Chromosome painting was employed to measure frequencies of reciprocal translocations in human lymphocytes induced by 60Co gamma rays, with emphasis on low doses. Translocations and dicentrics were distinguished by use of a pan-centromeric probe. A total of 41,151 metaphases were analyzed at doses below 0.2 Gy. We present here an alternative approach to the study of mosaic cell lines containing dicentric chromosomes. The approach is based on chromosome-specific non-radioactive in situ hybridization with centromere (alpha satellite DNA) probes. We report on a fetus with ring chromosome 18 and monosomy 18 mosaicism [mos 45,XX,-18/46,XX,r(18)] detected in cultured amniotic fluid cells at 15 weeks' gestation. Chromosome analysis of fetal blood, cultured chorionic villi and fetal fibroblasts confirmed the aberrant karyotype. The aberrant chromosome complement could not be detected in a short-term culture of chorionic villi (46,XX in 21 metaphases).
