The effect of amorphous pyrophosphate on calcium phosphate cement resorption and bone generation.

Pyrophosphate ions are both inhibitors of HA formation and substrates for phosphatase enzymes. Unlike polyphosphates their hydrolysis results simultaneously in the complete loss of mineral formation inhibition and a localised elevation in orthophosphate ion concentration. Despite recent advances in our knowledge of the role of the pyrophosphate ion, very little is known about the effects of pyrophosphate on bone formation and even less is known about its local delivery. In this work we first developed a self setting pyrophosphate based calcium cement system with appropriate handling properties

Pyrovanadolysis, a pyrophosphorolysis-like reaction mediated by pyrovanadate, Mn2+, and DNA polymerase of bacteriophage T7.

DNA polymerases catalyze the 3'-5'-pyrophosphorolysis of a DNA primer annealed to a DNA template in the presence of pyrophosphate (PP(i)). In this reversal of the polymerization reaction, deoxynucleotides in DNA are converted to deoxynucleoside 5'-triphosphates. Based on the charge, size, and geometry of the oxygen connecting the two phosphorus atoms of PP(i), a variety of compounds was examined for their ability to carry out a reaction similar to pyrophosphorolysis. We describe a manganese-mediated pyrophosphorolysis-like activity using pyrovanadate (VV) catalyzed by the DNA polymerase of bac

Amino acid dependent formation of phosphate anhydrides in water mediated by carbonyl sulfide.

Carbonyl sulfide (COS), a component of volcanic gas emissions and interstellar gas clouds, is shown to be an efficient condensing agent in the context of phosphate chemistry in aqueous solutions. We report that high-energy aminoacyl-phosphate anhydrides and aminoacyl adenylates are generated in solutions containing amino acids, COS, and the corresponding phosphate molecule. We further show that the mixed anhydrides of amino acids and inorganic phosphate are phosphorylating agents, producing pyrophosphate in better than 30% yield in the presence of Ca2+ precipitates. The amino acid dependent ac

Using electrophoresis to observe the interaction of nitrogenase with ions.

The two protein components of nitrogenase from Klebsiella pneumoniae were shown to interact with metal ions and ADP, altering their electrophoretic mobility in polyacrylamide gel electrophoresis. Both Mg+2 and Mn+2 caused reduced mobility of Fe protein relative to other proteins. The effect was about 50% complete at concentrations around 0.2 mM. Other ions including Fe+2, Ni+2 and Co+2 had no observable effect at levels up to 1 _mM. Both Cd+2 and Zn+2 appeared to interact with the protein; Cd+2 at 0.5 mM dramatically destabilized the protein. The effects of more than a dozen different mutation

Effect of oxygen on the respiratory system and cephalosporin-C production in Acremonium chrysogenum.

In this paper in vivo studies on the overall and cyanide-resistant respiration of Acremonium chrysogenum in connection with different oxygen transfer rates are discussed. Two parameters of the cyanide-resistant respiration, the activity and the capacity were examined separately during cultivation together with the possibly respiration-influencing factors like NADH/NAD content and ADP-limitation. It was finally concluded that the cyanide-resistant respiration seems to be independent of the amount and proportion of oxidized/reduced coenzymes and not limited by ADP or phosphate, but it is strongl

Phosphoinositide signalling system in human erythrocyte and its role in cell pathology.

Human erythrocytes contain 217.2 +/- 70.5 pmole of inositol 1,4,5-trisphosphate per ml of packed cells (n = 14). The increased generation of Ins 1,4,5P3, was induced by 1 microM A 23187, 20 microM Pb2+, and by AlF4-. Ins 1,4,5P3, Pb2+ and AlF4- evoke shape changes, disorganisation of spectrin network and vesiculation. In erythrocytes of patients with hereditary spherocytosis the level of Ins 1,4,5P3 was increased to 556.7 +/- 374 pmole per ml of packed cells. We suggest that activation of the phosphoinositide signalling system may represent a common denominator for many divergent stimuli and d

Potentiation by red blood cells of shear-induced platelet aggregation: relative importance of chemical and physical mechanisms.

Evidence has been reported to indicate that red blood cells (RBCs) may potentiate platelet adherence and platelet aggregation (PAG) in different flow systems in vitro as well as hemostatic platelet plug formation in response to vascular injury. In this study, we demonstrate that RBCs enhance PAG induced by well-defined, low-intensity, uniform, laminar shear stress. Potentiation by RBCs of shear-induced PAG was associated with appreciable loss of adenine nucleotides from 14C-adenine-labeled RBCs, the extent of which increased with increasing RBC concentration. The concentrations of RBC-derived

Metabolite control of L-fucose utilization.

Thyroid fucokinase is responsive to a number of metabolites which might serve in a regulatory capacity. In addition to inhibition by ADP and stimulation by GMP, fucokinase responds selectively to a series of nucleotide sugars. Of those studied, only guanine nucleotide sugars moderate the activity of the enzyme. GDP-alpha-D-mannose, GDP-alpha-D-glucose, GDP-alpha-D-rhamnose, and GDP-alpha-L-fucose on the other hand is strongly inhibitory. In the case of GDP-alpha-D-mannose stimulation, a physiological role seems possible, but the rationale is not entirely clear. The effects of GDP-beta-L-fucose

Regulation of glibenclamide-sensitive K+ current by nucleotide phosphates in isolated rabbit pulmonary myocytes.

OBJECTIVE: Although the existence of ATP-sensitive K+ (KATP) channels in vascular muscle is widely accepted, there appears to be little consensus as to what the primary regulator of these channels is under physiological or pathophysiological conditions. Recent evidence has suggested that nucleotide diphosphates (NDPs) may play a more important role than ATP. However, since the properties of vascular KATP channels are quite diverse, and the effects of these nucleotides are poorly understood, the aim of this study was to test the hypothesis that intracellular ATP can regulate whole-cell KATP cur…

Postexposure Liponucleotide Prophylaxis and Treatment Attenuates Acute Respiratory Distress Syndrome in Influenza-infected Mice.

There is an urgent need for new drugs for patients with acute respiratory distress syndrome (ARDS), including those with coronavirus disease (COVID-19). ARDS in influenza-infected mice is associated with reduced concentrations of liponucleotides (essential precursors for de novo phospholipid synthesis) in alveolar type II (ATII) epithelial cells. Because surfactant phospholipid synthesis is a primary function of ATII cells, we hypothesized that disrupting this process could contribute significantly to the pathogenesis of influenza-induced ARDS. The goal of this study was to determine whether p

X-ray structure of the magnesium(II)-pyrophosphate complex of the truncated head of Dictyostelium discoideum myosin to 2.7 A resolution.

The structure of the magnesium pyrophosphate complex of the truncated head of Dictyostelium myosin has been determined by molecular replacement at 2.7 A resolution and refined to a crystallographic R-factor of 16.0%. The crystals belong to the orthorhombic space group P2(1)2(1)2, where a = 105.2 A, b = 182.1 A, and c = 54.5 A. The conformation of the protein around the magnesium pyrophosphate is very similar to that seen when magnesium ADP-beryllium fluoride binds in the active site. The latter complex mimics the binding of ATP prior to hydrolysis. The pyrophosphate molecule occupies the beta-

Nm23 protein expression in ductal in situ and invasive human breast carcinoma.

BACKGROUND: Mortality associated with human breast carcinoma is almost entirely due to subsequent metastatic disease, but the molecular basis of this metastasis is not understood. Elucidation of the genetic control of metastatic propensity of a tumor is important in determining prognosis and choice of therapy. Expression of nm23, a putative metastasis suppressor gene, has been detected in human breast cancers, but studies have not consistently shown high levels of the Nm23 messenger RNA or protein to be associated with better histological differentiation. This inconsistency suggests that Nm23

Metabolic Engineering of Yarrowia lipolyticafor Efficient De Novo Synthesis of Astaxanthin Glucosides via Enhanced UDP-Glucose Supply.

Astaxanthin is a high-value carotenoid used in nutrition, cosmetics, pharmaceuticals, and biotechnology, but its poor water dispersibility and oxidative stability limit its formulation. Here, we engineered yeast Yarrowia lipolytica for the efficient production of glycosylated astaxanthin. Introducing zeaxanthin glucosyltransferase CrtX from Pantoea ananatis into an astaxanthin-producing strain enabled the synthesis of astaxanthin glucosides, confirmed by HR-ESI-MS, 1H NMR, and 2D NMR. Engineering UDP-glucose metabolism by reducing competing pathways and enhancing UDP-glucose biosynthesis impro

[The effectiveness of citicoline in the treatment of cognitive disorders].

Randomized controlled trials, systematic reviews, and meta-analyses have shown that citicoline is highly effective in the treatment of vascular cognitive impairments (CI) of various origins (post-stroke, chronic cerebral ischemia (CCI)), post-traumatic CI, mild CI in neurodegenerative diseases (Alzheimer's disease, Parkinson's disease), and in patients with age-related memory impairments. Citicoline administration regimens: 1) post-stroke CI at a dose of 1000 mg/day. for a course of 4-12 months; 2) mild CI of vascular origin at a dose of 500 mg/day for a course of 3-9 months; 3) CI against the

Alzheimer's Disease, Piezo2 Channelopathy, Piezo1 Channelopathy, and the Body-Wide Piezo2 System.

Alzheimer's disease initiates pathophysiology in the 10 to 20 years prior to detectable clinical symptoms. A recent genetic analysis implicated the critical role of Piezo2 in Alzheimer's disease pathophysiology. A recent genetic study, involving PIEZO1 manipulation, showed that phosphatidylinositol 4,5-bisphosphate (PIP2) rectified brain capillary endothelial Piezo1 channelopathy in a mouse model of Alzheimer's disease. However, the present opinion paper posits that the initiating microdamage is in the prefrontal cortex, further upstream of pathophysiology, namely an irreversible Piezo2 channe

Quantitative analysis of the UDP-glucuronosyltransferase transcriptome in human tissues.

UDP-glucuronosyltransferases (UGTs) are phase II drug metabolizing enzymes that play important roles in the detoxification of endogenous and exogenous substrates. The 22 human UGTs belong to four families (UGT1, UGT2, UGT3, and UGT8) and differ in their expression, substrate specificity, UDP-sugar preference, and physiological functions. Differential expression/activity of the UGTs contributes to interperson variability in drug responses and toxicity, hormone homeostasis, and disease/cancer risks. However, in normal tissues, the tissue-specific expression profiles and transcriptional regulatio
