Dynamic Palmitoylation of Red Cell Membrane Proteins Governs Susceptibility to Invasion by the Malaria Parasite, Plasmodium falciparum.

Phosphorylation and other post-translational modifications of red blood cell (RBC) proteins govern membrane function and have a role in the invasion of RBCs by the malaria parasite, Plasmodium falciparum. Furthermore, a percentage of RBC proteins are palmitoylated, although the functional consequences are unknown. We establish dynamic palmitoylation of 118 RBC membrane proteins using click chemistry and acyl biotin exchange (ABE)-coupled LC-MS/MS and characterize their involvement in controlling membrane organization and parasite invasion. RBCs were treated with a generic palmitoylation inhibi…

Synthesis, biological evaluation and molecular docking studies of novel 1,2,3-triazole-quinazolines as antiproliferative agents displaying ERK inhibitory activity.

ERK1/2 inhibitors have attracted special attention concerning the ability of circumventing cases of innate or log-term acquired resistance to RAF and MEK kinase inhibitors. Based on the 4-aminoquinazoline pharmacophore of kinases, herein we describe the synthesis of 4-aminoquinazoline derivatives bearing a 1,2,3-triazole stable core to bridge different aromatic and heterocyclic rings using copper-catalysed azide-alkyne cycloaddition reaction (CuAAC) as a Click Chemistry strategy. The initial screening of twelve derivatives in tumoral cells (CAL-27, HN13, HGC-27, and BT-20) revealed that the mo…

New bis- and tetrakis-1,2,3-triazole derivatives: Synthesis, DNA cleavage, molecular docking, antimicrobial, antioxidant activity and acid dissociation constants.

In this study, a series of bis- and tetrakis-1,2,3-triazole derivatives were synthesized using copper-catalyzed azide-alkyne cycloaddition (CuAAC) click chemistry in 73-95% yield. The bis- and tetrakis-1,2,3-triazoles exhibited significant DNA cleavage activity while the tetrakis-1,2,3-triazole analog 6g completely degraded the plasmid DNA. Molecular docking simulations suggest that compound 6g acts as minor groove binder of DNA by binding through several noncovalent interactions with base pairs. All bis- and tetrakis-1,2,3-triazole derivatives were screened for antibacterial activity against …

Design, synthesis and biological evaluation of novel carbohydrate-based sulfonamide derivatives as antitumor agents.

A series of novel carbohydrate-based sulfonamides were designed and synthesized by the sugar-tail approach. The classical aromatic sulfonamide pharmacophore (ArSO2NH2) was directly linked to a hydrophilic sugar-tail moiety through a rigid 1, 2, 3-triazole linker by the click chemistry reaction. The inhibitory activity against three carbonic anhydrase (CA, EC 4.2.1.1) isozymes (hCA I, hCA II and hCA IX) of all new compounds so designed were investigated in vitro and efficient inhibition against all three CA isoforms, especially the tumor-associated hCA IX, were observed. These glycoconjugate su…

Cell-type-specific quantification of protein synthesis in vivo.

Although protein synthesis is a conserved and essential cellular function, it is often regulated in a cell-type-specific manner to influence cell fate, growth and homeostasis. Most methods used to measure protein synthesis depend on metabolically labeling large numbers of cells with radiolabeled amino acids or amino acid analogs. Because these methods typically depend on specialized growth conditions, they have been largely restricted to yeast, bacteria and cell lines. Application of these techniques to investigating protein synthesis within mammalian systems in vivo has been challenging. The

A ROS responsive nanomedicine with enhanced photodynamic therapy via dual mechanisms: GSH depletion and biosynthesis inhibition.

Reduced glutathione (GSH, containing sulfydryl) inside cancer cells strongly consumes reactive oxygen species (ROS) in photodynamic therapy (PDT) process to reduce PDT effect. Thiol-ene click reaction is a classic method to couple sulfydryl and alkene groups. However, little effort was dedicated to developing medicines for GSH depletion based on this reaction. Our research indicated that GSH could effectively couple with docosahexaenoic acid (DHA, as alkene) in the presence of 2,2-dimethoxy-2-phenylacetophenone (DMPA, as photo-initiator) post 665 nm light irradiation through the thiol-ene clic

Design, Synthesis, and Use of Novel Photoaffinity Probes in Measuring the Serum Concentration of Glycogen Phosphorylase.

A procedure to measure the serum concentration of glycogen phosphorylase during acute myocardial infarction is presented. This method was based on the synthesis of photoaffinity probes, and used the semiquantitative protein electrophoretic mobility shift technique. Three novel photoaffinity probes bearing different secondary tags were synthesized. Their potency was evaluated in an enzyme inhibition assay against rabbit muscle glycogen phosphorylase a (RMGPa). The inhibitory activity of probe 1 was only 100-fold less potent than the mother compound CP-320626. The photoaffinity labeling experime

Click-crosslinkable and photodegradable gelatin hydrogels for cytocompatible optical cell manipulation in natural environment.

This paper describes the generation of "click-crosslinkable" and "photodegaradable" gelatin hydrogels from the reaction between dibenzocycloctyl-terminated photoclevable tetra-arm polyethylene glycol and azide-modified gelatin. The hydrogels were formed in 30 min through the click-crosslinking reaction. The micropatterned features in the hydrogels were created by micropatterned light irradiation; the minimum resolution of micropatterning was 10-μm widths for line patterns and 20-μm diameters for circle patterns. Cells were successfully encapsulated in the hydrogels without any loss of viabilit

Radiosynthesis and biological evaluation of an fluorine-18 labeled galactose derivative [18F]FPGal for imaging the hepatic asialoglycoprotein receptor.

The asialoglycoprotein receptor (ASGPR) is abundantly expressed on the surface of hepatocytes where it recognizes and endocytoses glycoproteins with galactosyl and N-acetylgalactosamine groups. Given its hepatic distribution, the asialoglycoprotein receptor can be targeted by positron imaging agents to study liver function using PET imaging. In this study, the positron imaging agent [18F]FPGal was designed to specifically target hepatic asialoglycoprotein receptor and its effectiveness was assessed in in vitro and in vivo models. The radiosynthesis of [18F]FPGal required 50 min with total radi

Analysis of mesenchymal stem cell proteomes in situ in the ischemic heart.

Rationale: Cell therapy for myocardial infarction is promising but largely unsuccessful in part due to a lack of mechanistic understanding. Techniques enabling identification of stem cell-specific proteomes in situ in the injured heart may shed light on how the administered cells respond to the injured microenvironment and exert reparative effects. Objective: To identify the proteomes of the transplanted mesenchymal stem cells (MSCs) in the infarcted myocardium, we sought to target a mutant methionyl-tRNA synthetase (MetRSL274G) in MSCs, which charges azidonorleucine (ANL), a methionine analog

Photoresponsive Chemistries for User-Directed Hydrogel Network Modulation to Investigate Cell-Matrix Interactions.

Synthetic extracellular matrix (ECM) engineering is a highly interdisciplinary field integrating materials and polymer science and engineering, chemistry, cell biology, and medicine to develop innovative strategies to investigate and control cell-matrix interactions. Cellular microenvironments are complex and highly dynamic, changing in response to injury and disease. To capture some of these critical dynamics in vitro, biomaterial matrices have been developed with tailorable properties that can be modulated in situ in the presence of cells. While numerous macromolecules can serve as a basis i

Guanidinium-rich cell-penetrating peptidomimetic-doxorubicin conjugates show linker-dependent, reduced acute toxicity to cardiomyocytes and normal breast epithelium while retaining activity against a HER2-positive breast cancer model (SKBR3 spheroids).

Doxorubicin (DOX) is a cornerstone anthracycline in breast cancer therapy, but its use is constrained by cumulative cardiotoxicity. We asked whether conjugation to a guanidinium-rich cell-penetrating peptidomimetic widens the window between anticancer activity and cardiomyocyte toxicity, or merely rescales potency. DOX was linked by azide-alkyne click chemistry to an eight-guanidinium peptidomimetic, H-O2Oc-[Dap(GO2)]8-O2Oc-NH2, through either a hexynoyl linker (CuAAC; DOX-ACA-HEX-G8) or a dibenzocyclooctyne linker (SPAAC; DOX-ACA-DBCO-G8), so that the pair differs in a single variable. Both c…

Click inspired new series of pyridazine-triazole-persulfonimide derivatives as DPP-4 inhibitors with exploratory HepG2 phenotypic screening.

A click-enabled pyridazine-1,2,3-triazole sulfonimide platform yielded two libraries: persulfonimides (7a-7f) and aryl sulfonamides (8a-8d). All synthesized compounds were characterized by 1H and 13C NMR spectroscopy and mass spectrometry. In a fluorometric DPP-4 assay at fixed concentrations, inhibition increased with dose, with the most favorable profiles observed for 7d and 7f and the highest activity at 100 nM for 8c (91.31%) versus sitagliptin. In HepG2 cells. 7f reduced viability to 53% at 50 μM, while 8c showed a dose-dependent effect. Sorafenib was more potent (37.45% viability at 10 μ

IEDDA: An Attractive Bioorthogonal Reaction for Biomedical Applications.

The pretargeting strategy has recently emerged in order to overcome the limitations of direct targeting, mainly in the field of radioimmunotherapy (RIT). This strategy is directly dependent on chemical reactions, namely bioorthogonal reactions, which have been developed for their ability to occur under physiological conditions. The Staudinger ligation, the copper catalyzed azide-alkyne cycloaddition (CuAAC) and the strain-promoted [3 + 2] azide-alkyne cycloaddition (SPAAC) were the first bioorthogonal reactions introduced in the literature. However, due to their incomplete biocompatibility and

Ester modification at the 3' end of anti-microRNA oligonucleotides increases potency of microRNA inhibition.

MicroRNAs (miRNAs) are short noncoding RNAs that play a fundamental role in gene regulation. Deregulation of miRNA expression has a strong correlation with disease and antisense oligonucleotides that bind and inhibit miRNAs associated with disease have therapeutic potential. Current research on the chemical modification of anti-miRNA oligonucleotides (anti-miRs) is focused on alterations of the phosphodiester-ribose backbone to improve nuclease resistance and binding affinity to miRNA strands. Here we describe a structure-guided approach for modification of the 3'-end of anti-miRs by screening

Synthesizing Covalent L‑Shaped Bacteriochlorin Dimers To Isolate Purely Coulombic Excitonic Coupling.

Using de novo bacteriochlorins, we employed a click-chemistry strategy to synthesize elusive oblique dimers with L-oriented transition dipole moments (TDM) designed to isolate purely Coulombic excitonic coupling. The resulting covalent bis-bacteriochlorins (bis-BC1 and bis-BC2) utilize a non-conjugating bridge to prevent spatial orbital overlap between the constituent nonidentical chromophores. To enable excitonic coupling, we incorporated 3- and 13-auxochromic groups, which enhanced the TDM magnitudes of the constituent monomers. As a result, both bis-bacteriochlorins exhibited the split abso
