Clinical, biochemical and histochemical studies on infants with Acrodermatitis enteropathica chronica.

7 infants diseased with Acrodermatitis enteropathica and 10 normal controls were included in this study. The values of anthranilic acid glucuronide, 6- aminohippuric, anthranilic acid, N-acetyl Kneurine, Kneurine and 30 H Kneurenine, were estimated in mg/24 hours urine, both basal and after tryptophane load. In addition, histopathological and histochemical studies for lactase, succinic dehydrogenase, alkaline phosphatase, acid phosphatase, and alpha-non-specific esterases activities were done for the intestinal mucosal biopsies. All the previous investigations were then repeated after two mont

Breath hydrogen as a diagnostic method for hypolactasia.

Breath hydrogen (H2), collected by end-expiratory sampling, was measured in twenty-five patients with abdominal symptoms or diarrhoea after ingesting 50 g. of lactose. This was compared with established tests of hypolactasia. Fifteen patients with a blood-glucose rise of more than 20 mg. per 100 ml. had less than 4 parts per million (p.p.m.) rise in breath H2 at 2 hours. In contrast, ten patients with blood-glucose rises of less than 20 mg. per 100 ml. had more than a 20 p.p.m. H2 rise (mean 85.8 p.p.m. plus or minus s.d. 44.3) at 2 hours. Similarly, two patients with normal jejunal lactase ac

Lactose Intolerance versus Cow's Milk Allergy in Infants: A Clinical Dilemma.

Due to its very early introduction, cow's milk is one of the first foods that can cause adverse reactions in human beings. Lactose intolerance (LI) and cow's milk allergy (CMA) are the most common adverse reactions to cow's milk. While LI is due to insufficient small intestinal lactase activity and/or a large quantity of ingested lactose, CMA is an aberrant immune reaction to cow's milk proteins, particularly casein or β-lactoglobulin. However, the clinical manifestations of LI and CMA, particularly their gastrointestinal signs and symptoms, are very similar, which might lead to misdiagnosis o

Lactose intolerance in patients with inflammatory bowel diseases and dietary management in prevention of osteoporosis.

Lactose intolerance affects 33% to 75% of the world population and may be associated with various genetic factors. Lactose in the diet can be found in milk and dairy products, which simultaneously constitute the primary sources of calcium. Gut microbiota also influences lactose tolerance. Patients with lactose intolerance often stop consuming milk and dairy products, which may lead to calcium and vitamin deficiency and osteoporosis. Insufficient production of lactase also occurs in patients with diseases of the gastrointestinal tract, such as inflammatory bowel diseases. Moreover, Crohn's dise

Familial aquagenic urticaria associated with familial lactose intolerance.

Aquagenic urticaria is a rare disorder characterized by the occurrence of pruritus and wheals after temporary contact with water. The familial occurrence of aquagenic urticaria over 3 generations is reported here in association with familial lactose intolerance, a condition in which the enzyme lactase encoded on chromosome 2, is deficient. In two patients, a young man and his mother, we verified the appearance of pruritic hives 5 to 10 minutes after contact with water of any temperature. Other types of physical urticaria were absent, and mastocytosis was excluded by extensive laboratory invest

Lactose absorption by postweaning rats from yogurt, quarg, and quarg whey.

Lactose-intolerant postweaning rats were fed experimental diets including yogurt, quargs prepared from yogurt culture and buttermilk culture, and two types of whey obtained from quarg processing. After feeding each diet for a period of 7 d, absence of blood glucose elevation and occurrence of diarrhea were used as indicators of lactose malabsorption. Blood glucose assays and absence of diarrhea indicated that yogurt and quargs prepared from yogurt and buttermilk culture were well tolerated by the rats. Wheys containing the same levels of viable organisms and lactose as the quargs caused severe

Lactose digestion and clinical tolerance to milk, lactose-prehydrolyzed milk and enzyme-added milk: a study in undernourished continuously enteral-fed patients.

We investigated: (1) the capacity to digest and tolerate the lactose administered by continuous infusion of intact milk to undernourished tube-fed patients, and (2) the effectiveness of lactose-prehydrolyzed milk, and of the addition of exogenous lactase to milk at infusion time, to reduce lactose maldigestion and increase clinical tolerance. Carbohydrate digestion was evaluated in 10 subjects with the hydrogen breath analysis test during 8 hr of observation. Lactose intolerance was determined by evaluation of subject's symptoms. With the infusion of intact milk (IM), none of the subjects were

Lactose malabsorption in children and adolescents with inflammatory bowel disease.

Lactose breath hydrogen tests were given to 70 children and adolescents with chronic ulcerative colitis and Crohn's disease in order to determine the prevalence of lactose malabsorption in childhood inflammatory bowel disease. Twenty-nine percent of these patients demonstrated lactose malabsorption; the majority of these children (70%) experienced gastro-intestinal symptoms during the test. The prevalence was not significantly different whether the diagnosis was ulcerative colitis or Crohn's disease. With the exception of those with diffuse small bowel disease, the location of intestinal invol

Serum lipids and proteins in lactose malabsorption.

It has been suggested that dietary lactose may reduce the intestinal absorption of fat and protein in individuals with lactase deficiency. On the other hand, it is known that a high carbohydrate diet increases serum lipids. The purpose of this study was to examine whether there are differences in the fasting serum lipid and protein concentrations between people with lactose malabsorption and people with normal lactose absorption. Therefore in the connection of a family study serum lipids and proteins were measured in 409 subjects belonging to 11 families. Of these 288 were relatives of the 11

Large-scale searching for causal relationships of milk consumption with multi-omics and disease susceptibility: A phenome-wide Mendelian randomization study.

The causal relationship between milk consumption and a broad spectrum of health outcomes remains incompletely elucidated. This study aimed to investigate potential causal effects and explore the underlying biological pathways using an integrative multi-omics framework. We conducted a large-scale 2-sample Mendelian randomization study. Genetic predisposition for milk consumption was instrumented by the lactase persistence variant (rs4988235). We assessed causal relationships with 2459 disease phenotypes from FinnGen, and 91 inflammatory proteins, 211 gut microbial taxa, 731 immunophenotypes, an

Two novel β-galactosidases from Aeromonas caviae with potential industrial applications in milk and catalytic mechanism analysis using molecular docking.

Lactose intolerance has been a significant global health concern, as it is caused by the absence of lactase, leading to the inability of the human body to absorb lactose. This study investigated two genes encoding maltose O-acetyltransferase with β-galactosidase activity from Aeromonas caviae to evaluate their potential application value for lactose degradation in bovine and human milk. The two novel β-galactosidases (AcGal25: 22.0 kDa and AcGal31: 21.3 kDa) were heterologously expressed and biochemically characterized. The optimal pH of both enzymes was 8.0, and the optimal temperature of AcG

Lactose Malabsorption and Presumed Related Disorders: A Review of Current Evidence.

BACKGROUND: Lactose malabsorption (LM) is a frequent clinical problem associated with several digestive and extra-digestive diseases. The aim of this manuscript was to clarify the real clinical impact of LM on these disorders. METHODS: A literature search for digestive and extra-digestive disorders related to LM was carried out using PubMed, Medline and Cochrane. RESULTS: A transient lactase deficiency is present in celiac disease (CD) on a normal diet. The persistence of symptoms in CD on a gluten-free diet may be instead, in part, attributed to a primary LM. Similar circumstances are prese

Role of gastric emptying in functional dyspepsia: a scintigraphic study of 94 subjects.

Many physiological factors can alter gastric emptying, and the role of gastric emptying in functional dyspepsia is controversial. The aim of this study was to assess the effects of different factors (age, sex, the degree of dyspeptic and irritable bowel symptoms, lactase deficiency, smoking habits, the use of antiinflammatory drugs, and H. pylori gastritis) on gastric emptying in patients with functional dyspepsia. The study population consisted of 83 patients with functional dyspepsia and 11 control subjects who underwent a standardized scintigraphic examination to study gastric emptying. Thi

Effects of dietary gluten on the hepatotoxic action of galactosamine and/or endotoxin in rats.

This study was done to clarify the effects of dietary wheat gluten on the hepatotoxic action of D-galactosamine (GalN) and endotoxin (Etx). Male Wistar rats fed a high casein or high gluten (supplemented with L-Lys and L-Thr) diet were injected with GalN or Etx, and the plasma glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, and lactase dehydrogenase activities were examined 20 h later. In rats fed the high gluten diet, these enzyme activities were lower than in the high casein group after injection of 800 mg/kg of GalN. But such a difference between the casein and gluten

[Enzymes and histology of the intestinal mucosa of breast-fed African infants].

Weight gain and statural growth are slow in the Kivu area (Zaïre), as compared with international reference curves. One hypothesis put forward to explain this well-documented but poorly understood fact is inadequate breast-feeding with early intake of foods responsible for the introduction of microorganisms with deleterious effects on the digestive tract. A study of intestinal mucosa specimens from 90 at least partially breast-fed infants (4 to 22 months) showed that the appearance of the jejunal mucosa was consistently abnormal, as compared with histologic standards used in Europe. [Adaptation of rats, after weaning, to a diet with a high concentration of lactose. I. Evaluation of water and diet ingestion].

Twelve female Wistar rats, weaned at 21 days, were allocated in two groups: Lactose Group composed by six rats receiving a normal laboratory diet added with 25 g of lactose for each 100 g of final mixture, and the Saccharose Group that was fed with the same diet but with the lactose being replaced by saccharose in the same proportion; both groups were studied for 28 days. The rats were weighed at the beginning of the experiment and on the 7th, 14th, 21st and 28th days; the relative ingestion of water and diet was evaluated for each animal on the same days except for the first. Effect of Strongyloides ratti on small bowel function in normal and immunosuppressed host rats.

Although Strongyloides stercoralis is a common parasite, little is known about its effect on intestinal function. Published clinical studies are difficult to evaluate and compare because of the inability to differentiate the effects of the parasite load from that of various other coexisting features such as bacterial overgrowth, multiparasitism, malnutrition, or tropical sprue. Using a rat model where these problems do not occur, we found that Strongyloides ratti did not inhibit intestinal function in the healthy rat. In fact, in normal rats S. ratti appeared to increase ileal sucrase activity

Dairy Intake and Parkinson's Disease: A Mendelian Randomization Study.

BACKGROUND: Previous prospective studies highlighted dairy intake as a risk factor for Parkinson's disease (PD), particularly in men. It is unclear whether this association is causal or explained by reverse causation or confounding. OBJECTIVE: The aim is to examine the association between genetically predicted dairy intake and PD using two-sample Mendelian randomization (MR). METHODS: We genotyped a well-established instrumental variable for dairy intake located in the lactase gene (rs4988235) within the Courage-PD consortium (23 studies; 9823 patients and 8376 controls of European ancestry)

Severe lactose intolerance in a patient with coronary artery disease and ischemic cardiomyopathy.

A 72-year-old man with severe lactose intolerance was admitted for non-ST-segment elevation myocardial infarction. The coronary angiogram revealed occlusion of the distal third of the first diagonal artery and several non-significant lesions. The pre-discharge echocardiogram revealed moderate left ventricular systolic dysfunction. Discharged on dual antiplatelet therapy, rosuvastatin, perindopril and carvedilol, he was repeatedly readmitted in the following days for abdominal pain/bloating, diarrhea and nausea despite avoiding food products containing lactose. To date, there has been no compre
