Tissue distribution of 35S-metabolically labeled neublastin (BG00010) in rats.

Neublastin (NBN) is a neurotrophic growth factor that promotes the survival and regenerative properties of nociceptive neurons and has been tested in clinical trials as a treatment for neuropathic pain in individuals with sciatica and painful lumbosacral radiculopathy. Like many low molecular weight heparin binding proteins, NBN is rapidly cleared from the blood following systemic administration. To explore ADME properties of NBN in rats, we used metabolically 35S-labeled NBN following IV and SC administration quantifying counts and intact protein in kidney, liver, brain, serum, and urine at 5

The accumulation of T cells within acellular nerve allografts is length-dependent and critical for nerve regeneration.

Repair of traumatic nerve injuries can require graft material to bridge the defect. The use of alternatives to bridge the defect, such as acellular nerve allografts (ANAs), is becoming more common and desired. Although ANAs support axon regeneration across short defects (<3 cm), axon regeneration across longer defects (>3 cm) is limited. It is unclear why alternatives, including ANAs, are functionally limited by length. After repairing Lewis rat nerve defects using short (2 cm) or long (4 cm) ANAs, we showed that long ANAs have severely reduced axon regeneration across the grafts and contain S

Nerve regeneration using tubular scaffolds from biodegradable polyurethane.

INTRODUCTION: In severe nerve lesion, nerve defects and in brachial plexus reconstruction, autologous nerve grafting is the golden standard. Although, nerve grafting technique is the best available approach a major disadvantages exists: there is a limited source of autologous nerve grafts. This study presents data on the use of tubular scaffolds with uniaxial pore orientation from experimental biodegradable polyurethanes coated with fibrin sealant to regenerate a 8 mm resected segment of rat sciatic nerve. METHODS: Tubular scaffolds: prepared by extrusion of the polymer solution in DMF into w

Retinal ischaemia and delayed fibrovascular proliferation associated with an optic nerve coloboma.

CASE REPORT: A patient with a left optic nerve coloboma with late development (at 15 years of age) of vitreoretinal fibrovascular proliferation and tractional retinal detachment (TRD). Sectorial retinal photocoagulation was performed with regression of the proliferative tissue and exudation. DISCUSSION: Congenital optic nerve anomalies may be associated with significant retinal avascularity, ischaemia and late fibrovascular proliferation. Laser is effective for regression of the neovascular tissue and preventing TRD progression.

Late superior gluteal nerve palsy following posterior fracture-dislocation of the hip.

An unusual case of late superior gluteal nerve palsy complicating posterior fracture-dislocation of the hip is reported. Posterior fracture-dislocation of the hip was Grade V according to the classification of Thompson and Epstein, and Type IV according to the subclassification of Pipkin. The palsy resulted from traction by scar tissue formation. Excision of the scar tissue and decompression of the superior gluteal nerve led to complete recovery.

Ectopic thymic tissue in the mediastinum: limitations for the operative treatment of myasthenia gravis.

OBJECTIVES: The aim of the study was to investigate the distribution of ectopic thymic tissue in the mediastinum and to evaluate the possible relevance of this distribution to the therapeutic yield of thymectomies in patients with myasthenia gravis. METHODS: In this prospective autopsy study, mediastinal dissections were performed on 50 cadavers without any previously known intrathoracic pathology. The initial dissection was performed in the same way as during the maximal thymectomy. The second stage consisted of dissecting areas of fatty tissue located out of reach of the standard maximal th

Surgical management of trigeminal neuropathic pain.

Surgical treatment of mechanical trigeminal nerve injuries resulting in neuropathic pain varies with the type of injury (i.e., amputation or compression), presence of painful neuromas, and adjacent hard/soft tissues. Microsurgical principles that include access, preparation, and repair of the various nerve injuries are recommended for surgical treatment. The choice of surgical repair depends on the microscopic pathology and the presence or absence of adequate distal nerve tissue.

Progress toward understanding the molecular mechanisms of neurotrophic factor signalling.

Understanding the mechanism of action of the neurotrophic factors is central to unravelling of the mysteries of some of the neurodegenerative disorders. In this review we will discuss recent advances in our understanding of neurotrophic factor signalling in primary cultured neurons, in particular those from the superior cervical and dorsal root ganglia, as well as cerebellar granule cells, cortical neurons and oligodendrocytes.

Novel Histologic Finding: Adipose Tissue Is Prevalent Within Penile Tunica Albuginea and Corpora Cavernosa: An Anatomic Study of 63 Specimens and Considerations for Cancer Invasion.

Adipose tissue, along with arteries, veins, and peripheral nerves, is a normal constituent of mesenchymal tissues encasing the corpora cavernosa at the level of the penile shaft, variously designated as penile fascia or Bucks fascia. To our knowledge, the presence of fat has not been previously reported within the corpora cavernosa. One or 2 transversal histologic sections at the level of the surgical margin at the shaft of 63 consecutive partial penectomy specimens for squamous cell carcinoma were evaluated. From outer to inner tissues, 3 anatomic levels were identified: (1) outer fascia comp

Coil-reinforced hydrogel tubes promote nerve regeneration equivalent to that of nerve autografts.

Despite spontaneous sprouting of peripheral axons after transection injury, peripheral regeneration is incomplete and limited to short gaps, even with the use of autograft tissue, which is considered to be the "gold" standard. In an attempt to obviate some of the problems associated with autografts, including limited donor tissue and donor site morbidity, we aimed to synthesize a synthetic nerve guidance channel that would perform as well as the nerve autograft. Given that the patency of the nerve guidance channel is critical for repair, we investigated a series of nerve guidance channel desig

Ninjurin, a novel adhesion molecule, is induced by nerve injury and promotes axonal growth.

Peripheral nerve injury results in axonal degeneration and in phenotypic changes of the surrounding Schwann cells, whose presence is critical for nerve regeneration. Using differential screening strategies, we identified a novel protein, termed ninjurin (for nerve injury-induced protein), that is up-regulated after axotomy in neurons and in Schwann cells surrounding the distal nerve segment. Ninjurin is located on the cell surface, is capable of mediating homophilic adhesion, and promotes neurite extension of dorsal root ganglion neurons in vitro. Ninjurin is also expressed in a number of othe

Differences in quantitative architecture of sciatic nerve may explain differences in potential vulnerability to nerve injury, onset time, and minimum effective anesthetic volume.

BACKGROUND: In sciatic nerve (SN) blocks, differences are seen in risk of nerve damage, minimum effective anesthetic volume, and onset time. This might be related to differences in the ratio neural:nonneural tissue within the nerve. For the brachial plexus, a higher proximal ratio may explain the higher risk for neural injury in proximal nerve blocks. A similar trend in risk is reported for SN; however, equivalent quantitative data are lacking. The authors aimed to determine the ratio neural:nonneural tissue within SN in situ in the upper leg. METHODS: From five consecutive cadavers, the regi

Calretinin-immunoreactivity in the oro-facial and pharyngeal regions of the rat.

Calretinin-immunoreactivity (CR-ir) was examined in the rat oro-facial and pharyngeal tissues using an immunofluorescence method. CR-ir was distributed in the entire size range of trigeminal ganglion neurons. CR-ir was also observed in nerve fibers surrounding neuronal cell bodies in autonomic ganglia, and in nerve endings in the lip, tongue, incisal papilla, soft palate, pharynx and epiglottis. CR-immunoreactive nerve endings were all in close proximity to the epithelium, and classified into 2 types; simple (free nerve ending) and taste-bud-related types. In the salivary gland, positive nerve

A new type of Schwann cell graft transplantation to promote optic nerve regeneration in adult rats.

Like other parts of the central nervous system, the adult mammalian optic nerve is difficult to regenerate after injury. Transplantation of the peripheral nerve or a Schwann cell (SC) graft can promote injured axonal regrowth. We tried to develop a new type of tissue-engineered SC graft that consisted of SCs seeded onto a poly(lactic-co-glycolic acid)/chitosan conduit. Meanwhile, SCs were transfected along the ciliary neurotrophic factor (CNTF) gene in vitro by electroporation to increase their neurotrophic effect. Four weeks after transplantation, GAP-43 labelled regenerating axons were found
