The pharmacokinetics and pharmacodynamics of micafungin in experimental hematogenous Candida meningoencephalitis: implications for echinocandin therapy in neonates.

BACKGROUND: Hematogenous Candida meningoencephalitis (HCME) is a relatively frequent manifestation of disseminated candidiasis in neonates and is associated with significant mortality and neurodevelopmental abnormalities. The outcome after antifungal therapy is often suboptimal, with few therapeutic options. Limited clinical data suggest that echinocandins may have role to play in the treatment of HCME. METHODS: We studied the pharmacokinetics and pharmacodynamics of micafungin in a rabbit model of neonatal HCME and bridged the results to neonates by use of population pharmacokinetics and Mon

Bayesian inference of conformational state populations from computational models and sparse experimental observables.

We present a Bayesian inference approach to estimating conformational state populations from a combination of molecular modeling and sparse experimental data. Unlike alternative approaches, our method is designed for use with small molecules and emphasizes high-resolution structural models, using inferential structure determination with reference potentials, and Markov Chain Monte Carlo to sample the posterior distribution of conformational states. As an application of the method, we determine solution-state conformational populations of the 14-membered macrocycle cineromycin B, using a combin

Source apportionment and source-oriented health risk assessment of heavy metal(loid)s in soils of Zibo City, a typical industrial City in China: insights from APCS-MLR and Monte Carlo simulation.

Soil heavy metal(loid) pollution in industrial cities has become an increasingly concerning issue worldwide. The contamination status, source and health risk of Cr, Ni, Cu, Zn, As, Cd, Pb and Hg in the soils of Zibo city, a typical industrial city of China, were investigated. Results demonstrated that 2.33%, 1.16%, and 0.75% of samples exceeded the thresholds for Cu, Zn, and Cd, respectively. Cu, Zn, and Hg exhibited moderate pollution, and 1.50% of the sampling sites were classified as heavily polluted. According to the Influence Index of Comprehensive Quality (IICQ), the soil-vegetable syste

iProDNet: integrated probabilistic differential network inference under heterogeneous biological conditions.

Understanding phenotype-specific network rewiring is essential for elucidating the molecular mechanisms underlying complex diseases. Although numerous differential network analysis methods have been developed, most focus on only a limited aspect of network information. Consequently, these approaches often fail to jointly characterize regulatory activity and network architecture, provide limited statistical evidence at the network level, and offer little capability for prioritizing key genes that drive phenotype-specific network rewiring. To address these limitations, we developed a novel compu

Organ and Effective Dose Assessments Using MCNP with Computational PIMAL Phantoms and Realistic Eye Model in Radiological Workplaces.

The female and male computational phantom with moving arms and legs (PIMAL) are useful models for the calculation of the effective doses to workers in radiological occupational settings. In this study, Monte Carlo N-Particle (MCNP) Version 6.2 is used with PIMAL phantoms to simulate exposure in nuclear and medical settings. The PIMAL phantoms, defined by mathematical representation of the human body, allow for detailed assessment of equivalent dose to organs. These values are converted to effective dose (E) using sex- and age-average tissue weighting factors from ICRP Publication 103. Initial

A two-stage approach for segmenting spatial point patterns applied to multiplex imaging.

Recent advances in multiplex imaging have enabled researchers to locate different types of cells within a tissue sample. This is especially relevant for tumor immunology, as clinical regimes corresponding to different stages of disease or responses to treatment may manifest as different spatial arrangements of tumor and immune cells. Spatial point pattern modeling can be used to partition multiplex tissue images according to these regimes. To this end, we propose a two-stage approach: first, local intensities and pair correlation functions are estimated from the spatial point pattern of cells

Monte Carlo simulation of uveal melanoma treatment by ocular plaque brachytherapy using gold nanoparticles.

Uveal melanoma is a serious ocular disease that can be fatal. One of its treatment methods involves the use of radioactive seeds, which are temporarily attached to a plaque and placed on the eye. The dose distribution around the seeds used in ocular plaques strongly depends on the seeds' structure and the ocular plaque's design. Recently, there has been a high interest in using gold nanoparticles (GNPs) as a biocompatible and non-toxic dose enhancer in cancer treatment via ionizing radiation. This study used the Monte Carlo code GATE 9.1 to calculate the dose distribution in the tumour and the

Photon minibeam-based LATTICE radiotherapy for small and medium-sized tumors: A dosimetric planning study.

BACKGROUND: Lattice radiotherapy (LRT) is a spatially fractionated technique that delivers three-dimensional high-dose vertices within tumors. However, conventional LRT is constrained by geometric limitations when applied to small and medium-sized tumor volumes, primarily due to the large beam sizes that restrict optimal lattice pattern formation. PURPOSE: This study aims to investigate a photon minibeam-based LATTICE radiotherapy (mini-LRT) approach designed to overcome the geometric limitations of conventional LRT and enable effective spatially fractionated treatment for small and medium-si

Computational design of HLA class I superbinders for broad T cell immunogenicity.

Human leukocyte antigen (HLA) class I molecules are highly polymorphic, restricting peptide binding to narrow sequence subsets. Designing peptides that bind multiple HLA supertypes-termed superbinders-offers a promising strategy for broad-spectrum T cell vaccines and immunotherapies. Here, we present superHLA, a computational framework that combines Markov Chain Monte Carlo optimization with state-of-the-art major histocompatibility complex binding predictors to design synthetic 9-mer peptides with broad HLA-binding profiles. Using superHLA, we generated over 190,000 candidate superbinders pre

Cell survival comparison of proton, helium, and carbon ion interlaced minibeams in water phantom simulations.

BACKGROUND: Radiotherapy is a key in cancer treatment, with particle therapy providing better tumor targeting and sparing healthy tissues. Particle Minibeam Radiotherapy (PMBT) integrates the advantages of spatial fractionation into particle radiotherapy by employing submillimeter-sized beams, thus improving the therapeutic ratio by reducing side effects. Former simulation studies have shown that interlaced proton minibeams from opposing directions in Single Energy Distal-Edge (1E) mode better protect normal tissue compared to the conventional spread-out Bragg peak (SOBP) mode. ETODA: Automatic three-dimensional error tolerance grid generation for dosage adaptation in precision medicine.

BACKGROUND AND OBJECTIVE: Model-informed precision dosing (MIPD) relies on drug concentration measurements to individualize dosage regimens, yet emerging point-of-care (POC) technologies may introduce substantial measurement uncertainty compared with conventional laboratory assays. Existing frameworks lack systematic tools to evaluate how such inaccuracies propagate to dosage adaptation decisions and affect therapeutic outcomes across different sampling times. We propose ETODA (error tolerance of dosage adaptation), a computational framework that constructs automatic three-dimensional error-to

Global feasibility of cervical cancer elimination: a multidimensional scenario-based modelling analysis across 175 countries.

BACKGROUND: Cervical cancer remains a major cause of preventable cancer mortality worldwide. Progress towards elimination remains uneven, with persistent disparities in health system capacity, governance, and education. Existing models often rely on income classifications, overlooking structural determinants critical for success. We aimed to evaluate the global feasibility of cervical cancer elimination under structurally differentiated policy scenarios. METHODS: We developed a multidimensional scenario-based model integrating 270 structural indicators obtained from WHO, IARC, UN, World Bank,

Bayesian hierarchical models for multivariate mixed responses with repeated measures: A case study in arterial occlusive disease.

Modeling repeated measures of arterial occlusive diseases, such as peripheral artery disease (PAD), using data with mixed-type outcomes poses unique challenges due to complex dependency structures and diverse distributional assumptions. This study proposes a comprehensive Bayesian hierarchical modeling framework for the simultaneous analysis of binary and continuous outcomes observed repeatedly within individuals. We focus on the methodological comparison of three major Markov Chain Monte Carlo (MCMC) Bayesian computational methods-Metropolis-Hastings, Gibbs sampling, and Hamiltonian Monte Car

[Comparison of Measured Dose and Calculated Dose by Various Algorithms in an Early-stage Glottic Cancer Treatment Model].

PURPOSE: To evaluate the dose calculation accuracy in areas exposed to air on a radiotherapy treatment planning system using a phantom model simulating early-stage glottic cancer. METHODS: An air gap was placed between water-equivalent phantoms, and 6 MV X-rays were delivered at 200 MU. The absorbed dose in the depth direction was measured using Gafchromic film. These measurements were compared with calculations from Pinnacle3 (collapsed cone convolution: CCC) and RayStation (photon Monte Carlo: pMC, CCC). RESULTS: The difference between the measured film values after passing through the air

Neural parameter calibration for dengue outbreak forecasting.

Dengue fever poses a growing public health challenge in tropical and subtropical regions, with transmission driven by complex interactions among viral and host. Computational models, often expressed as ordinary differential equations (ODEs), are widely used to understand complex systems such as dengue fever transmission dynamics. However, traditional parameter estimation methods such as Markov chain Monte Carlo (MCMC) often require complex setups and are computationally expensive. In this study, we choose a compartmental model extended to human and mosquito populations, estimate its parameters

QSAR-based evaluation of plant-derived larvicidal agents against Zika vector Aedes aegypti via Monte Carlo optimization and SMILES-based descriptors and molecular docking.

Aedes aegypti is the principal vector responsible for the transmission of several arboviral diseases, including dengue, Zika, chikungunya, and yellow fever, posing a significant threat to global public health. Controlling its population is crucial to limiting the spread of these infections. Among the most effective strategies is the use of larvicides derived from plant sources, which interfere with the mosquito's development and reduce its proliferation. This study presents a QSAR analysis of 255 plant-based larvicidal compounds against Aedes aegypti (Zika vector), using pLC50 values as the me

Femoral neck anteversion alters patellofemoral joint mechanics during walking: insights from musculoskeletal modeling and Monte Carlo simulations.

Femoral neck anteversion (FNA) may lead to compensatory mechanisms during gait but the isolated influence of abnormal FNA on patellofemoral joint (PFJ) mechanics is not yet well understood. We used a detailed musculoskeletal model of the knee with 12 degrees of freedom. Seven models with different FNA, ranging from -8° to 52°, were developed by rotating the proximal part of the femur and the corresponding muscle attachment points along the mechanical axis of the femur, in the transverse plane. OpenSim COMAK and Monte Carlo simulations were used to estimate muscle force and patellar kinematics

Dosimetric impact of positional uncertainties and a robust optimization approach for rectal intensity-modulated brachytherapy.

BACKGROUND: Intensity-modulated brachytherapy (IMBT) employs rotating high-Z shields during treatment to decrease radiation in certain directions and conform the dose distribution to the target volume. Prototypes for dynamic IMBT have been proposed for prostate, cervical, and rectal cancer. PURPOSE: We considered two shielded applicators for IMBT rectal cancer treatment and investigated how rotational uncertainties in the shield angle and translational uncertainties in the source position affect plan evaluation criteria. METHODS: The effect of rotational errors of 3 ∘ $3^\circ$ , 5 ∘ $5^\
