196 +/- 6 microm to ult

Implant site development: clinical realities of today and the prospects of tissue engineering.

The success of dental implants depends on their placement in bone of adequate density and volume in order to achieve primary stability. Optimal esthetics of implants requires their placement in a position approximating that of the natural teeth they replace. However, there is generally at least some degree of atrophy in the sites where implants are to be placed. This atrophy may occur either before or after tooth extraction. Following extraction of teeth, there is commonly alveolar ridge resorption in horizontal and vertical dimensions. Alternatively, some of the oral hard and soft tissues may

Quantitative analysis of three-dimensional fluid flow in rotating bioreactors for tissue engineering.

Tissue engineering has emerged as a viable alternative to the problem of organ and tissue shortage. Our laboratory has developed matrices for bone tissue engineering based on sintered spherical particles and, using bioreactor technology, has demonstrated the ability to produce highly mineralized matrices in vitro. In this study, porous microcapsule scaffolds were developed for bone tissue engineering in the high aspect ratio vessel rotating bioreactor. The motion of individual microcapsules as well as scaffolds in the bioreactor were studied by numerical simulation and in situ imaging analysis…

Experimental study on allogenic decalcified bone matrix as carrier for bone tissue engineering.

The biocompatibility and osteogenic activity of allogenic decalcified bone matrix (DBM) used as a carrier for bone tissue engineering were studied. Following the method described by Urist, allogenic DBM was made. In vitro, DBM and bone marrow stromal cell (BMSC) from rabbits were co-cultured for 3-7 days and subjected to HE staining, and a series of histomorphological observations were performed under phase-contrast microscopy and scanning electron microscopy (SEM). In vivo the mixture of DBM/BMSC co-cultured for 3 days was planted into one side of muscules sacrospinalis of rabbits, and the DB…

Pore architecture effects on chondrogenic potential of patient-specific 3-dimensionally printed porous tissue bioscaffolds for auricular tissue engineering.

OBJECTIVE: This study aims to determine the effect of auricular scaffold microarchitecture on chondrogenic potential in an in vivo animal model. METHODS: DICOM computed tomography (CT) images of a human auricle were segmented to create an external anatomic envelope. Image-based design was used to generate 1) orthogonally interconnected spherical pores and 2) randomly interspersed pores, and each were repeated in three dimensions to fill the external auricular envelope. These auricular scaffolds were then 3D printed by laser sintering poly-l-caprolactone, seeded with primary porcine auricular

Graphene-based materials offer tunable mechanical, electrical, and biochemical properties that address longstanding challenges in cartilage tissue engineering. This review synthesizes recent advances in graphene derivatives, scaffold fabrication strategies, and emerging mechanisms governing cell-graphene interactions. We evaluate how graphene enhances mechanical reinforcement, bioactivity, and matrix production across hydrogels, polymer composites, and ECM-derived scaffolds. Key opportunities and translational pathways are identified to guide the development of next-generation cartilage regene…

Hydroxyapatite-Polysaccharide Composites Synthesized from Maize Lime-Cooking Wastewater for Bone Tissue Engineering.

Hydroxyapatite (HAp) is a widely used bioceramic in bone tissue engineering due to its biocompatibility and osteoinductive capacity; however, sustainable low-cost synthesis routes remain a challenge. This study evaluated HAp-polysaccharide composite synthesis from nejayote, the alkaline wastewater of maize nixtamalization, via chemical precipitation with (NH4)3PO4 under controlled and uncontrolled pH, followed by calcination at 550 °C for 2 or 4 h. Controlled pH synthesis yielded higher solid recovery (89.8% vs. 76.4%), better calcium removal (99.8% vs. 87.4%), and smaller particle sizes (423.…

Comparing Primary Human Myogenic Spheroids and Single Cells for Skeletal Muscle Tissue Engineering.

Skeletal muscle tissue engineering aims to create bioartificial muscle (BAM) constructs that recapitulate key structural and functional features of native skeletal muscle for applications including regenerative medicine, disease modeling, and drug screening. While most BAM models are generated using myoblasts, multicellular building blocks such as myogenic spheroids may better mimic native cell-cell interactions and microarchitectural organization. In this study, we compared the use of primary human myogenic spheroids with single cells for skeletal muscle tissue engineering. Myogenic spheroids…

Synergistic integration of biomaterials in orthopedic implantation for infection preventing and tissue engineering.

The treatment of infected bone defects (IBD) is challenging in orthopedics because of their complex pathological mechanisms and high recurrence rates. An ideal therapeutic strategy should simultaneously eradicate infection and promote bone regeneration. However, conventional approaches rarely achieve both objectives effectively. It has concurrently high recurrence rates, prolonged treatment courses and limited therapeutic efficacy. Clinically, implant-associated infections leading to IBD are often exacerbated by patients'underlying diseases, material-related risks and perioperative factors, th
