Using a phylogenetic framework to characterize natural selection, we investigate the hypothesis that zoonotic viruses require adaptation prior to zoonosis to sustain human-to-human transmission. Examining the zoonotic emergence of Ebola virus, Marburg virus, influenza A virus, SARS-CoV, and SARS-CoV-2, we find no evidence of a change in the intensity of natural selection immediately prior to a host switch, compared with typical selection within reservoir hosts. We conclude that extensive pre-zoonotic adaptation is not necessary for human-to-human transmission of zoonotic viruses. In contrast,

Yersinia pestis has spilled over from wild rodent reservoirs to commensal rodents and humans, causing three historically recorded pandemics. Depletion in the copy number of the plasmid-encoded virulence gene pla occurred in later-dated strains of the first and second pandemics, yet the biological relevance of the pla deletion has been difficult to test. We identified modern Y. pestis strains that independently acquired the same pla depletion as ancient strains and herein show that excision of pla from the multicopy pPCP1 plasmid is accompanied by the integration of a separate full pPCP1 harbor

OBJECTIVES: The outbreak of the COVID-19 pandemic has had wide-ranging outcomes on adolescents' well-being. However, less attention has been paid to the adolescent's mental health during the pandemic. The pandemic may impair adolescents' mental health through stress spillover from other family members, contextual and policy changes, and the disruption of everyday life routines. Therefore, our research is motivated by a need to address the relative scarcity of research examining adolescent mental health during the pandemic.

The role of extracorporeal membrane oxygenation (ECMO) in the management of severe acute respiratory failure, including acute respiratory distress syndrome, has become better defined in recent years in light of emerging high-quality evidence and technological advances. Use of ECMO has consequently increased throughout many parts of the world. The coronavirus disease (COVID-19) pandemic, however, has highlighted deficiencies in organizational capacity, research capability, knowledge sharing, and resource use. Although governments, medical societies, hospital systems, and clinicians were collect

Ebola virus is among the most dangerous, contagious and deadly etiological causes of viral diseases. However, Ebola virus has never extensively spread in human population and never have led to a pandemic. Why? The mechanistic biophysical model revealing the biothermodynamic background of virus-host interaction) could help us to understand pathogenesis of Ebola virus disease (earlier known as the Ebola hemorrhagic fever). In this paper for the first time the empirical formula, thermodynamic properties of biosynthesis (including the driving force of virus multiplication in the susceptible host),

Ever since the devastating 1918-1919 influenza pandemic, policy makers have employed mathematical models to predict the course of epidemics and pandemics in an effort to mitigate their worst impacts. But while Britain has long been a pioneer of predictive epidemiology and disease modellers occupied influential positions on key committees that advised the government on its response to the coronavirus pandemic, as in 1918 Britain mounted one of the least effective responses to Covid-19 of any country in the world. Arguing that this 'failure of expertise' was the result of medical and political c

BACKGROUND: The current COVID-19 pandemic is not the first time New South Wales prisons have faced contagion. This paper examines the current responses in New South Wales prisons to the threat of COVID-19 to prisoner health, by contrasting contemporary activities with actions and policy developed during two historical epidemics: the influenza epidemic of 1860 and pandemic of 1919.
METHOD: Epidemiological information relating to cases of disease in NSW prisons during the 1860 and 1919 influenza epidemics was obtained from the Comptroller-General's reports for the specific outbreak years and fo

The outbreak of COVID-19 revealed the vulnerability of commercially available face masks. Without having antibacterial/antiviral activities, the current masks act only as filtering materials of the aerosols containing microorganisms. Meanwhile, in surgical masks, the viral and bacterial filtration highly depends on the electrostatic charges of masks. These electrostatic charges disappear after 8 h, which leads to a significant decline in filtration efficiency. Therefore, to enhance the masks' protection performance, fabrication of innovative masks with more advanced functions is in urgent dema

Pandemics such as influenza, smallpox, and plague have caused the loss of hundreds of millions of lives and have occurred for many centuries. Fortunately, they have been largely eliminated by the use of vaccinations and drugs. More recently, Severe Acute Respiratory Syndrome (SARS), Middle East Respiratory Syndrome (MERS), and now Coronavirus Disease 2019 (COVID-19) have arisen, and given the current absence of highly effective approved vaccines or drugs, brute-force approaches involving physical barriers are being used to counter virus spread. A major basis for physical protection from respir

When confronted by the novel ethical challenges posed by a pandemic, it is helpful to turn to history for guidance and direction. In this essay, the author revisits Thucydides's description of the Plague of Athens from The Peloponnesian War as he considers the New York State Task Force on Life and the Law's 2015 guidelines on ventilator allocation. Confronted by the exigencies of the Covid-19 surge that struck New York, he questions the task force's decision not to give any degree of preference to health care workers who might become ill. He posits that they are due a compensatory ethic and so

In the twenty-first century, severe acute respiratory syndrome (SARS), 2009 A(H1N1) influenza, and Ebola have all placed strains on critical care systems. In addition to the increased patient needs common to many disasters, epidemics may further degrade ICU capability when staff members fall ill, including in the course of direct patient care. In a large-scale pandemic, shortages of equipment and medications can further limit an ICU's ability to provide the normal standard of care. Hospital preparedness for epidemics must include strategies to maintain staff safety, secure adequate supplies, a

INTRODUCTION: We aimed to develop tools to ease the burden on medical staff while improving patient care. The central question is whether Support Vector Machines (SVMs), trained solely on patients' self-reported data (symptoms, demographics, comorbidities), can achieve triage accuracy comparable to or surpassing traditional score-based systems in emergency departments under pandemic conditions.
METHODS: We propose an SVM-based approach to replicate COVID-19 triage using patient-provided information rather than laboratory tests. The system automates triage scoring, classifying patients into re

OBJECTIVE: Hospitals play a crucial role in responding to respiratory pandemics, yet the degree of their readiness varies widely across health systems. Identifying and prioritizing the most critical indicators of hospital preparedness can guide policymakers in resource allocation and strategic planning. This study aimed to determine and rank the key indicators influencing hospital readiness for respiratory pandemics in referral hospitals in Kerman Province, Iran.
METHODS: A mixed-method, multicriteria decision-making (MCDM) approach was applied. Initially, a semi-structured Delphi process inv

BACKGROUND: Many studies examined the impact of behavioural interventions on COVID-19 outcomes. We conducted a systematic review to gain insight into transmission models, following PRISMA 2020 guidelines. We included peer-reviewed studies published in English until December 31, 2022, focusing on human subjects, modelling, and examining behavioural interventions during COVID-19 using real data across diverse geographical regions.
METHODS: We searched seven databases. We used descriptive analysis, network analysis for textual synthesis, and regression analysis to identify the relationship betwe

The COVID-19 pandemic revealed the urgent need for faster and more accessible drug discovery for future coronavirus outbreaks. We participated in the Polaris Antiviral Drug Discovery 2025 competition to evaluate how AI can accelerate this process, comparing three molecular docking approaches: AutoDock Vina (physics-based), GNINA (deep learning-assisted), and Boltz-2 (data-driven), using SARS-CoV-2 and MERS-CoV data sets. Our results demonstrate that AI has transformed drug discovery: Boltz-2 reached over 80% accuracy, greatly improving docking precision. However, accurate prediction of binding

In recent decades, the world has gained experience of the dangerous effects of pandemic events caused by emerging respiratory viruses. In particular, annual epidemics of influenza are responsible for severe illness and deaths. Even if conventional influenza vaccines represent the most effective tool for preventing virus infections, they are not completely effective in patients with severe chronic disease and immunocompromised and new small molecules have emerged to prevent and control the influenza viruses. Thus, the attention of chemists is continuously focused on the synthesis of new antivir

Throughout history, pandemics of infectious diseases caused by emerging viruses have spread worldwide. Evidence from previous outbreaks demonstrated that pregnant women are at high risk of contracting the diseases and suffering from adverse outcomes. However, while some viruses can cause major health complications for the mother and her fetus, others do not appear to affect pregnancy. Viral surface proteins bind to specific receptors on the cellular membrane of host cells and begin therewith the infection process. During pregnancy, the molecular features of these proteins may determine specifi
