Meningococcal resistance to antimicrobial peptides is mediated by bacterial adhesion and host cell RhoA and Cdc42 signalling.

Antimicrobial peptides (AMPs) constitute an essential part of the innate immune defence. Pathogenic bacteria have evolved numerous strategies to withstand AMP-mediated killing. The influence of host epithelia on bacterial AMP resistance is, however, still largely unknown. We found that adhesion to pharyngeal epithelial cells protected Neisseria meningitidis, a leading cause of meningitis and sepsis, from the human cathelicidin LL-37, the cationic model amphipathic peptide (MAP) and the peptaibol alamethicin, but not from polymyxin B. Adhesion to primary airway epithelia resulted in a similar i

Platelet endothelial cell adhesion molecule-1 in neutrophil emigration during acute bacterial pneumonia in mice and rats.

Platelet endothelial cell adhesion molecule-1 (PECAM-1) (CD31) is an adhesion molecule believed to mediate transendothelial migration of neutrophils and other leukocytes after CD11/CD18-mediated adhesion. Our study evaluated the role of PECAM-1 in neutrophil emigration across the pulmonary capillaries and the bronchial microvasculature using blocking anti-PECAM-1 antibodies in mice and rats. Neutrophil emigration was induced by Escherichia coli, a stimulus eliciting CD11/CD18-dependent emigration, or Streptococcus pneumoniae, a stimulus inducing CD11/CD18-independent emigration. Although anti-

Targeting bacterial adhesion: Recent advances and therapeutic potential of FimH and PapG inhibitors against Escherichia coli infection.

The widespread use and abuse of antibiotics have led to an increase in antibiotic resistance, and new methods and approaches are needed to combat bacterial infections. One promising alternative is to intervene early stage in the infection process. Adhesion is a prerequisite for bacteria to colonize on the cell surface. Various pathogenic Escherichia coli strains utilize carbohydrate-specific lectins to bind to specific glycoconjugate receptors on host cells for cell adhesion. The most important and best-studied bacterial lectins are FimH and PapG. Therefore, blocking the adhesion of bacteria t

Self-Patterned Nanoscale Topography of Thin Copolymer Films Prepared by Evaporative Assembly-Resist Early-Stage Bacterial Adhesion.

Biofilm formation on the surfaces of indwelling medical devices has become a growing health threat due to the development of antimicrobial resistance to infection-causing bacteria. For example, ventilator-associated pneumonia caused by Pseudomonas and Staphylococci species has become a significant concern in treatment of patients during COVID-19 pandemic. Nanostructured surfaces with antifouling activity are of interest as a promising strategy to prevent bacterial adhesion without triggering drug resistance. In this study, we report a facile evaporative approach to prepare block copolymer film…

Fish borne Edwardsiella tarda eha involved in the bacterial biofilm formation, hemolytic activity, adhesion capability and pathogenicity.

Edwardsiella tarda (E. tarda) is distributed widely in a variety of hosts including humans, other mammals and fish, and it is worthwhile to notice that E. tarda -caused fish infections lead to the most important bacterial disease in fish. Considering Eha acting as a transcriptional regulator in E. tarda strain ET13 have been reported previously, to better understand its pathogenesis due to this, a type of cell of epithelial cell line (Caco-2) infection model for the pathogen was established in the laboratory. We focused on studying various parameters such as lactate dehydrogenase release (to m

Interactions of bacteria with contact lenses: the effect of soluble protein and carbohydrate on bacterial adhesion to contact lenses.

PURPOSE: During contact lens wear, components in the tear film adsorb to the contact lens surface to form a protein-rich coating. In this study, we investigated the importance of these components in bacterial adhesion to contact lenses. METHODS: Five strains of bacteria were tested for their ability to adhere to contact lenses (patient-worn; coated with lactoferrin, lysozyme, or an artificial tear formulation; or left uncoated) in the presence of various solutes (protein, carbohydrate). Bacterial adhesion was quantified by both a total count (light microscopy) and viable count (culturing on a…

Impact of an alpha helix and a cysteine-cysteine disulfide bond on the resistance of bacterial adhesion pili to stress.

Escherichia coli express adhesion pili that mediate attachment to host cell surfaces and are exposed to body fluids in the urinary and gastrointestinal tracts. Pilin subunits are organized into helical polymers, with a tip adhesin for specific host binding. Pili can elastically unwind when exposed to fluid flow forces, reducing the adhesin load, thereby facilitating sustained attachment. Here we investigate biophysical and structural differences of pili commonly expressed on bacteria that inhabit the urinary and intestinal tracts. Optical tweezers measurements reveal that class 1a pili of urop

Down Syndrome Cell Adhesion Molecule Triggers Membrane-to-Nucleus Signaling-Regulated Hemocyte Proliferation against Bacterial Infection in Invertebrates.

Down syndrome cell adhesion molecule (Dscam) generates tens of thousands of isoforms by alternative splicing, thereby providing crucial functions during immune responses. In this study, a novel Dscam signaling pathway was investigated in crab, which remains poorly characterized in invertebrates. Bacterial infection induced the cytoplasmic cleavage of Dscam intracellular domains (ICDs) by γ-secretase, and then the released ICDs carrying specific alternatively spliced exons could directly interact with IPO5 to facilitate nuclear translocation. Nuclear imported ICDs thus promoted hemocyte prolife

Direct interaction between avian pathogenic Escherichia coli and H9N2 avian influenza virus promotes bacterial adhesion during their infections.

H9N2 subtype avian influenza virus (AIV) and avian pathogenic Escherichia coli (APEC) are prevalent pathogens in the poultry industry, and their co-infection results in synergistic pathogenicity in chickens and significant economic losses. However, the direct interactions between H9N2 AIV and APEC during infection remain unclear. We preincubated H9N2 AIV particles with APEC and found that H9N2 AIV could bind directly to the surface of APEC. The H9N2 AIV-APEC complex enhanced bacterial adhesion to chicken embryo fibroblasts and chicken macrophages (HD11), which could be inhibited by antiserum a…

Adhesion of Staphylococcus aureus to Candida albicans During Co-Infection Promotes Bacterial Dissemination Through the Host Immune Response.

Interspecies interactions greatly influence the virulence, drug tolerance and ultimately the outcome of polymicrobial biofilm infections. A synergistic interaction is observed between the fungus Candida albicans and the bacterium Staphylococcus aureus. These species are both normal commensals of most healthy humans and co-exist in several niches of the host. However, under certain circumstances, they can cause hospital-acquired infections with high morbidity and mortality rates. Using a mouse model of oral co-infection, we previously showed that an oral infection with C. albicans predisposes t

Cynaroside attenuates uropathogenic Escherichia coli adhesion and invasion by suppressing bacterial virulence and down-modulating urothelial Caveolin-1 signaling.

BACKGROUND: Uropathogenic Escherichia coli (UPEC) constitutes a primary pathogen responsible for urinary tract infections (UTIs), characterized predominantly by colonization initiated through adhesion, persistence via biofilm formation, and invasion facilitated by lipid rafts. Cynaroside (Cyn), a natural flavonoid, emerges as a potential anti-virulence compound; nonetheless, the involvement of Caveolin-1 (CAV1) in UPEC adhesion and invasion processes has yet to be fully elucidated. PURPOSE: To investigate the effects and mechanisms of Cyn on UPEC adhesion and invasion in bladder epithelial ce
