Trypanothione reductase: a target protein for a combined in vitro and in silico screening approach.

With the goal to identify novel trypanothione reductase (TR) inhibitors, we performed a combination of in vitro and in silico screening approaches. Starting from a highly diverse compound set of 2,816 compounds, 21 novel TR inhibiting compounds could be identified in the initial in vitro screening campaign against T. cruzi TR. All 21 in vitro hits were used in a subsequent similarity search-based in silico screening on a database containing 200,000 physically available compounds. The similarity search resulted in a data set containing 1,204 potential TR inhibitors, which was subjected to a sec

Multi-analytical platform metabolomic approach to study miltefosine mechanism of action and resistance in Leishmania.

Miltefosine (MT) (hexadecylphosphocholine) was implemented to cope with resistance against antimonials, the classical treatment in Leishmaniasis. Given the scarcity of anti- Leishmania (L) drugs and the increasing appearance of resistance, there is an obvious need for understanding the mechanism of action and development of such resistance. Metabolomics is an increasingly popular tool in the life sciences due to it being a relatively fast and accurate technique that can be applied either with a particular focus or in a global manner to reveal new knowledge about biological systems. Three analy

Tumor targeting of gene expression through metal-coordinated conjugation with dextran.

Источник пометил публикацию как отозванную

Tumor targeting of plasmid DNA was achieved through the conjugation of dextran derivatives with chelate residues based on metal coordination. Diethylenetriamine pentaacetic acid (DTPA), spermidine (Sd), and spermine (Sm) were chemically introduced to the hydroxyl groups of dextran to obtain dextran-DTPA, dextran-Sd and dextran-Sm derivatives. Conjugation of the dextran derivative by Zn(2+) coordination decreased the apparent size of the plasmid DNA, depending on the derivative type. The negative zeta potential of plasmid DNA became almost 0 mV after Zn(2+)-coordinated conjugation with dextran-

Autoradiographic distribution and characteristics of high- and low-affinity polyamine-sensitive [3H]ifenprodil sites in the rat brain: possible relationship to NMDAR2B receptors and calmodulin.

We have studied the regional distribution and characteristics of polyamine-sensitive [3H]ifenprodil binding sites by quantitative autoradiography in the rat brain. In forebrain areas ifenprodil displaced [3H]ifenprodil (40 nM) in a biphasic manner with IC50 values ranging from 42 to 352 nM and 401 to 974 microM. In hindbrain regions, including the cerebellum, ifenprodil displacement curves were monophasic with IC50 values in the high micromolar range. Wiping studies using forebrain slices (containing both high- and low-affinity sites) or cerebellar slices (containing only the low-affinity site

Occurrence and associated factors of biogenic amines in selected Mediterranean dairy products.

Fermented products, particularly cheese, represent a susceptible reservoir of biogenic amines (BA), which can pose health risks when present at elevated levels. This study aimed to detect the presence and concentration of 6 major BA (histamine [Hist], phenylethylamine [Phenyl], cadaverine [Cad], spermine [Sper], spermidine [Sped], and putrescine [Putr]) in selected Mediterranean dairy products, investigating the effects of packaging, fat content, and pH on BA levels. A total of 39 stock-keeping units (SKU) of 15 dairy products (yogurt, labneh, Baladi, Halloumi Baladi, Halloumi kobersi, Akkawi,

A biomarker-driven approach to targeting polyamine depletion in triple negative breast cancer.

BACKGROUND: Triple negative breast cancer (TNBC) is the most aggressive and heterogenous breast cancer subtype. Polyamines are essential for TNBC growth and proliferation; however, therapeutic strategies targeting polyamine metabolism have shown limited efficacy in TNBC. In this study, we demonstrate a biomarker-driven approach to treating TNBC through polyamine depletion. As the rate limiting catabolic enzyme in the polyamine pathway, spermidine/spermine N1-acetlytransferase (SAT1) reduces polyamines and generates the metabolite, diacetylspermine. METHODS: TNBC cell lines, ex vivo tumor slic

Deep learning of multiplexed mitochondrial morphology identified natural compound combinations against skin photoaging.

BACKGROUND: Skin photoaging is primarily driven by mitochondrial dysfunction. Although natural products have demonstrated protective effects against Ultraviolet (UV)-induced damage, efficient screening strategies for mitochondrial-targeting compounds remain limited. Artificial Intelligence (AI)-assisted high-content screening strategy offers a valuable approach to identify bioactive candidates and elucidate their mechanisms. PURPOSE: This study aimed to employe an AI-assisted high-content screening strategy to identify mitochondrial-enhancing natural compounds that prevent skin photoaging and

Geroprotective Agents, Including Glucagon-Like Peptide-1 Receptor Agonists, for Ocular Health.

Aging has long been implicated in the onset and progression of major retinal diseases, including age-related macular degeneration (AMD), diabetic retinopathy (DR), and retinal vein occlusion (RVO). Glaucoma is likewise increasingly recognized as an age-related disorder. Across these conditions, converging patterns of neurodegeneration and microvascular injury contribute to age-associated ocular decline. Structural and neuronal degeneration of the retina, including loss of retinal ganglion cell axons, along with impaired microvascular circulation and chronic inflammation, contribute to the path

Stepwise neutrophil-to-neuron targeting nanotherapy for mitochondrial protection and blood-brain barrier preservation in ischemic stroke.

Ischemic stroke is a leading cause of mortality and long-term disability worldwide, yet the clinical efficacy of reperfusion therapies remains severely constrained by a narrow therapeutic time window and secondary ischemia-reperfusion injury. Preserving the viability of the ischemic penumbra through early, multi-target neuroprotection represents a promising strategy to extend this window; however, effective approaches that simultaneously address neuronal mitochondrial dysfunction and blood-brain barrier (BBB) disruption are lacking. Here, we report a dual-modal, stepwise-targeting lipid nanopa

Integrated Multiomics Analyses Reveal Molecular Insights into How Intermittent Fasting Ameliorates Obesity and Increases Fertility in Male Mice.

Background: Intermittent fasting (IF) has been increasingly recognized for its potential to mitigate obesity and diabetes. However, it remains unclear whether IF can alleviate metabolic disorder-induced male infertility. The aim of this study was to investigate the potential of IF to improve fertility outcomes in obese mice. Methods: Eight-week-old C57BL/6J mice were fed a high-fat diet (HFD) for 24 weeks to induce obesity, followed by alternate-day fasting for 6 weeks. We assessed obesity-related metabolic changes and fertility issues postintervention. Comprehensive metabolomic and transcript

Genetic and Phenotypic Features of the Five Known Polyaminopathies: A Critical Narrative Review.

Polyaminopathies are a recently described family of rare genetic neurodevelopmental disorders. Polyaminopathies disrupt the biosynthesis of the primary polyamines: putrescine, spermidine, and spermine. Snyder-Robinson syndrome results from hemizygous loss-of-function variants in the spermine synthase (SMS) gene, resulting in decreased or complete loss of spermine synthase enzyme activity. Bachmann-Bupp syndrome results from heterozygous gain-of-function variants in the ornithine decarboxylase 1 (ODC1) gene, resulting in increased ornithine decarboxylase enzyme activity. Faundes-Banka syndrome

Longitudinal exposure to antiseizure medications shape gut-derived microbiome, resistome, and metabolome landscape.

The influence of chronically administered host-targeted drugs on the gut microbiome remains less understood compared to antibiotics. We investigated repetitive exposure effects of three common antiseizure medications [carbamazepine (CBZ), valproic acid, and levetiracetam] on the gut microbial composition, resistome, and metabolome using microcosms constructed from feces of young children. Microcosms were established by cultivating feces for 24 h (C0). These microcosms were daily transferred into fresh media for seven cycles (C1-C7) with antiseizure medications or carrier molecules, followed by

Unravelling the Metabolic Underpinnings of Gestational Diabetes Mellitus: A Comprehensive Mendelian Randomisation Analysis Identifying Causal Metabolites and Biological Pathways.

BACKGROUND: Gestational diabetes mellitus (GDM) has a strong genetic predisposition. Integrating metabolomics with Mendelian randomisation (MR) analysis offers a potent method to uncover the metabolic factors causally linked to GDM pathogenesis. OBJECTIVES: This study aims to identify specific metabolites and metabolic pathways causally associated with GDM susceptibility through a comprehensive MR analysis. Additionally, it seeks to explore the potential of these identified metabolites as circulating biomarkers for early GDM detection and risk assessment. Furthermore, it aims to evaluate the

Progesterone and interferon tau regulate expression of polyamine enzymes during the ovine peri-implantation period†.

Progesterone (P4) and interferon tau (IFNT) are important for establishment and maintenance of pregnancy in ruminants. Agmatine and polyamines (putrescine, spermidine, and spermine) have important roles in the survival, growth, and development of mammalian conceptuses. This study tested the hypothesis that P4 and/or IFNT stimulate the expression of genes and proteins involved in the metabolism and transport of polyamines in the ovine endometrium. Rambouillet ewes (n = 24) were surgically fitted with intrauterine catheters on Day 7 of the estrous cycle. They received daily intramuscular injecti

Human Salivary Protein Histatin 5 Has Potent Bactericidal Activity against ESKAPE Pathogens.

ESKAPE (Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumanni, Pseudomonas aeruginosa, and Enterobacter species) pathogens have characteristic multiple-drug resistance and cause an increasing number of nosocomial infections worldwide. Peptide-based therapeutics to treat ESKAPE infections might be an alternative to conventional antibiotics. Histatin 5 (Hst 5) is a salivary cationic histidine-rich peptide produced only in humans and higher primates. It has high antifungal activity against Candida albicans through an energy-dependent, non-lytic process; but it

Food Quality Monitor: Paper-Based Plasmonic Sensors Prepared Through Reversal Nanoimprinting for Rapid Detection of Biogenic Amine Odorants.

This paper describes the fabrication of paper-based plasmonic refractometric sensors through the embedding of metal nanoparticles (NPs) onto flexible papers using reversal nanoimprint lithography. The NP-embedded papers can serve as gas sensors for the detection of volatile biogenic amines (BAs) released from spoiled food. Commercial inkjet papers were employed as sensor substrates-their high reflectance (>80%) and smooth surfaces (roughness: ca. 4.9 nm) providing significant optical signals for reflection-mode plasmonic refractometric sensing and high particle transfer efficiency, respectivel

Bittersweet dynamics from flowers to fruits: chemometric molecular networking reveals metabolic changes towards reduced toxicity over ontogeny in above-ground Solanum dulcamara chemotypes.

Poisonous plants frequently deploy toxic metabolites in an organ- and ontogenetic-specific manner, yet the developmental dynamics of these plant specialised metabolites remain poorly understood. In the Solanaceae, steroidal glycosides (SGs), including steroidal glycoalkaloids (SGAs) and steroidal saponin glycosides (SSGs), are major defence metabolites with potent ecological and pharmacological activities. Here, we investigated how their metabolic profiles are reorganised during flower and fruit development in Solanum dulcamara using untargeted metabolomics and chemometric molecular networking

Bergamot (Citrus bergamia) preserves cardiac structure and function and modulates myocardial redox and metabolic profiles in diet-induced obesity.

Obesity promotes cardiac remodeling and dysfunction through oxidative stress, inflammation, and metabolic disturbances. Citrus bergamia (bergamot) has shown beneficial effects on obesity-related cardiometabolic risk factors, but its cardiac effects remain unclear. We evaluated bergamot supplementation in a rat model of diet-induced obesity using integrated assessments of cardiac structure and function, redox and inflammatory status, biogenic amines, fatty acids, and metabolites. Wistar rats (n = 54) were randomized to Control, Obese, or Obese + Bergamot groups for 20 weeks; bergamot was admini

Targeting SAT1 alleviates high glucose-induced tubular ferroptosis and fibrosis: implications for diabetic kidney disease.

Renal tubular damage and interstitial fibrosis are highly linked to diabetic kidney disease (DKD) progression. Ferroptosis in renal tubular epithelial cells has emerged as one of the key mechanisms of DKD. Spermidine/spermine N1-acetyltransferase 1 (SAT1) knockdown has been found to alleviate repetitive low-dose cisplatin-induced kidney damage and fibrosis, and importantly, SAT1 silencing represses cellular sensitivity to ferroptosis. However, the effect of SAT1 on DKD-associated ferroptosis and its potential mechanism remain understood. In this study, we constructed a high-fat diet/streptozot
