Cementum proteins: role in cementogenesis, biomineralization, periodontium formation and regeneration.

Destruction of the periodontium is normally associated with periodontal disease, although many other factors, such as trauma, aging, infections, orthodontic tooth movement and systemic and genetic diseases, can contribute to this process. Strategies (such as guided tissue regeneration) have been developed to guide and control regeneration using bioresorbable membranes and bone grafts. Although effective to a certain point, these strategies have the problem that they are not predictable and do not completely restore the architecture of the original periodontium. To achieve complete repair and r…

Wnt5a regulates dental follicle stem/progenitor cells of the periodontium.

INTRODUCTION: Dental follicle gives rise to one or several tissues of the periodontium including the periodontal ligament, cementum and/or alveolar bone. Whether Wnt5a is expressed in the postnatal periodontium or regulates dental follicle stem/progenitor cells is unknown. METHODS: Dental follicle stem/progenitor cells were isolated from postnatal day 1 (p1) to p11 from rat mandibular first molars. Immunolocalization mapped Wnt5a expression in the alveolar bone, periodontal ligament, and the developing ameloblast and odontoblast layers. Mononucleated and adherent cells were isolated from p7 d…

Neutrophils in the periodontium: Interactions with pathogens and roles in tissue homeostasis and inflammation.

Neutrophils are of key importance in periodontal health and disease. In their absence or when they are functionally defective, as occurs in certain congenital disorders, affected individuals develop severe forms of periodontitis in early age. These observations imply that the presence of immune-competent neutrophils is essential to homeostasis. However, the presence of supernumerary or hyper-responsive neutrophils, either because of systemic priming or innate immune training, leads to imbalanced host-microbe interactions in the periodontium that culminate in dysbiosis and inflammatory tissue b…

Influence of Female Sex Hormones in Different Stages of Women on Periodontium.

The primary etiologic factor for periodontal diseases is "Dental plaque." Although pathogenic bacteria in dental plaque are required for the incidence of periodontal disease, a susceptible host is also very important. The susceptibility of the host can be modified by many systemic factors with hormones level being one. The periodontium shows an exaggerated inflammatory response to plaque modified by female sex hormones during puberty, pregnancy, in women taking oral contraceptives, and at the postmenopausal stage. This review provides an in detail analysis of how periodontium is influenced by …

ACVR1 is essential for periodontium development and promotes alveolar bone formation.

OBJECTIVE: To explore the role of a BMP type I receptor (ACVR1) in regulating periodontium development, Acvr1 was conditionally disrupted in Osterix-expressing cells. METHODS: Mandibles from both control (Acvr1 fx/+; Osterix-Cre (+)/(-)) and cKO (Acvr1 fx/-; Osterix-Cre (+)/(-)) mice at postnatal day 21 (PN21) were scanned by micro-CT, followed by decalcification and histological observations. Distributions and levels of differentiation markers of fibroblasts, osteoblasts and cementocytes in the periodontium were detected by immunohistochemical (IHC) staining. RESULTS: Micro-CT results showe…

Citrullination in the periodontium--a possible link between periodontitis and rheumatoid arthritis.

OBJECTIVES: The aim of the present study was to assess human and bacterial peptidylarginine deiminase (PAD) activity in the gingival crevicu … on with high antibody levels against that species. CONCLUSIONS: PAD and PPAD activities within the periodontium are elevated in RA and non-RA patients with periodontitis. PPAD secreted by P. gingivalis residing in epithelial cells may exert its citrullinating activity in distant regions of the periodontium or even distant tissues. CLINICAL RELEVANCE: In peri …

[Does good oral hygiene guarantee the maintenance of a health periodontium?].

On the last European Workshop for Periodontology, it was accepted that the prevalence of periodontitis in certain regions of Europe and USA has decreased. It remains difficult to phrase a decision concerning the prevalence of periodontitis in general. This article wants to highlight the need of a good oral hygiene and different forms of prevention (primary, secondary & tertiary) in the maintenance of a healthy periodontium. The relationship between gingivitis and tooth loss is pointed out. The prevention is described in the complexity of the periodontitis proces and it's modifying factors. Whe…

[The local immune mechanisms of the involvement of the teeth and periodontium in periodic disease].

The aim of our investigations was to elucidate some immune aspects of combination of caries and periodontitis with periodic disease (PD), also known as familial Mediterranean fever. In this regard in patients with active and non-active stage of PD we have studied dynamic changes of concentration of secretory immunoglobulin A (SIgA) in saliva and phagocytic activity of neutrophils derived from gum blood. It has been shown that in patients with PD these indices of local immunity of oral cavity had tendency to a decrease especially in case of PD and periodontitis combination. Disturbances of loca

Calcium oxalate deposition in the periodontium secondary to chronic renal failure.

Deposition of calcium oxalate in the periodontium with accompanying bone loss and tooth resorption, occurred in a 55-year-old woman on long-term renal dialysis for end-stage renal disease. These deposits were birefringent upon examination with polarized light. Scanning electron microscopy of the deposits revealed clusters of sharply angular oblong and needlelike crystals. Microincineration followed by staining with alizarin red S was positive for calcium oxalate. X-ray diffraction studies of the crystals in the tissue specimen revealed a pattern identical to that of commercially prepared calci…

Changes of the marginal periodontium as a result of labial tooth movement in monkeys.

Tooth position has been suggested to be an important factor in gingival recession. Due to conflicting reports in the literature, this study was undertaken to examine the effects of labial tooth movement on changes in the marginal periodontium. Orthodontic tooth movement was performed in five monkeys (Macaca nemistrina). Oral health was established and exploratory surgery was performed to assess the level of the connective tissue attachment and marginal bone. Measurements of the gingival margin and mucogingival junction were taken and orthodontic forces were applied. The central incisors were m…

Deteriorating effect of occlusal disorders on the periodontium of rats with experimental arteriosclerosis.

The periodontal effects of food consistency and of experimental occlusal stresses were clinically studied with healthy and arteriosclerotic rats. Experimental arteriosclerosis was induced with a hypercholesterolemic diet continued for 6 -- 12 months. By using food that was finely powdered and moistened, mechanical irritation of the periodontium was reduced to a minimum. The special occlusal and gingival irritants lasting six weeks were an occlusal overload with an overhigh amalgam filling, or an occlusal hypofunction caused by extracting the antagonist tooth. The arteriosclerotic animals with …

Gingiva and periodontium in lepromatous leprosy. A clinical, radiological, and microscopical study.

Thirty patients with lepromatous leprosy of long duration were examined for gingival and periodontal changes. All patients presented with chronic gingivitis and periodontitis of an unspecific type. Etiologic factors affecting the development of gingival and periodontal changes in this disease are: lack of oral hygiene with severe plaque and calculus formation due to the inability of the patients to perform oral hygiene, mouth breathing, specific granulomatous infiltrations, and a possible specific drug action. Specific changes in form of 'facies leprosa' are not pathognomonic of lepromatous le
