Effect of prolonged cryptorchidism on germ cell apoptosis and testicular sperm count.

AIM: To evaluate the long term effect of experimental cryptorchidism on germ cell apoptotic rate and testicular sperm content in adult rats. METHODS: Bilateral cryptorchidism was created in 40 adult male Sprague-Dawley rats by surgically manipulating the testes into the abdominal cavity and closing the internal inguinal ring. The rats were sacrificed and the testes removed 6 hours and 2, 4, 7, 21, 28 and 56 days after cryptorchidism. Germ cell apoptosis was quantified by means of TUNEL assay and apoptosis was confirmed using transmission electron microscopy. RESULTS: The rate of apoptosis pe

Correlation of platelet count changes with liver cell destruction in HELLP syndrome.

OBJECTIVE: HELLP syndrome is defined as a subset of preeclampsia involving a combination of hemolysis, low platelets affecting the circulatory system, and elevated liver enzymes, reflecting liver cell destruction. Our objective was to assess the correlation between the platelet count changes and liver cell destruction as indicated by aspartate aminotransferase (AST) levels. METHOD: We reviewed the records of 15 preeclamptic patients who developed HELLP syndrome as defined by 1) hemolysis, 2) elevated liver enzymes with an AST greater than 70 U/L (>2 SD above mean), and 3) low platelets (<100x

Testicular volume does not predict germ cell count in patients with cryptorchidism.

PURPOSE: A germ cell count of less than 0.2 germ cell per tubule on the prepubertal biopsy of cryptorchid testes predicts abnormal spermiograms and decreased fertility in adulthood, and may be used to select patients for post-orchiopexy hormonal therapy. Testicular volume directly correlates with testicular function and spermiogenesis. We determined whether testicular volume would predict the total germ cell count accurately enough to replace testicular biopsy in the modern management of cryptorchidism. MATERIALS AND METHODS: At our hospital 723 patients younger than 9 years with cryptorchidi…

Utility of Symptom Duration and C-Reactive Protein, White Blood Cell Count, and Absolute Neutrophil Count in the Evaluation of Pediatric Appendicitis.

BACKGROUND: Appendicitis is a common pediatric surgical emergency, and the diagnosis may be delayed or missed because of nonspecific findings in children. Not all patients with abdominal pain need to be imaged for appendicitis, and laboratory evaluation may improve diagnostic accuracy in this population. OBJECTIVE: To determine if C-reactive protein (CRP) and symptom duration could be used to improve diagnosis of appendicitis compared with white blood cell count (WBC) and absolute neutrophil count (ANC). METHODS: This was a retrospective chart review from June 2017 to 2019 at our tertiary ac…

Comparison of Blood Cell Count Results Between MgSO4 and K2EDTA. Stability and Accuracy of Platelet Counting With MgSO4.

INTRODUCTION: We investigated the stability and accuracy of platelets with MgSO4 and compared the results of blood counts between MgSO4 and K2EDTA. METHODS: Platelet stability as a function of time was assessed on the XN Sysmex analyzer by comparing the percentage deviation to the desirable bias of the EFLM. Accuracy was determined by calculating the LoQ of platelets corresponding to a CV of 10%. Results from 250 patients were compared between K2EDTA and MgSO4. RESULTS: At 20°C, platelets were stable for 24 h in impedance, 10 h in fluorescence and at 4°C for 4 h in impedance, 2 h in fluoresc

Mini-FLOTAC for counting Toxoplasma gondii oocysts from cat feces--comparison with cell counting plates.

Oocysts of Toxoplasma gondii represent one of the most common environmental contaminants causing the zoonotic infection toxoplasmosis. The aim of the present study was to compare the Mini-FLOTAC device with traditional cell counting plates (Kova Slide) for the detection of T. gondii oocysts from feline feces. Two types of experiments were performed: (i) purified oocysts were counted in different dilutions and (ii) specific pathogen free T. gondii-negative cat feces was inoculated with numbers of purified oocysts and counting was performed directly from feces. Our analysis showed a thousand tim…

Intra-apheresis CD34+ cell count: A dynamic approach to predicting peripheral blood stem cell collection yield.

BACKGROUND: Accurate prediction of peripheral blood stem cell (PBSC) yield is essential for optimizing hematopoietic stem cell collections. Traditional CD34+ prediction formulas often overlook the ongoing recruitment of CD34+ cells during the collection procedure. This study evaluated whether a single intra-apheresis CD34+ measurement could improve yield prediction compared to existing prediction models. METHODS: We retrospectively analyzed 150 PBSC collections from 123 autologous and allogenic donors. A single intraprocedural CD34+ measurement and final collection yield were obtained, and un

White blood cell count as a predictor of the severity of sickle cell disease during pregnancy.

OBJECTIVE: Increasing epidemiological evidence implicates leukocytosis as a major risk factor for poor outcome in non-pregnant sickle cell disease (SCD) patients. The aim of this study was to investigate whether steady-state white blood cell (WBC) count can predict SCD-related problems in pregnancy. DESIGN: Retrospective observational study of 40 SCD pregnant women who had their antenatal care at Guy's & St Thomas' NHS Trust, London. Twenty-six women developed clinical complications related to SCD during their pregnancy (painful crisis, chest symptoms, infections, severe anaemia requiring blo

CD38+ T-Cell Count Is an Immunologic Phenotype Associated With Blood Pressure.

BACKGROUND: T lymphocytes play a crucial role in the development of hypertension and associated end-organ damage. CD38 is a well-established surface marker for T-cell activation. However, clinical evidence linking CD38+ T cells, or other specific T-cell subsets, with blood pressure (BP) changes remains limited. We therefore sought to determine whether CD38+ T-cell abundance correlates with BP and whether anti-CD38 therapy influences BP in humans and mice. METHODS: We performed correlation analyses between peripheral immune cell counts and BP in 197 normotensive and 53 hypertensive subjects. T…

Lateral flow assay-based CD4 cell count testing for advanced HIV disease.

The global policy shift to treating all people living with HIV, regardless of CD4 cell count, has inadvertently led donors and national programmes to reduce their investments in CD4 testing. The subsequent decline in testing volumes has caused manufacturers to discontinue major point-of-care CD4 testing instruments, despite these tests being crucial for the diagnosis of advanced HIV disease (AHD). Mortality from AHD remains high, with an estimated 630 000 deaths among people living with HIV in 2023. CD4 lateral flow assay (LFA) testing could provide pragmatic screening for AHD. Published studi…

AxoNet: A deep learning-based tool to count retinal ganglion cell axons.

In this work, we develop a robust, extensible tool to automatically and accurately count retinal ganglion cell axons in optic nerve (ON) tissue images from various animal models of glaucoma. We adapted deep learning to regress pixelwise axon count density estimates, which were then integrated over the image area to determine axon counts. The tool, termed AxoNet, was trained and evaluated using a dataset containing images of ON regions randomly selected from whole cross sections of both control and damaged rat ONs and manually annotated for axon count and location. This rat-trained network was

Quantitative analysis of mast cell count and density in chronic periodontal disease.

BACKGROUND: Mast cells play a crucial role in activation of acquired immune response to inflammatory conditions of periodontal diseases. They promote inflammation by releasing pro-inflammatory mediators and bring about angiogenesis, degeneration of the extracellular matrix, and tissue remodeling. Since there is little literature regarding the role of mast cells in periodontitis, the present study was aimed to evaluate mast cell count (MCC) and density in periodontitis. MATERIALS AND METHODS: A total of eighty participants, Group I (n = 40) healthy participants and Group II (n = 40) participan…

Validity of somatic cell count as indicator of pathogen-specific intramammary infections.

The objective of this study was to determine whether somatic cell count (SCC) was an effective test, with a sensitivity exceeding 85%, to determine species-specific bacterial infections. In addition, the relation between the SCC and various udder pathogen groups was investigated. SCC thresholds of greater than 200 000 cells/mL were used in quarter and greater than 150 000 cells/mL in composite milk samples. A retrospective study was conducted on a data set for 89 635 quarter and 345 467 composite cow milk samples. Eleven SCC threshold values were used to evaluate the diagnostic efficacy for th…

Bronchoalveolar lavage total cell count in interstitial lung diseases--does it matter?

Bronchoalveolar lavage (BAL) is a useful technique for differential diagnosis of various interstitial lung diseases (ILDs) and is usually realized by analysis of the differential cell count. This study was conducted to estimate the value of bronchoalveolar lavage fluid (BALF) total cell count (TCC) in the diagnosis of ILD. We analyzed 237 BAL samples from patients with ILD: sarcoidosis (SA), idiopathic pulmonary fibrosis (IPF), cryptogenic organizing pneumonia (COP), hypersensitivity pneumonitis (HP), chronic eosinophilic pneumonia (CEP), and smoking-related ILD (sr-ILD). The control group con…

The value of white blood cell count in patients with swollen discs.

BACKGROUND: A broad differential diagnosis has to be considered in a patient with swollen discs. Myeloproliferative disorders such as leukemia and lymphoma can in rare cases cause infiltrative optic neuropathy. HISTORY AND SIGNS: Two patients initially presented with slowly progressive severe visual loss. History was unremarkable except for previously noted slightly elevated white blood cell count for which - according to their general physicians - no treatment or work-up was required. At presentation, bilateral disc swelling was present. Magnetic resonance imaging showed enhancement of the e…
