Esterification-deesterification of estradiol by human mammary cancer cells in culture.

Formation of lipoidal derivatives of estradiol-17 beta (E2) esterified to long-chained fatty acids has been reported to occur in estrogen target tissues. Employing human breast cancer cells in culture, we have detected the rapid synthesis of such compounds upon exposure of the cells to concentrations of [3H]E2 as low as 1 nM. When exposed to 10 nM [3H]E2 in the culture medium, synthesis of E2-lipoidal derivative (E2-L) reached 270 fmol/mg DNA in 2 h in the estrogen receptor positive MCF-7 human mammary cancer cell line. Higher rates (approximately 900 fmol/mg DNA in 2 h) were reached in 2 estr

Cellular effects of combined Ctn-2 peptide and doxorubicin treatment in breast cancer cells: A preliminary study of membrane interaction, ROS production, and cell cycle distribution.

BACKGROUND: Triple-negative breast cancer (TNBC) lacks targeted therapies and is commonly treated with doxorubicin, whose efficacy is limited by toxicity and chemoresistance. Bioactive peptides have emerged as potential adjuvants due to their membrane activity and antitumor properties. To address the potential of the combined treatment, we explored the cellular effects and underlying mechanisms of Ctn-2 peptide and doxorubicin in TNBC cells. METHODS AND RESULTS: MDA-MB-231 cells were treated with Ctn-2 (15 µM), doxorubicin (1.25-5 µM), and their combination. Membrane interactions were evaluat…

Unraveling the therapeutic potential of Elaeagnus angustifolia extract on triple-negative breast cancer (TNBC): An investigation using zebrafish model.

Breast cancer is a widespread and aggressive disease, with 2.3 million new cases globally in 2022. Metastasis, the spread of cancer cells to distant organs, remains a leading cause of breast cancer-related mortality. Current treatment options, particularly traditional chemotherapeutic drugs, are often associated with severe side effects, emphasizing the urgent need for safer and more effective therapeutic alternatives. Triple-negative breast cancer (TNBC) represents one of the most aggressive breast cancer subtypes, characterized by the absence of estrogen receptors (ER), receptors (PR), and H

Discovery of a novel PI3Kα inhibitor for breast cancer therapy via virtual screening method, molecular dynamics simulation and biological evaluation.

Phosphatidylinositol-4,5-bisphosphate 3-kinase alpha (PI3Kα) is a central signaling enzyme driving cell proliferation and growth in cancers including breast cancer. Selective inhibition of PI3Kα isoform has become a promising therapeutic approach. In this work, 2000 in-house natural compounds were virtually screened against the ATP-binding site of PI3Kα. Of these, 618 compounds were predicted to have acceptable drug-likeness, pharmacokinetic, and toxicity properties based on in silico ADMET screening. Docking analysis highlighted four candidates forming stable hydrogen bonds with key residues

Design, synthesis and biological evaluation of novel 1,3,5-triazine derivatives containing benzo[d]imidazole scaffold as PI3K inhibitors.

Twenty-seven novel 1,3,5-triazine derivatives containing benzo[d]imidazole scaffold were designed, synthesized, and evaluated for anti-tumor activity as PI3Kα inhibitors. Assessment of PI3Kα inhibitory effects revealed that some compounds were highly potent in vitro, with seventeen compounds (LNP1, LNP2, LNP4, LNP7-LNP10, LNP12-LNP18, LNP20, LNP23, and LNP24) showing IC50 values less than 100 nM. Notably, LNP23 and LNP24 showed remarkable potency against PI3Kα with IC50 values of 5.15 nM and 8.88 nM, respectively, and thus they were more potent than positive control drug ZSTK474 (PI3Kα, IC50 =

Expression of GPR68, an Acid-Sensing Orphan G Protein-Coupled Receptor, in Breast Cancer.

UNLABELLED: Breast cancer (BC) is the most diagnosed cancer and the leading cause of global cancer incidence in 2020. It is quite known that highly invasive cancers have disrupted metabolism that leads to the creation of an acidic tumor microenvironment. Among the proton-sensing G protein-coupled receptors is GPR68. In this study, we aimed to explore the expression pattern of GPR68 in tissues from BC patients as well as different BC cell lines. METHODS: In-silico tools were used to assess the expression of GPR68 in BC patients. The expression pattern was validated in fresh and paraffin-embedd

Targeting PD-L1 and disrupting Siglec-9 cooperatively potentiate macrophage phagocytosis of breast cancer via a trispecific macrophage engager (TriME).

Breast cancer (BC) remains a leading cause of mortality, highlighting the need for novel immunotherapies capable of overcoming treatment resistance. Programmed death-ligand 1 (PD-L1) is recognized as a therapeutic target, particularly within aggressive subtypes like triple-negative breast cancer (TNBC). To effectively leverage the powerful phagocytic function of macrophages, we engineered a novel trispecific macrophage engager (TriME) designed for simultaneous engagement of three components: CD16 (macrophage activation), IL-15 (immune cell survival and activation), and PD-L1 (tumor recognition

Clinically accessible drug-based nano-assemblies with self-targeting ability for NIR-II fluorescence imaging-guided surgery in triple-negative breast cancer.

RATIONALE: Accurate intraoperative visualization is critical for reducing margin positivity during breast-conserving surgery for triple-negative breast cancer (TNBC). Second near-infrared (NIR-II) fluorescence imaging represents a promising approach for precision surgery by combining lesion detection with real-time guidance. Nevertheless, the clinical translation of most fluorescence agents remains hampered by carrier-related toxicity and complex synthesis. Therefore, a "green" drug-repurposing strategy was adopted here to construct carrier-free pure-drug nano-assemblies (PDNAs), aiming to pro

ROS-Driven PKCζ Signaling as a Widely Involved Mechanism for Cancer Cell Motility and Metastasis.

The enhancement of cell motility by bioactive molecules such as growth factors, hormones, and tissue factors is pivotal in cancer invasion and metastasis. However, the molecular mechanisms underlying this enhancement remain incompletely understood. In this study, we demonstrate that hepatoblastoma HepG2 cell motility is significantly increased following hepatocyte growth factor (HGF) treatment, as assessed by phagokinetic track assays, Transwell assays, and scratch assays. This enhancement is mediated by reactive oxygen species (ROS), which activate the PKCζ/Rho GTPase signaling pathway. Notab
